Description
<B>Verfahren zur Herstellung eines neuen</B> Imidazolinabkömmlings. Die hervorragende physiologische Wirkung von 2-substituierten Imidazolinen, besonders ihre Fähigkeit, den Blutdruck zu beeinflussen, ist bekannt (C. R. Scholz, Industrial Engineer ing Chemistry, Ind. Ed.1945, 120).
In neuerer Zeit wurde festgestellt, dass man in gewissen Arzneimitteln, welche einen Dialkylamino- alkylrest enthalten, besonders in solchen mit Antihistaminwirkung, den genannten Rest ohne Beeinträchtigung der Wirkung, speziell also der Antihistaminwirkung, durch den 2-Methyl-imidazolin-Rest ersetzen kann (C. Djerassi und C. R. Scholz, J. Am. Chem. Soc. 69, 1688, 1947; M. Protiva und J.
Urban, Col- lection 13, 340, 1948).
So konnte man, nachdem die antihistami- nöse und analgetische Wirkung von a,a-Di- phenyl-co-dialkylaminoalkanen der Formel

bekannt war (C. P. Huttrer, Enzy mologia 12, 311, 1948), eine ähnliche Wirkung auch bei den 2-Diphenylalkyl-imidazolinen der Formeln

wobei n 1 oder 2 bedeutet, voraussetzen.
Die Herstellung des der Formel 1 entsprechenden Stoffes, das ist des 2-Benzhydryl-imidazolins, ist in der Literatur schon beschrieben; danach kann dieser Stoff auf die übliche Art aus dem Iminoäther des entsprechenden Äthylen- diamins (Gesellschaft für chem. Ind. in Basel: 0. P.150, 307; C.1937, 1I, 3039) und ferner durch Cyklisierung von Diphenylacetyläthylen- dia.min (S. R. Aspinall: J.
Am. Chem. Soe. 61, 3195, 1939) hergestellt werden. Die Herstel lung der der Formel II entsprechenden Stoffe, das ist des 2-Benzhydrylmethyl-imidazolins und 2-Benzhydryläthyl-imidazolins, wurde bis her nicht beschrieben.
Es wurde nun festgestellt, da.ss man die Stoffe der obigen Formeln nach der Methode von P. Oxley und W. F. Short (J. Chem. Soe. 1947, 497) herstellen kann, nämlich durch Schmelzen der entsprechenden Nitrite mit dem Äthylendiaminsalz der p-Toluolsulfonsäure.
Gegenstand des vorliegenden Patentes ist nun ein Verfahren zur Herstellung des neuen 2-Benzhydrylmethyl-imidazolins, welches da- durch gekennzeichnet ist, dass man ss,ss-Di- phenyl-propionitril mit dem Äthylendiamin- salz der p-Toluolsulfonsäure kondensiert.
Zweckmässig geht man so vor, dass man äquimolekulare lIengen der beiden Stoffe gründlich vermischt und das Gemisch einige Stunden auf 200 bis 210 C erwärmt. Das Reaktionsgemisch zerschmilzt dabei zu einer homogenen Schmelze, welche nach der Beendi gung der Reaktion und nach dem Erkalten zu einer glasigen Masse erstarrt. Diese Masse kann man in kristallinische Form überführen; sie besteht aus dem Salz des Imidazolins mit der p-Toluolsulfonsäure. Die Base kann aus diesem Salz z.
B. in der Weise freigesetzt werden, dass man das Salz in wässriger Sus pension mit einem Alkalihydroxyd behandelt, worauf man die Base mit Chloroform extra hieren und nach dem Abdampfen des Chloro forms in kristallinischer Form gewinnen kann. Das aus der Base auf die übliche Art (z. B. aus einer Acetonlösung durch Zusatz von Chlorwasserstoff) erhältliche Hydrochlorid ist in Wasser gut löslich und für die therapeu tische Anwendung besonders geeignet.
Was die biologische Wirkung des Imida.zolinderi- vates anbelangt, so wurde gefunden, da.ss die ser Stoff ausser der Wirkung auf den Blut druck auch antihistaminöse, spasmolytische und analgetische Wirkung'besitzt. <I>Beispiel:</I> Ein Gemisch von 10,3 g ss,ss-Diphenyl- propionitril und 11,6 g Äthylendiamin-mono- p-toluolsulfonat erwärmt man 3 Stunden auf 200 C.
Die entstandene Schmelze erstarrt beim Erkalten. Sie besteht aus 2-Benzhydryl- methyl-imidazolin-p-toluolsulfonat, welches nach einigen Kristallisationen aus verdünntem Äthanol den Schmelzpunkt 170 bis 17-1 C be sitzt. Aus diesem Abkömmling wird die Base in der oben beschriebenen Weise freigesetzt. Die Base schmilzt, aus Aceton kristallisiert., bei l02 C. Das aus der Base in der ebenfalls oben angegebenen Weise erhältliche Hydrochlorid schmelzt, aus Aceton kristallisiert, bei 88 bis 90 C.
Die Analyse des p-Toluolsulfonats und des Hydrochlorids ergeben für die Slinimar- formel der Base C17H18N..
<B> Process for the preparation of a new </B> imidazoline derivative. The excellent physiological effect of 2-substituted imidazolines, especially their ability to influence blood pressure, is known (C. R. Scholz, Industrial Engineering Chemistry, Ind. Ed. 1945, 120).
It has recently been found that in certain drugs that contain a dialkylaminoalkyl radical, especially those with an antihistamine effect, the mentioned radical can be replaced by the 2-methylimidazoline radical without impairing the effect, especially the antihistamine effect ( C. The preparation of the substance corresponding to formula 1, that is 2-benzhydryl-imidazoline, has already been described in the literature; then this substance can be prepared in the usual way from the imino ether of the corresponding ethylene diamine (Gesellschaft für chem. Ind. in Basel: 0. P. 150, 307; C.1937, 11, 3039) and further by cyclizing diphenylacetylethylene dia .min (SR Aspinall: J.
At the. Chem. Soe. 61, 3195, 1939). The manufacture of the substances corresponding to formula II, that is 2-benzhydrylmethyl-imidazoline and 2-benzhydrylethyl-imidazoline, has not been described so far.
It has now been found that the substances of the above formulas can be prepared by the method of P. Oxley and WF Short (J. Chem. Soe. 1947, 497), namely by melting the corresponding nitrites with the ethylenediamine salt of the p- Toluenesulfonic acid.
The present patent now relates to a process for the preparation of the new 2-benzhydrylmethyl-imidazoline, which is characterized in that ss, s-diphenylpropionitrile is condensed with the ethylenediamine salt of p-toluenesulfonic acid.
It is advisable to proceed in such a way that equimolecular lengths of the two substances are thoroughly mixed and the mixture is heated to 200 to 210 ° C. for a few hours. The reaction mixture melts into a homogeneous melt, which solidifies to a vitreous mass after the end of the reaction and after cooling. This mass can be converted into crystalline form; it consists of the salt of imidazoline with p-toluenesulfonic acid. The base can be made from this salt, for.

As far as the biological effect of the imida zoline derivative is concerned, it has been found that this substance has antihistaminic, spasmolytic and analgesic effects in addition to the effect on blood pressure. <I> Example: </I> A mixture of 10.3 g ss, ss-diphenyl propionitrile and 11.6 g ethylenediamine mono-p-toluenesulfonate is heated to 200 ° C. for 3 hours.
The resulting melt solidifies when it cools. It consists of 2-benzhydryl methyl imidazoline p-toluenesulfonate, which, after a few crystallizations from dilute ethanol, has a melting point of 170 to 17-1 ° C. The base is released from this derivative in the manner described above. The base melts, crystallized from acetone, at 10 ° C.
