本発明は、メントールの効果が最大限に引き出され、かつメントールの苦味が軽減されたメントール配合口腔用固形製剤に関する。

【従来の技術】
一般に、口腔用固形製剤は、薬効成分が長時間にわたって作用するように、薬効成分が製剤表面から順次溶解・崩壊するように設計されている。このような薬効成分の一つとしてメントールがあり、メントールは、食物やタバコ等の臭いをマスキングする効果に優れ、しかも口腔に清涼感を与える成分である。

しかしながら、メントールを口腔用固形製剤に多量に配合すると、これらの効果がより向上する反面、メントールの苦味が助長されるため、服用が困難になるという問題がある。そこで、口腔用固形製剤におけるメントールの苦味を低減させる方法として、例えば甘味成分の多量添加、発泡成分の添加により苦味の低減(特開平4−327526号、特開昭61−281183号公報)等の方法が提案されているが、臭いのマスキング効果や苦味の低減の点で十分に満足し得るものではない。

【発明が解決しようとする課題】
本発明の目的は、メントールによる臭いのマスキング効果が高く、かつメントールの有する苦味等の不快な味が軽減されたメントール配合口腔用固形製剤を提供することにある。

【課題を解決するための手段】
本発明者は、口腔用固形製剤におけるメントールの効果を高め、かつメントールの有する苦味等の不快な味を軽減すべく鋭意検討を行った結果、メントール配合量の異なる2層以上の層を有する錠剤又は飴とし、舌に接触するメントール濃度が経時的に徐々に高くなるようにすることにより、メントールによる異臭マスキング効果が高まり、かつメントールの苦味等が軽減されることを見出し、本発明を完成した。

すなわち、本発明は、メントールを配合し、3層以上の積層構造を有するか又は内層と外層で形成された2層以上の層を有する口腔用錠剤又は飴であって、口腔内でより遅く溶解する層のメントール配合量を当該層よりも早く溶解する層のメントール配合量よりも高くし、各層のメントール配合量差を0.6〜3.0重量%(各層の重量に対する重量%)とし、かつ、メントールを配合した層の1層以上を発泡性を有する層としたことを特徴とするメントール配合口腔用錠剤又は飴を提供するものである。

【発明の実施の形態】
本発明の口腔用錠剤又は飴は、口腔内でより遅く溶解する層のメントール配合量が当該層よりも早く溶解する層のメントール配合量よりも高くした構造を有するもので3層以上の積層構造を有するもの、内層の全部又は一部が外層で被覆された2層以上の層からなるものが挙げられる。また、本発明の口腔用錠剤又は飴は、これらの最上下層又は最外層等はメントールを含有しない層としてもよい。

本発明の口腔用固形製剤におけるメントール配合量は、苦味等の点から高メントール配合層において5重量%以下とすることが好ましい。また、各層のメントール配合量差は、メントールの苦味の軽減等の点から0.6〜3.0重量%が特に好ましい。例えば上中下層からなる3層構造の口腔用固形製剤、内層及び外層それぞれ1層からなる口腔用固形製剤の場合、上下層又は外層におけるメントール配合量を0.05〜1重量%とし、中層又は内層におけるメントール配合量を0.1〜5重量%とすることができる。

本発明の口腔用固形製剤のメントール配合は、炭酸塩と有機酸を配合した発泡性の層とすることが、メントールの有する苦味等の不快な味をより軽減させ得る点から好ましい。

炭酸塩としては、炭酸水素ナトリウムが代表的であるが、その対イオンとしてカリウム、カルシウム等も使用でき、これらは単独で又は2種以上組合わせて用いることができる。また、有機酸としては、例えばリンゴ酸、酒石酸、アスコルビン酸、コハク酸、クエン酸などが挙げられ、これらは1種又は2種以上を組合わせて用いることができる。
炭酸塩として炭酸水素ナトリウムを用いる場合、各層当たり0.5〜40重量%が好ましく、1〜30重量%が特に好ましい。この場合、有機酸は各層当たり1〜40重量%が好ましく、1〜30重量%が特に好ましい。

甘味料は1種を単独で又は2種以上を組合わせて用いることができ、製剤の味覚を矯正するために配合する場合、甘味の強さによって配合量が異なる。例えば、蔗糖、マンニトール、エリスリトール、ソルビトールの配合量は各層当たり通常5〜90重量%、好ましくは10〜80重量%であり、サッカリンやアスパルテームの配合量は0.01〜5重量%、好ましくは0.1〜1.5重量%である。

本発明の口腔用固形製剤は、メントールの有する異臭マスキングや清涼感付与等の効果が最大限に引き出され、かつメントールの有する苦味等の不快な味が軽減されたものである。

以下、実施例を挙げて本発明を更に説明するが、本発明はこれらの実施例に限定されるものではない。
なお、以下の実施例中、実施例1及び6は、特許請求の範囲外の参考例である。

実施例1及び比較例1
表1に示す組成のメントール配合量の異なる3層錠(1g、直径15mm、重量比:上層/中層/下層=1/1/1)をマシーナ(株)社製油圧式多層錠打錠機を用いて調製した。また、比較のために均一にメントールを配合した錠剤(1g、直径15mm)を調製した。マスキング効果を13名の喫煙者に喫煙してもらい、調製した錠剤を口に含み服用後、30分後に呼気袋に呼気を取り、これを3名のパネラーにタバコの臭いの有無を評価してもらった評価基準はタバコの臭いがしないを◎、ほとんどしないを○、若干するを△、するを×とした。結果を表2に示す。
