【発明の属する技術分野】
本発明は従来利用されることの無かった、イカの内臓由来の新規原料を含有する化粧料に関するもので、更に詳細にはイカの包卵腺より可溶化処理の後に、抽出された粘性物質を含有した化粧料及び前記粘性物質の採取方法に関する。

【従来の技術】
イカは産卵の際、卵は無色透明なゼリー状の材料に包まれている。このゼリー状の物質は産卵の前に包卵腺の中に原料として蓄積され、それが産卵の際に使われている。従来、イカの包卵腺を含む内蔵類は、ほとんど利用されることなく廃棄されていて、わずかに練り物類、中華料理の食材として利用されているのみであった。この包卵腺に蓄積されたゼリー状物質の利用については全く検討されたことが無く、更には抽出、精製方法についても知られていなかった。

【発明が解決すべき問題点】
本発明は、従来利用されることの無かったこのイカの包卵腺の新規利用を開拓することを目的としたもので、利用可能な原料の抽出、精製法を開発し、これを新規な化粧品原料として開発することを目指したものである。

【問題点を解決するための手段】
上記目的を達成するために鋭意検討の結果、包卵腺をアルカリによって処理することで抽出が可能となり、その抽出物は無色透明な粘性な物質であって、この粘性物質は化粧料原料として優れた特性を持つことを明らかにし、本発明を完成した。
即ち、本願の第1の発明の要旨はイカ包卵腺よりアルカリにより可溶化処理し、抽出した粘性物質を原料の一つとして含有することを特徴とした化粧料である。また、第2の発明の要旨はイカ包卵腺をアルカリにより可溶化処理し、得られた粘稠な溶液にアルコール及び中性塩を添加して沈殿物を生成させ、この沈殿物を濾別、回収することを特徴とするイカ包卵腺抽出物の採取方法である。
包卵腺は雌イカの腹を開いた際に中心部に位置する、比較的大きな内臓器官でこの内臓器官は産卵時期になると肥大化し、マイカでは50g程度、スルメイカでは20g程度の重量となる。

【発明の実施の形態】
本発明に用いることが出来るイカの種類に特に制限はないが、アカイカ科、ヤリイカ科、コウイカ科のスルメイカ、アカイカ、トビイカ、ヤリイカ、ケンサキイカ、アオリイカ、ジンドウイカ、スミイカ、モンゴイカ(別名マイカ、アカイカ、コウイカ、ゴウドウイカ、ムラサキイカ、バカイカ、ツメイカ、ニュウドウイカ、タコイカ、ドスイカ、シリヤケイカ、ハリイカ、ヒメイカ、コウイカ、ホタルイカ、ソデイカ、ダイオウイカ等)が望ましい。
集めた包卵腺をそのまま、あるいは乾燥させ、これをアルカリ性の溶液に入れ包卵線に含まれている粘性物質を可溶化して抽出を行う。用いるアルカリ性の溶液としては、NaOH、KOH等水酸イオンを持つ無機塩で、溶液としてのpHは12以上が望ましい。これよりpHが低い場合には、可溶化を十分に行うことが出来ない。具体的には0.1〜1N程度の溶液を用いることが出来る。なお通常生体内に粘質物として存在するムチン(粘素)はアルカリで抽出する場合、β―位脱離を起こし分解を生じるが、本発明で使用するこの粘性物質の場合にはβ―位脱離を起こすことが無いために、従来β―位脱離を抑えるために行われるNaBH4等を用いる還元処理は不用であることを見いだした。
アルカリ処理時の温度は0℃〜室温付近が望ましい。0℃以下では可溶化が進行しにくく抽出の効率が悪く、室温を超える場合には粘性物質の分解が起こり、化粧料原料としての特性を損なうことになる。また処理時間はアルカリ溶液の濃度、温度に依存するが、1時間から10時間の処理により可溶化、抽出は終了する。

この可溶化、抽出の終了した溶液にアルコール溶液、並びに中性塩を加えることによって、抽出した粘性物質を沈殿させることが出来る。用いるアルコール溶液はメタノール、エタノール、プロパノール、イソプロパノール等通常の糖タンパク質を沈殿させる有機溶媒を用いることが出来、例えば50〜60%のエタノール溶液等が使用出来る。さらにこのアルコール溶液にNaCl等の中性塩を含有させることによって、抽出物の沈殿を得ることが出来る。沈殿させた粘性物質は遠心分離、濾別等によって集めることが出来る。
使用できる中性塩に特に制限はなく、NaClの他にKCl、Na2SO4等水溶性の中性塩を用いることが出来る。

得られた沈殿は水に容易に溶解することが出来、その溶液の固有粘度を測定したところ30dl/g以上の範囲の固有粘度を持つ溶液であった。また得られた粘性物質はおおよそタンパク質35〜38%、糖質62〜65%、その他成分から構成された糖タンパク質で、約100万〜200万の分子量を持つと予想される。

本発明で得られた粘性物質は、無色透明、無臭で高い粘度を持ち、保湿、湿潤、増粘、乳化安定、乳化助剤、整肌、肌保護、潤滑、感触改善等の効果を得ることが出来る。具体的には基礎化粧品、化粧水(ローション)、乳液、クリーム、美容液、パック剤、下地クリーム、ファンデーション、口紅等の皮膚化粧料、シャンプー、リンス、ヘアーリキッド、ヘアークリーム等の頭髪化粧料等に用いることが出来、更に具体的には冬季の肌荒れ、乾燥による肌荒れ、髭剃り後の肌手入れ、洗髪、染毛・パーマ後の手入れ用化粧品に配合して使用することが出来る。

【実施例】
以下の実施例により本発明を具体的に説明するが、これによって本発明が限定されるものではない。
実施例1
スルメイカより取り出した包卵腺(湿重量約3g)を水(150mL)に入れ、ホモジナイズした後に、NaOHを加えNaOH濃度として0.4Nの溶液とする。この溶液を4℃にし4時間ゆっくりと攪拌する。処理後の粘調な溶液に50%エタノールとNaClを加え沈殿を生成させる。得られた沈殿を遠心分離によって回収し、イカ粘性物質(乾燥重量0.2g)を得る。本粘性物質(0.2g)を水100mLに溶解し、イカ粘性物質溶液を得た。

実施例1で得られたイカ粘性物質溶液を使用し、以下の組成からなる栄養乳液を調製した。
