イネ品種「あきだわら環1号」(国立研究開発法人農業・食品産業技術総合研究機構所有)の種子を、室温にて、200倍希釈の殺菌剤(スポルタック(登録商標)スターナ(登録商標)SE、住友化学株式会社製)、1、000倍希釈の殺虫剤(スミチオン(登録商標)乳剤、住友化学株式会社製)、及び1質量%過酸化水素水に24時間浸漬させ、シャーレ上に播種し、温度30℃、相対湿度100%の条件下で3日間置き、発芽させた。発芽させた種子は、イネ用育苗培土(ボンソル2号、住友化学株式会社製)を入れた園芸用プラグトレー(RL-40PT、株式会社東海化成製)に移植して栽培を行った。窒素不足により葉の黄化が認められた場合は、100個体に対し5%(w/v)の尿素を50mL追肥した。

次いで、切穎した種子親としてのイネ品種「あきだわら環1号」と、花粉親としてのイネ系統「sbe1あきだわら」と共に1つの交配袋(グラシン紙を用い、縦:32cm×横:8cmの袋状にして作成)に入れ、4日間、毎日開花している時間に30分間毎に手で交配袋を振り、種子親のめしべに、花粉親の花粉を振りかけた。

第1の工程で交配したイネ個体(種子親)を、交配直後(播種後47日目)に、植物育成用インキュベーター(LPH-350-SPCに、光源としてLEDチューブ(図1Eに示す発光スペクトルを有する、発光スペクトルにおける波長380nm以上780nm以下の波長領域において、波長615nm以上650nm以下の波長領域におけるピークトップの光強度が最大となる光を照射し得るLEDチューブ)を増設して改変した植物育成用インキュベーター;以下「改変インキュベーター1」と称することがある)に移し、図1Bに示す発光スペクトルの光環境下で、照射距離を20cmとし、明期が10時間(温度:27℃~30℃)、暗期が14時間(温度:18℃~25℃)の10時間日長で、CO濃度600ppm、相対湿度85%の条件下で育成した。
図1Bに示す発光スペクトルは、分光光度計(ライトアナライザーLA-105、株式会社日本医化器械製作所製)により測定した。改変インキュベーター1内の植物体付近で、光度計(ライトアナライザーLA-105、株式会社日本医化器械製作所製)により測定した照度は15,057ルクス、光量は、PPFD:228μmol m-2-1であった。
これにより、イネ品種「あきだわら環1号」と、イネ系統「sbe1あきだわら」との交配種子Fを得た。交配種子Fは、交配から27日間後に収穫した。

植物育成用インキュベーター(LPH-350-SPC、株式会社日本医化器械製作所製)内で、温度30℃、相対湿度100%の条件下で発芽させ、10日間育成した。
DNA Sui-Suiキット(Rizo Inc, Tsukuba, Ibaraki, Japan)を用いて発芽した苗(n=10)からDNAを抽出し、sbe1遺伝子のDNAマーカーを用いて交配種子Fが、イネ品種「あきだわら環1号」と、イネ系統「sbe1あきだわら」との交配により得られたものであることを確認した。
sbe1遺伝子を検出するために、プライマー対、Sbe1_5end_12I/D_U(配列番号:1)及びSbe1_5end_12I/D_L(配列番号:2)(特開2013-172710号公報参照)を使用した。
PCRの温度条件は、タッチダウンPCR法(Don et al.1991)に基づいた。PCRプログラムは以下の通りであった。DNAを完全に変性させるために94℃で5分間、続いて94℃で30秒間、アニーリング温度(後述)で60秒間、72℃で30秒間の34サイクル、その後72℃で10分間、完全な二本鎖DNAの合成を行った。アニーリング温度は、第1サイクルでは62℃であったが、第2サイクルから第14サイクルまでは1サイクル当たり0.5℃ずつ下げ、最後の20サイクルでは55℃に維持した。

To detect the sbe1 gene, the primer pair Sbe1_5end_12I/D_U (SEQ ID NO: 1) and Sbe1_5end_12I/D_L (SEQ ID NO: 2) (see JP 2013-172710 A) were used.
The temperature conditions for PCR were based on the touchdown PCR method (Don et al. 1991). The PCR program was as follows: 94°C for 5 min to completely denature DNA, followed by 34 cycles of 94°C for 30 s, annealing temperature (described below) for 60 s, and 72°C for 30 s, followed by 72°C for 10 min to synthesize complete double-stranded DNA. The annealing temperature was 62°C in the first cycle, decreased by 0.5°C per cycle from cycle 2 to cycle 14, and maintained at 55°C for the final 20 cycles.

イネ系統「sbe1あきだわら」(国立研究開発法人農業・食品産業技術総合研究機構所有)を用い、播種から出穂まで、図1Aに示す発光スペクトルの光環境下(LPH-350-SPC)、図1Bに示す発光スペクトルの光環境下(改変インキュベーター1)、図1Cに示す発光スペクトルの光環境下(改変インキュベーター2)、又は図1Dに示す発光スペクトルの光環境下(改変インキュベーター3)の一定の光環境下で育成した。
いずれの光環境においても、明期が10時間(温度:27℃~30℃)、暗期が14時間(温度:18℃~25℃)の10時間日長で、CO濃度600ppm、相対湿度85%の条件にて行った。
具体的には、各光環境下で、イネ系統「sbe1あきだわら」の種子を、シャーレ上に播種し、温度30℃、相対湿度100%の条件下で3日間置き、発芽させた。発芽させた種子は、イネ用育苗培土(ボンソル2号、住友化学株式会社製)を入れた園芸用プラグトレー(RL-40PT、株式会社東海化成製)に移植して栽培を行った。窒素不足により葉の黄化が認められた場合は、100個体に対し5%(w/v)の尿素を50mL追肥した。結果を下記表1に示す。

Specifically, seeds of the rice line "sbe1 Akidawara" were sown on a petri dish under each light environment and left for 3 days under conditions of a temperature of 30°C and a relative humidity of 100% to germinate. The germinated seeds were transplanted into a horticultural plug tray (RL-40PT, manufactured by Tokai Kasei Co., Ltd.) containing rice seedling soil (Bonsol No. 2, manufactured by Sumitomo Chemical Co., Ltd.) and cultivated. When yellowing of the leaves was observed due to nitrogen deficiency, 50 mL of 5% (w/v) urea was added as a top dressing for 100 individuals. The results are shown in Table 1 below.

本発明のイネ科植物の育成方法は、簡便で迅速な育種を行うことができ、また切穎法により得られた果実であっても十分な肥大を示すため、種子質量が大きい果実を得ることができ、更に得られた果実の発芽率及びその後の生育が良好である高品質の交配種子を得ることができるため、一般的なイネ科植物の育成方法のみならず、イネ科植物の高速世代促進技術において好適に利用することができる。
また、本発明のイネ科植物育成用装置は、イネ科植物の育成のために用いられるものであり、特にその光環境は、簡便で迅速な育種を行うことができ、また切穎法により得られた果実であっても十分な肥大を示し、種子質量が大きい果実を得ることができる光環境である。したがって、一般的なイネ科植物の育成に使用できるだけでなく、イネ科植物の高速世代促進技術において好適に使用することができる。

外部光を遮蔽して人工光のみをイネ科植物に照射してイネ科植物を育成させるイネ科植物の育成方法であって、
前記人工光として、国際照明委員会(CIE)が規定する標準光源を用い、色温度が5,000K~6,500Kの白色光を、種子親であるイネ科植物及び花粉親であるイネ科植物にそれぞれ照射して育成させる第1の工程と、
前記第1の工程で育成した前記種子親であるイネ科植物の穎花の上部を切除し、前記第1の工程で育成した前記花粉親であるイネ科植物と交配させる交配工程と、
前記人工光として、前記標準光源と、発光スペクトルにおける波長380nm以上780nm以下の波長領域において、波長615nm以上650nm以下の波長領域におけるピークトップの光強度が最大となる光を照射し得るLED光源と、から共に照射した光であって、発光スペクトルにおける波長380nm以上780nm以下の波長領域において、波長550nm以上780nm以下の波長領域におけるピークトップの光強度が最大となる光を、前記交配工程で得られた前記種子親であるイネ科植物に照射して育成させる第2の工程と、
を含むことを特徴とするイネ科植物の育成方法。
