Abstract

The invention relates to prostate specific membrane antigen humanized antibodies (anti-PSMA) and anti-PSMA antibody drug conjugates. The invention also relates to methods and compositions for using anti-PSMA antibody drug conjugates in inhibiting, preventing or treating PSMA related diseases or cancers.

Claims (18)

CLAIMS :
1. An antibody drug conjugate comprising an anti-prostate specific membrane antigen antibody (anti-PSMA antibody) conjugated to a drug-linker, wherein the conjugation occurs via a para-acetyl phenylalanine incorporated in the heavy chain sequence of the antibody, wherein the anti-PSMA antibody comprises the heavy chain sequence of SEQ ID NO: 8 and the light chain sequence of SEQ ID NO: 9, in which the para-acetyl phenylalanine is incorporated at heavy chain position A114 according to the Kabat numbering scheme, and wherein the drug-linker is a non-cleavable monomethyl auristatin F having the structure:
2. A pharmaceutical composition comprising the antibody drug conjugate of claim 1 and at least one pharmaceutically acceptable adjuvant, binder, buffer, carrier, diluent or excipient.
3. The pharmaceutical composition of claim 2, further comprising a chemotherapeutic agent, hormonal agent, antitumor agent, immunostimulatory agent, immunomodulator, corticosteroid or a combination thereof.
4. The pharmaceutical composition of claim 2, further comprising a hormonal agent, wherein the hormonal agent is enzalutamide.
5. Use of the antibody drug conjugate of claim 1 in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell.
6. Use of the pharmaceutical composition of any one of claims 2-4 in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA- expressing cancer or cancer cell. MARKED UP COPY
7. Use of the antibody drug conjugate of claim 1 and an additional therapeutic agent in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell.
8. Use of the antibody drug conjugate of claim 1 in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell in combination with an additional therapeutic agent.
9. Use of an additional therapeutic agent in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell in combination with an effective amount of an antibody-drug conjugate of claim 1.
10. Use of the pharmaceutical composition of claim 2 and an additional therapeutic agent in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell.
11. Use of the pharmaceutical composition of claim 2 in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell in combination with an effective amount of an additional therapeutic agent.
12. Use of an additional therapeutic agent in the manufacture of a medicament for reducing or inhibiting tumor growth or progression in a PSMA-expressing cancer or cancer cell in combination with the pharmaceutical composition of claim 2.
13. The use according to any one of claims 5, 7 or 9-12, wherein the medicament comprises at least one pharmaceutically acceptable adjuvant, binder, buffer, carrier, diluent or excipient.
14. The use according to any one of claims 5-13, wherein the additional therapeutic agent is a chemotherapeutic agent, hormonal agent, antitumor agent, immunostimulatory agent, immunomodulator, corticosteroid or a combination thereof.
15. The use of claim 14, wherein the hormonal agent is enzalutamide. MARKED UP COPY
