Abstract
Antibodies that bind the apple 3 domain of human coagulation Factor XI and inhibit activation of FXI by coagulation factor XIIa as well as activation of FIX by FXIa are described.
Claims (17)
1. An antibody comprising a heavy chain (HC) comprising the amino acid sequence set forth in SEQ ID NO: 49, 51, 73 or 75 and a light chain (LC) comprising the amino acid sequence set forth in SEQ ID NO: 26.
2. An antibody comprising a heavy chain (HC) comprising the amino acid sequence set forth in SEQ ID NO: 49 and a light chain (LC) comprising the amino acid sequence set forth in SEQ ID NO: 26. 10
3. An antibody comprising a heavy chain (HC) comprising the amino acid sequence set forth in SEQ ID NO: 51 and a light chain (LC) comprising the amino acid sequence set forth in SEQ ID NO: 26.
4. An antibody comprising a heavy chain (HC) comprising the amino acid sequence set 15 forth in SEQ ID NO: 73 and a light chain (LC) comprising the amino acid sequence set forth in SEQ ID NO: 26.
5. An antibody comprising a heavy chain (HC) comprising the amino acid sequence set forth in SEQ ID NO: 75 and a light chain (LC) comprising the amino acid sequence set forth in 20 SEQ ID NO: 26.
6. A composition comprising the antibody of any one of the foregoing claims and a pharmaceutically acceptable carrier or diluent. 25
7. Use of an antibody of any one of the foregoing claims in the manufacture of a medicament for treating a thromboembolic disorder or disease.
8. The use of claim 7, wherein the medicament is for treating or preventing a thrombotic disorder or disease in a subject with severe or end-stage renal disease (ESRD). 30 9. The use of claim 7, wherein the medicament is for treating Stroke Prevention in Atrial

10. The use of claim 7, wherein the medicament is for treating or preventing venous thromboembolism (VTE).

12. The use of claim 11, wherein the medical device is a stent, endovascular stent graft, cardiac or venous catheter, continuous flow ventricular assist device (CF-LVAD), hemodialysis, cardiopulmonary bypass and Extracorporeal Membrane Oxygenation (ECMO), or ventricular assist device (VAD).
13. A method for producing an antibody of any one of claims 1-5, the method comprising: 10 providing a host cell comprising a nucleic acid molecule encoding the heavy chain of the antibody of any one of claims 1-5 and a nucleic acid molecule encoding the light chain of the antibody of any one of claims 1-5; and cultivating the host cell under conditions and a time sufficient to produce the antibody of any one of claims 1-5. 15
14. The antibody of any one of claims 1-5 substantially as herein described and with reference to any example thereof and with or without reference to the figures.
15. The composition of claim 6 substantially as herein described and with reference to any example thereof and with or without reference to the figures.
16. The use of any one of claims 7-12 substantially as herein described and with reference to any example thereof and with or without reference to the figures.
