この発明は新規な化粧料用の素材、特に優れた美白効果と保湿効果のある担子菌の子実体抽出物に関するものであって、さらに詳しくはイグチ科に属するアメリカウラベニイロガワリ、アシベニイグチ、イロガワリ、ウツロベニハナイグチ、アカジコウ、ヤマドリタケモドキ、ドクヤマドリ、ニセアシベニイグチ 、コガネヤマドリ、コウジタケ、キノボリイグチ、シロヌメリイグチ 、ヌメリツバイグチ、ハナイグチ、ヌメリイグチ 、アカヤマドリ、キンチャヤマイグチ、ヤマイグチ 、アケボノアワタケ 、ホオベニシロアシイグチ、ニガイグチ 、コショウイグチ、ウツロイイグチ、ヌメリコウジタケ、ハンノキイグチ、アミハナイグチ、キヒダタケ 、クロアザアワタケまたはオニイグチ科に属するオオキノボリイグチ、クリカワヤシャイグチあるいはヒダハタケ科に属するヒダハタケから選ばれる担子菌の子実体抽出物中に含まれるチロシナーゼ活性抑制作用に基づく美白効果を有する物質と、担子菌の子実体抽出物中に含まれる保湿効果を有する物質とを有効成分とするもので、かつ安全性が高い化粧料用素材に関する。

【従来の技術】
皮膚の黒化には色素のメラニンが深く関与しているものと考えられる。すなはち、メラニンが紫外線などの外的刺激を受けて肌の皮膚組織で生産され、そのために肌の黒化が促進され、シミ、ソバカス色黒等の症状が引き起こされるものと考えられる。
肌の美白の作用機序としていくつか挙げられるが、そのひとつとしてメラニンの生成に関与するチロシナーゼの活性化を抑制することが提案されている。
従来、チロシナーゼの活性化を抑制する物質として、美白化粧料には、主としてアスコルビン酸、グルタチオン、コロイドイオウ等が配合されており、このような美白化粧料は皮膚の色黒、シミ、ソバカスの防止など美容効果を得る上で有用である。
また、生薬などの天然物の抽出物を化粧料に配合し、美白効果を得ようとする試みがなされている。

【発明が解決しようとする問題点】
しかしながら、アスコルビン酸は、それ自体が酸化され易いため効果の発現が期待しにくいばかりか、配合した化粧品が経時的に変色、変臭する。また、グルタチオンやコロイドイオウは特有の臭気や安定性に問題があり、製品化に支障があった。一方、生薬類は、安全性が高いことからその有用性が期待されているものの、その美白効果はいまだ不十分であった。
また、これらの成分は美白効果は備えているものの、保湿効果については、充分に有していないものが多く、化粧料の重要な効果で有る保湿効果については他の有効成分を添加する必要があった。

(1)試料溶液の調整
前記各種子実体抽出液を、精製水により1.0 mg/ml濃度に溶解したものを試料溶液として調整する。
また、水に不溶の試料は、ポリオキシエチレン(50)硬化ひまし油により可溶化させ、1.0 mg/ml試料濃度に調整したものを試料溶液とする。
(2)チロシナーゼ溶液の調整
マッシュルーム由来のチロシナーゼ(シグマ社製) 10mg をMcllvaine Buffer(PH 6.8) 20ml で希釈し調整した。
(3)チロシナーゼ活性抑制効果の測定方法
試験管にそれぞれL−チロジン水溶液(0.3mg/ml)1.0 mlとMcllvaine Buffer(ph 6.8) 1.0 ml を加え、これらの各試験管に前記試料溶液およびコントロールテスト液を0.9ml加え、37℃で10分間インキュベートする。これに調整したチロシナーゼ溶液を0.1ml添加し、良く攪拌後、直ちに各反応液を分光光度計にセットし、475nmにおける吸光度を経時的に測定する。
各吸光度値を次式に代入してチロシナーゼ活性抑制率を算出する。なお、本発明のチロシナーゼ活性抑制率の算出には、チロシナーゼ溶液添加後10分後の吸光度値を使用した。
