【発明の属する技術分野】
本発明は、線維芽細胞によるヒアルロン酸の産生を活発化する作用を有するヒアルロン酸産生促進剤、並びに皮膚の老化予防・改善作用を付与した皮膚化粧料及び美容作用を付与した飲食品(美容用飲食品)に関するものである。

【従来の技術】
皮膚の真皮及び表皮は、表皮細胞、線維芽細胞及びこれらの細胞の外にあって皮膚構造を支持するヒアルロン酸等の細胞外マトリックスによって構成されている。若い皮膚においては、これらの皮膚組織の相互作用が恒常性を保つことにより水分保持、柔軟性、弾力性等が確保され、肌は外見的にも張りや艶があってみずみずしい状態に維持される。

ところが、紫外線、空気の著しい乾燥、過度の皮膚洗浄等、ある種の外的因子の影響があったり加齢が進んだりすることによって、また外的因子の影響や加齢に伴う線維芽細胞の増殖率低下によって、天然保湿因子であるヒアルロン酸の産生量の低下を引き起こす。その結果、皮膚は保湿機能や弾力性が低下し、角質は異常剥離を始めるから、肌は張りや艶を失い、荒れ、シワ、くすみ等の老化症状を呈するようになる。

このように、皮膚の老化に伴う変化、即ち、シワ、くすみ、きめの消失、弾力性の低下等には、ヒアルロン酸等の真皮マトリックス成分の減少・変性と、線維芽細胞の増殖率の低下とが関与している。

【発明が解決しようとする課題】
そこで、本発明の第一の目的は、線維芽細胞によるヒアルロン酸の産生を促進して皮膚の老化を防止及び/又は改善し得る物質を見出し、それを有効成分として含有するヒアルロン酸産生促進剤を提供することにある。

また、本発明の第二の目的は、ヒアルロン酸産生促進作用を有し、皮膚の老化を防止及び/又は改善する上で有用な皮膚化粧料を提供することにある。

さらに、本発明の第三の目的は、ヒアルロン酸産生促進作用を有し、皮膚の老化を防止及び/又は改善する上で有用な美容用飲食品を提供することにある。

【課題を解決するための手段】
上記目的を解決するために、本発明のヒアルロン酸産生促進剤は、吹雲草からの抽出物を有効成分として含有することを特徴とし、本発明の皮膚化粧料及び美容用飲食品は、吹雲草からの抽出物を配合したことを特徴とする。

【発明の実施の形態】
以下、本発明について詳細に説明する。
本発明において、「抽出物」には、吹雲草、鋪地草及び通大海からなる群より選ばれる1種又は2種以上の植物を抽出原料として得られる抽出液、該抽出液の希釈液若しくは濃縮液、該抽出液を乾燥して得られる乾燥物、又はこれらの粗精製物もしくは精製物のいずれもが含まれる。

抽出原料として用いる植物は、吹雲草、鋪地草及び通大海からなる群より選ばれる1種又は2種以上の植物である。2種以上の植物を抽出原料として用いる場合、上記植物を任意に組み合わせることができる。

吹雲草(スイウンソウ)(学名:Salomonia cantoniensis)(別名:サラモウ,一碗泡(イチワンホウ),公児草(コウジソウ),過山蛇(カザンダ),斬蛇剣(ザンジャケン))は、ヒメハギ科に属する一年生の草本であり、広東、広西、雲南、貴州、湖南、江西、福建等に分布しており、これらの地域から容易に入手可能である。また、鋪地草(ホチソウ)(学名:Euphorbia chamaesyce,Euphorbia prostrata)(別名:乳汁草(ニュウジュウソウ),ハイニシキソウ)は、トウダイグサ科に属する一年生の草本であり、広東、広西、福建、四川、雲南、貴州等に分布しており、これらの地域から容易に入手可能である。また、通大海(ツウタイカイ)(学名:Sterculia lychnophera)は、アオギリ科に属する落葉高木であって、ベトナム、インド、マレーシア、タイ、インドネシア等の熱帯地区に分布しており、これらの地域から容易に入手可能である。

抽出原料として用いる植物の構成部位は特に限定されるものではなく、吹雲草については、例えば全草、葉部、茎部、花、種子、根部等の構成部位を、鋪地草については、例えば全草、葉部、茎部、花、種子、根部等の構成部位を、通大海については、例えば葉部、枝部、樹皮、花、種子、根部等の構成部位を抽出原料として用いることができる。これらの構成部位のうち、吹雲草については特に葉部及び/又は枝部を、鋪地草については特に葉部を、通大海については特に種子を抽出原料として用いることが好ましい。

上記植物からの抽出物に含有されるヒアルロン酸産生促進作用を有する物質の詳細は不明であるが、植物の抽出に一般に用いられている抽出方法によって、上記植物からヒアルロン酸産生促進作用を有する抽出物を得ることができる。例えば、抽出原料を乾燥した後、そのまま、又は粗砕機を用いて粉砕し、抽出溶媒による抽出に供することにより得ることができる。この際、抽出原料の乾燥は天日で行ってもよいし、通常使用される乾燥機を用いて行ってもよい。また、上記植物は、ヘキサン、ベンゼン等の非極性溶媒によって脱脂等の前処理を施してから抽出原料として使用してもよい。脱脂等の前処理を行うことにより、極性溶媒による抽出処理を効率よく行うことができる。

抽出溶媒としては、水若しくは親水性有機溶媒又はこれらの混合液を室温又は溶媒の沸点以下の温度で用いることが好ましい。

抽出溶媒として使用し得る水としては、純水、水道水、井戸水、鉱泉水、鉱水、温泉水、湧水、淡水等の他、これらに各種処理を施したものが含まれる。水に施す処理としては、例えば、精製、加熱、殺菌、ろ過、イオン交換、浸透圧の調整、緩衝化等が含まれる。従って、本発明において抽出溶媒として使用し得る水には、精製水、熱水、イオン交換水、生理食塩水、リン酸緩衝液、リン酸緩衝生理食塩水等も含まれる。

抽出溶媒として使用し得る親水性有機溶媒としては、例えば、メタノール、エタノール、プロピルアルコール、イソプロピルアルコール等の炭素数1〜5の低級アルコール;アセトン、メチルエチルケトン等の低級脂肪族ケトン;1,3−ブチレングリコール、プロピレングリコール、グリセリン等の炭素数2〜5の多価アルコールなどが挙げられる。

2種以上の極性溶媒の混合液を抽出溶媒として使用する場合、その混合比は適宜調整することができる。例えば、水と低級脂肪族アルコールとの混合液を使用する場合には、水と低級脂肪族アルコールとの混合比を7:3〜2:8(重量比)とすることができる。

抽出処理は、上記植物に含有される可溶性成分を抽出溶媒に溶出させ得る限り特に限定されるものではなく、常法に従って行うことができる。抽出処理の際には特殊な抽出方法を採用する必要はなく、室温又は還流加熱下において任意の装置を用いることができる。

具体的には、抽出溶媒を満たした処理槽に抽出原料を投入し、必要に応じて時々攪拌しながら、通常1〜3時間静置して可溶性成分を溶出した後、ろ過して固形物を除去し、得られた抽出液から抽出溶媒を留去し、乾燥することにより抽出物が得られる。抽出溶媒量は抽出原料の通常5〜15倍量(重量比)であり、抽出温度は、通常、常温〜95℃である。

得られた抽出液は、該抽出液の希釈液若しくは濃縮液、該抽出液の乾燥物、又はこれらの粗精製物若しくは精製物を得るために、常法に従って希釈、濃縮、乾燥、精製等の処理を施してもよい。

得られた抽出液はそのままでもヒアルロン酸産生促進剤として使用することができるが、濃縮液又は乾燥物としたものの方が利用しやすい。上記植物からの抽出物の製剤化は常法に従って行うことができる。製剤化する場合、保存や取扱いを容易にするために、デキストリン、シクロデキストリン等の薬学的に許容され得るキャリアーその他任意の助剤を添加することができ、上記植物からの抽出物を粉末状、果粒状、錠剤状等、任意の剤形に製剤化することができる。

上記植物からの抽出物は特有の匂いを有しているため、その生理活性の低下を招かない範囲で脱色、脱臭等を目的とする精製を行うことも可能であるが、皮膚化粧料や飲食品などに添加する場合には大量に使用するものではないから、未精製のままでも実用上支障はない。精製は具体的には、活性炭処理、吸着樹脂処理、イオン交換樹脂処理等によって行うことができる。

本発明のヒアルロン酸産生促進剤は、線維芽細胞によるヒアルロン酸の産生を活発化させ、ヒアルロン酸の減少、変性等によって生じる皮膚の老化を防止及び/又は改善することができる。

〔皮膚化粧料〕
上記植物からの抽出物は、皮膚の老化を防止及び/又は改善する作用を有するとともに、皮膚に適用した場合の使用感と安全性に優れているので、皮膚化粧料に配合するのに好適である。皮膚化粧料には、本発明のヒアルロン酸産生促進剤を配合してもよい。

上記植物からの抽出物を配合し得る皮膚化粧料の種類は特に限定されず、その具体例としては、軟膏、クリーム、乳液、ローション、パック、口紅、入浴剤等が挙げられる。

本発明の皮膚化粧料における上記植物からの抽出物の配合量は、皮膚化粧料の種類や抽出物の生理活性等によって適宜調整することができるが、上記植物からの抽出物の総配合量が、標準的な抽出物に換算して約0.00001〜10重量%となるように配合することが好ましい。

本発明の皮膚化粧料には、上記植物からの抽出物のヒアルロン酸産生促進作用の妨げにならない限り、皮膚化粧料の製造に通常使用される各種主剤及び助剤、その他任意の助剤を使用することができる。本発明の皮膚化粧料は、皮膚の老化防止・改善に関し、上記植物からの抽出物のみが主剤となるものに限られるわけではない。

本発明の皮膚化粧料において、上記植物からの抽出物と共に皮膚化粧料構成成分として利用可能なものとしては、例えば、収斂剤、殺菌・抗菌剤、美白剤、紫外線吸収剤、保湿剤、細胞賦活剤、消炎・抗アレルギー剤、抗酸化・活性酸素消去剤等が挙げられ、上記植物からの抽出物とともに上記構成成分を併用した場合、上記植物からの抽出物と併用された構成成分との間の相乗作用が、通常期待される以上の優れた使用効果をもたらすことがある。

上記植物からの抽出物を配合した皮膚化粧料を製造する場合、他の製造原料の選択が制限されることはほとんどなく、油脂類、ロウ類、炭化水素類、脂肪酸類、アルコール類、エステル類、界面活性剤、香料等の一般的な基材や助剤はいずれも使用可能である。

〔美容用飲食品〕
上記植物からの抽出物は、皮膚の老化を防止及び/又は改善する作用を有するとともに、消化管で消化されるようなものではないことが確認されているので、任意の飲食品や栄養補助食品に配合するのに好適である。飲食品や栄養補助食品には、本発明のヒアルロン酸産生促進剤を配合してもよい。

上記植物からの抽出物を配合した飲食品には、皮膚の老化を防止及び/又は改善する作用が付与され、これを美容用飲食品として使用することができる。ここで、「美容用飲食品」とは、美肌又は皮膚の老化の防止及び/又は改善を図ることを目的とした飲食品を意味する。

本発明の美容用飲食品は、上記植物からの抽出物をその活性を妨げないような任意の飲食品に配合したものであってもよいし、上記植物からの抽出物を主成分とする栄養補助食品であってもよい。

本発明の美容用飲食品を製造する際には、例えば、デキストリン、デンプン等の糖類;ゼラチン、大豆タンパク、トウモロコシタンパク等のタンパク質;アラニン、グルタミン、イソロイシン等のアミノ酸類;セルロース、アラビアゴム等の多糖類;大豆油、中鎖脂肪酸トリグリセリド等の油脂類等の任意の助剤を添加して任意の剤形に製剤化することができる。

本発明の美容用飲食品における上記植物からの抽出物の配合量は、添加対象飲食品の一般的な摂取量を考慮して成人1日当たりの抽出物総摂取量が約1〜1000mg程度になるようにするのが適当である。

上記植物からの抽出物を配合し得る飲食品の種類は特に限定されないが、その具体例としては、清涼飲料、炭酸飲料、栄養飲料、果実飲料、乳酸飲料等の飲料(これらの飲料の濃縮原液及び調整用粉末を含む);アイスクリーム、アイスシャーベット、かき氷等の冷菓;そば、うどん、はるさめ、ぎょうざの皮、しゅうまいの皮、中華麺、即席麺等の麺類;飴、チューインガム、キャンディー、ガム、チョコレート、錠菓、スナック菓子、ビスケット、ゼリー、ジャム、クリーム、焼き菓子等の菓子類;かまぼこ、ハム、ソーセージ等の水産・畜産加工食品;加工乳、発酵乳等の乳製品;サラダ油、てんぷら油、マーガリン、マヨネーズ、ショートニング、ホイップクリーム、ドレッシング等の油脂及び油脂加工食品;ソース、たれ等の調味料;スープ、シチュー、サラダ、惣菜、漬物などが挙げられる。

以上説明した本発明のヒアルロン酸産生促進剤、皮膚化粧料及び美容用飲食品は、ヒトに対して好適に適用されるものであるが、それぞれの作用効果が奏される限り、ヒト以外の動物に対して適用することもできる。

【実施例】
以下、製造例、試験例及び配合例を示して本発明を具体的に説明するが、本発明は、下記の各例に何ら限定されるものではない。

〔製造例1〕
吹雲草の葉部及び枝部、鋪地草の葉部、通大海の種子の粗粉砕物300gをそれぞれ抽出溶媒3000mLに投入し、穏やかに攪拌しながら2時間、80℃に保った。水、50%エタノール又はエタノールの3種類の抽出溶媒を用いて上記抽出処理を行った後、ろ過し、ろ液を40℃で減圧下にて濃縮し、さらに減圧乾燥機で乾燥させた。抽出溶媒として水、50%エタノール、エタノールを用いたときの各抽出率を表1に示す。なお、抽出溶媒が混合物の場合、その混合比は重量基準によるものである。
