Abstract

The present disclosure relates to recombinant adeno-associated virus (rAAV) delivery of a GALGT2 polynucleotide. The rAAV comprises a plasmid which comprises a nucleic acid comprising a first AAV2 inverted terminal repeat sequence (ITR), a muscle creatine kinase core promoter sequence, a nucleotide sequence encoding a human GALGT2 polypeptide and a second AAV2 ITR sequence. The disclosure provides rAAV and methods of using the rAAV for GALGT2 gene therapy of neuromuscular disorders. Exemplary neuromuscular disorders include, but are not limited to, muscular dystrophies such as Duchenne muscular dystrophy, Congenital Muscular Dystrophy 1A and Limb Girdle Muscular Dystrophy 2D.

Claims (13)

We claim:
1. A plasmid sing nucleotides 53-2762 of SEQ ID NO: 2.
2. The d of claim 1, wherein said plasmid lacks AAV rep and cap genes.
3. The plasmid of claim 1 or 2, r comprising a selectable marker.
4. An ed cell comprising the plasmid of any one of claims 1-3.
5. The isolated cell of claim 4, wherein the cell is a bacterial cell.
6. A packaging cell comprising the plasmid of any one of claims 1-3, provided that if the cell is a human cell, it is ex vivo.
7. The packaging cell of claim 6 selected from the group consisting of HeLa cells, PerC6 cells, 293 cells, low passage 293 cells, MRC-5 cells, WI-38 cells, Vero cells and FrhL-2 cells.
8. A method of producing a recombinant AAV (rAAV) le comprising contacting a cell with the plasmid of any one of claims 1-3, provided that the method is not performed in a human.
9. The method of claim 8, wherein the cell is the cell of any one of claims 4-7.
10. The method of claim 8 or claim 9, further comprising culturing the cell.
11. The method of any one of claims 10-12, further comprising recovering an rAAV particle from the supernatant of the cell.
12. A recombinant AAV particle produced by the method of any one of claims 8-11.
