Abstract
本发明涉及一种金蝉花提取物及其在制备神经保护和抗衰老药物中的应用。所述提取物为金蝉花醇提水溶性部位;按照下述方法获得:金蝉花粉碎过筛,用乙醇回流提取,乙醇提取物用水分散,依次用石油醚,乙酸乙酯,正丁醇分级萃取,减压浓缩回收溶剂,冷冻干燥分别得到石油醚部位,乙酸乙酯部位,正丁醇部位,水部位,取水部位浓缩;过AB-8大孔吸附树脂,先用水洗至近无色,再用80%乙醇洗脱;收集乙醇洗脱液,浓缩,冷冻干燥得水溶性部位(JCHH2O)。该提取物可明显抑制谷氨酸诱导的PC12细胞的衰老损伤,能降低细胞内的氧自由基水平,并且在清除DPPH·和超氧阴离子(O2 · -)自由基实验中表现出较好的体外抗氧化活性。

技术领域
本发明属于医药技术领域,具体涉及从中药材金蝉花中制备具有神经保护和抗衰老活性的部位及在制备神经保护和抗衰老药物方面的应用。
背景技术
随着我国人口老龄化加剧,神经退行性疾病的发病率日益增高,衰老与神经退行性疾病的研究已成为神经科学的热点。人体的衰老,往往会导致加快神经细胞数目的减少,从而产生一些以阿尔茨海默病(Alzheimer’s Disease,AD)为代表的神经退行性疾病。目前,神经退行性疾病,病因不明,而且尚无有效的治疗措施,仅有少数几个药物可用于治疗神经退行性疾病。
金蝉花为麦角菌科真菌大蝉草(Cordyceps cicadae Shing)及其寄主山蝉(Cicada flammata Dist)若虫形成的干燥复合体,其性味甘寒,与冬虫夏草的无性型同属,功效相近。甄权所著《药性论》云:“其蜕壳,头上有一角,如冠状,谓之蝉花,最佳。味甘寒,无毒。主小儿天吊,惊痫瘈,夜啼心悸。《证类本草》:“蝉花能解痉、散风热”。《本草图经》曾有记载:“今蜀中有一种蝉,其蜕壳上有一角,如花冠状,谓之蝉花。西人有赍至都下者,医工云,入药最奇”。《本草纲目》中亦记载:“蝉花可治疗惊痫,夜啼心悸,功同蝉蜕”。现代药理研究表明,金蝉花为传统的补益中药,具有明显的抗衰老、免疫调节、改善肾功能、抗肿瘤等药理活性。近年来研究发现,金蝉花中主要含有多糖、腺苷、虫草酸、氨基酸、多球壳菌素、麦角甾醇等多种化学成分。
蝉花水煎剂能明显延长实验小鼠的游泳时间,明显提高常压缺氧状态下的存活时间及在高温下的存活时间,表明蝉花水煎剂有抗应激、抗疲劳的作用;机体处在不良环境下,蝉花水煎剂能促进内环境保持相对稳定,从而增强了机体对有害刺激的抵抗力。蝉花水煎剂高剂量组对雄性果蝇能显著地延长寿命,表明其有一定的抗衰老作用(王砚,等.蝉花药理作用的初步探讨[J].浙江中医杂志,2001,36(5):219-220)。
鉴于蝉花和名贵中药冬虫夏草的无性型同属真菌(虫草属),且蝉拟青霉有免疫等多种药理作用,并具有毒性小、易培养等优点,有希望作为冬虫夏草的代用品,但对其相应的活性功能成分尚未十分明确,大多的药理实验仅停留在原药材或粗提物的基础上,尚无金蝉花提取物进一步精制至活性部位并用于制备神经保护和抗衰老药物的报道。
为了测试和评估本发明金蝉花活性部位的神经保护和抗衰老方面的应用潜力,已使用本领域技术人员所熟悉的方法测试药材各部位的生物活性。这些已知的测试方法包括谷氨酸损伤神经细胞PC12衰老模型,抗氧化模型等。结果表明选定的金蝉花醇提水溶性部位活性部位具有较好的神经保护和抗衰老的活性。
发明内容
本发明的目的是提供金蝉花具有神经保护和抗衰老活性应用潜力的有效部位。
金蝉花提取物的各部位经过反复多次的神经保护和抗衰老活性筛选,确认金蝉花60%一80%乙醇提取物的水溶性部位具有可明显抑制谷氨酸诱导的PC12细胞的衰老损伤,阻止细胞LDH释放,提高细胞的存活率,能降低细胞内的氧自由基水平,提高谷胱甘肽还原酶(GSH-Px)和超氧化物歧化酶(SOD)活性,显示出良好的神经保护和抗衰老活性,并且在清除DPPH·和超氧阴离子(O2·-)自由基实验中表现出较好的体外抗氧化活性,表明金蝉花的提取物具有良好的抗氧化作用,具有抗衰老的潜力。
因此,本发明的第一个方面涉及提供金蝉花醇提水溶性部位及其制备方法:金蝉花60%一80%乙醇提取物的水溶性部位制备方法,按照下述步骤进行:干燥金蝉花药材,粉碎过药典2号筛,粉末加入10一15倍的60%一80%乙醇回流提取,并过滤回收乙醇。金蝉花乙醇提取物用水分散,依次用0.5~1.5倍石油醚,乙酸乙酯,正丁醇分级萃取,共萃取3~5次,合并,减压浓缩回收溶剂,冷冻干燥分别得到石油醚部位,乙酸乙酯部位,正丁醇部位,水部位,水部位浓缩至原体积的1/40一1/10;过AB-8大孔吸附树脂,先用水洗至近无色,再用1BV一10BV80%(v/v)乙醇洗脱;流速为10一100mL/min;收集80%乙醇洗脱液,浓缩,冷冻干燥得水溶性部位(JCHH2O)。
本发明第二个方面涉及对所述金蝉花醇提水溶性部位用于制备神经保护和抗衰老药物的用途。
为了检测本发明所得的神经保护和抗衰老活性组分的性能,将本发明所得的神经保护和抗衰老活性组分按照配制成50~200μg/ml药液,用于大鼠肾上腺嗜铬细胞的PC12细胞处理,分别观察样品在不同剂量下对谷氨酸诱导的PC12细胞衰老的保护作用。结果表明金蝉花醇提水部位具有显著的神经保护和抗衰老活性,可用于制备治疗神经退行性疾病等疾病的辅助药物;同时在体外模型中具有显著的抗氧化活性,表明金蝉花活性部位在制备抗衰老药物方面具有良好的潜力。
本发明第三个方面涉及提供金蝉花活性部位与药学上可接受的辅料组成的药物组合物。
金蝉花活性部位可单独或几种部位组合,再进一步与辅料组合,剂型包括:片剂、胶囊剂、丸剂、颗粒剂、混悬剂、滴丸、口服液体制剂等。
本发明所述的载体或赋形剂包括药剂学常规应用的载体和赋形剂,例如溶剂、崩解剂、矫味剂、防腐剂、着色剂、粘合剂等。
下面的实施例和药理活性实验是对本发明的进一步详细说明,以下所列举的实施例不以任何方式构成限制。
具体实施方式
实施例1金蝉花80%乙醇提取物的水溶性部位(JCHH2O)制备:
取粉碎过药典2号筛干燥金蝉花粉末80g,加入10倍的80%乙醇回流提取,并过滤回收乙醇。金蝉花乙醇提取物用水分散,依次用等体积石油醚,乙酸乙酯,正丁醇分级萃取,共萃取3次,合并,减压浓缩回收溶剂,分别得到石油醚部位,乙酸乙酯部位,正丁醇部位,水部位,水部位浓缩,过AB-8大孔吸附树脂,先用水洗至近无色,再用3BV80%(v/v)乙醇洗脱;收集80%乙醇洗脱液,浓缩,冷冻干燥得JCHH2O4.24g。
实施例2JCHH2O对谷氨酸诱导的PC12细胞衰老的保护作用
(1)MTT法、LDH法测定细胞活力:
取对数生长期PC12细胞(由江苏大学医学院提供),以2×104/孔接种于96孔培养板,200μl/L。在37℃、5%CO2条件下培养过夜后,将细胞分为对照组、模型组、药物处理组。即对照组(不含Glu的完全培养基);模型组(完全培养基+Glu);药物处理组(完全培养基+JCHH2O+Glu,浓度分别为200μg/mL,100μg/mL,、50μg/mL,用培养液稀释。)JCHH2O预处理1h后加Glu孵育24h每组设4个平行孔。培养24h后,每孔加5g/L MTT20μl,继续培养4h后终止培养,小心吸取孔内上清液,做LDH实验。每孔加入DMSO150μL,使结晶充分溶解,用酶联免疫仪在波长570nm处读取吸光度(OD)。取4孔OD值的均数按公式计算细胞存活率结果见表1。细胞存活率%=实验组OD/对照组OD×100%。数据表明,JCHH2O具有保护Glu诱导PC12细胞的损伤作用。
收集培养液,按试剂盒说明书测定乳酸脱氢酶(LDH)(购买于南京建成生物工程研究所)的活性结果见表2。LDH的释放抑制率按下式计算:LDH抑制率(%)=(LDH模型组-LDH给药组)/(LDH模型组-LDH正常组)×100%。数据表明,JCHH2O具有抑制Glu诱导PC12细胞LDH释放效果。

(2)细胞内ROS水平、GSH-Px和SOD活力的测定:
取对数生长期PC12细胞,消化、计数,以4×104/mL的密度接种于24孔培养板中,在37℃、5%CO2条件下培养过夜后,按分组要求给以不同处理因素,每组设3个平行孔。继续培养24h后吸去培养基,轻轻用PBS洗一次,加入无血清培养基稀释的DCFH-DA溶液,使其终浓度为10μM,37℃下负载探针30min,PBS洗两次,胰酶消化,收集细胞,混匀后加入黑板透明底96孔培养板中,每孔100微升,每组设三个复孔,荧光酶标仪测定DCF荧光强度,最后对每组细胞计数,根据荧光强度及细胞密度计算每组细胞的DCF荧光强度/104Cells。结果见表3。数据表明,JCHH2O能够抑制Glu诱导PC12细胞ROS生成量。
取对数生长期细胞,消化、计数,以4×104/mL的密度接种于24孔培养板中,在37℃、5%CO2条件下培养过夜后,按分组要求给以不同处理因素,每组设3个平行孔。继续培养24h后吸去培养基,用PBS漂洗2次,留少许PBS,用细胞刮轻轻刮下细胞,收集到离心管中,1500rpm离心5min,500μl PBS悬浮细胞于eppendorf管中,细胞裂解液裂解细胞;12000rpm离心6min,取上清液,依试剂盒说明书进行各步骤反应;用酶标仪分别测定各组细胞。结果见表4。

实施例3JCHH2O体外抗氧化活性测定:
(1)DPPH·自由基清除能力的测定:
lmL不同浓度的金蝉花提取物溶液中加入1.0mL200μmol/DPPH·的乙醇溶液,再加入2.0mL80%乙醇后混合均匀,黑暗处放置30min后,用紫外分光光度计在517nm处测定吸光值A样品,同时测定1.0mL DPPH·的乙醇溶液与3.omL乙醇溶液混合液的吸光值A空白和3mL乙醇与1.0mL样品混合液的吸光值A对照,清除率公式:DPPH清除率=[A空白-(A样品-A对照)/A空白]×100%。数据表明,JCHH2O具有清除DPPH·能力。结果见表5
(2)超氧阴离子(O2·-)自由基清除能力的测定:
lmL不同浓度的金蝉花提取物溶液中,加3mlTris-HCl(PH8.2),25℃水浴20min后,加100μL10mmol/L邻苯三酚,精确反应4min,滴入100μL6mol/LHCl终止反应,在325nm处测定吸光度A样品,以去离子水代替样品浓液,其他同上,于325nm处测定吸光度A空白。超氧阴离子(O2·-)清除率按式计算。O2·-清除率=[(A空白-A样品)/A空白]×100%。数据表明,JCHH2O部位能够清除O2·-能力。结果见表6。

实施例4滴丸的制备
分别称取400g聚乙二醇4000,在水浴上熔化,再加入JCHH2O450g冻干粉末,搅拌均匀,倾入保温管中,调节恒温装置,使药液在80—90℃下滴入冷却过的液体石蜡中(温度±4℃),滴完后,将药丸倾入滤纸上吸干石蜡油,再加入少量滑石粉,混匀,得JCHH2O滴丸1000粒。
实施例5胶囊剂的制备
JCHH2O冻干粉末1000g,与药用淀粉500g混合均匀,烘干,按每粒0.45g制成胶囊。
实施例6片剂的制备
JCHH2O冻干粉末1000g,淀粉500g,混合均匀,用适量乙醇制粒,经整粒机整粒,压片,每片0.35g。
实施例7颗粒剂的制备
JCHH2O冻干粉末1500g,淀粉1000g,糖粉400g,混合均匀,用适量乙醇制粒,干燥、整粒、分装即得。
实施例8注射液的制备
100gJCHH2O冻干粉末,加入2500mL水,加热至80℃,加入2%(V/V)苯甲醇,混合均匀后过0.22μm滤膜,得到的澄明液灌封于2mL安瓶中,蒸汽灭菌3Omin,备用。
Claims (4)
1.一种金蝉花提取物,具有神经保护和抗衰老活性,为金蝉花60%一80%乙醇提取物的水部位;其特征在于所述金蝉花60%一80%乙醇提取物的水部位按照下述方法获得:干燥金蝉花药材,粉碎过药典2号筛,粉末加入10一15倍的60%一80%乙醇回流提取,并过滤回收乙醇;金蝉花乙醇提取物用水分散,依次用0.5~1.5倍石油醚,乙酸乙酯,正丁醇分级萃取,共萃取3~5次,合并,减压浓缩回收溶剂,冷冻干燥分别得到石油醚部位,乙酸乙酯部位,正丁醇部位,水部位;水部位浓缩至原体积的1/40一1/10;过AB-8大孔吸附树脂,先用水洗至近无色,再用1BV一10BV 80%(v/v)乙醇洗脱;流速为10一100mL/min;收集80%乙醇洗脱液,浓缩,冷冻干燥得水溶性部位,即金蝉花60%一80%乙醇提取物的水部位。
2.权利要求1所述金蝉花提取物在制备神经保护和抗衰老药物中的用途。
3.一种以金蝉花制备的神经保护和抗衰老药物,其特征在于该药物含有治疗有效量的权利要求1所述的金蝉花提取物。
4.一种药物制剂,其特征在于含有有效剂量的权利要求1所述金蝉花提取物和一种或多种药学上可接受的药物赋形剂,或可与金蝉花提取物组方的其他药物。
