本発明はシミ、ソバカスなどの色素沈着を消去、軽減又は防御して、白く美しい肌をつくる、白く美しい肌にかえる又は白く美しい肌を維持するという美白化粧料に関する。

【従来の技術】
これまでの美白化粧料は、生体内でメラニン色素が形成される際の主要酵素であるチロシナーゼに対する活性阻害作用を有する成分やメラニン産生細胞のメラニン合成量を制御する成分、あるいは還元作用を有する成分等が配合され、局所に塗布する方法がとられている。しかしながら、これらの成分の多くは有効性が弱く、また不安定であり、未だ十分な美白効果が得られていないのが実情である。

【発明が解決しようとする課題】
本発明は、従来の美白化粧料では満足し得なかった色素沈着の消去、軽減又は防御という目的に対して、従来よりも積極的に作用する美白化粧料を提供するものである。

本発明者らは、色素沈着の軽減について鋭意研究を重ねた結果、レブリン酸またはその塩と、1種又は2種以上の硫黄化合物、あるいはさらに1種又は2種以上のメラニン生成抑制成分を組合わせた成分を組合わせた成分を含有することを特徴とする美白化粧料が優れた色素沈着の消去、軽減又は防御作用を有することを見出し、本発明を完成するに至った。

本願発明は、下記の請求項1〜請求項4により構成されている。
〔請求項1〕レブリン酸またはその塩と、硫酸塩、亜硫酸塩、チオ硫酸塩、又はピロ亜硫酸塩から選ばれる1種又は2種以上の硫黄化合物を組合わせた成分を含有することを特徴とする美白化粧料。
〔請求項2〕請求項1の美白化粧料に、更に1種又は2種以上のメラニン生成抑制成分を組合わせた成分を含有することを特徴とする美白化粧料。
〔請求項3〕硫黄化合物が、硫酸ナトリウム、亜硫酸ナトリウム、チオ硫酸ナトリウム、ピロ亜硫酸ナトリウムである請求項1、又は請求項2記載の美白化粧料。
〔請求項4〕メラニン生成抑制成分が、アスコルビン酸、アルブチン、エラグ酸、カテコール、グルタチオン、コウジ酸、システイン、パンテテイン−S−スルホン酸、ハイドロキノン、レゾルシン、レチノイン酸、乳酸、過酸化水素、臭素酸、塩素酸、プラセンタエキスおよびそれらの誘導体である請求項2、又は請求項3に記載する美白化粧料。
本発明の美白化粧料を以上のように構成する理由は、美白化粧料に、レブリン酸またはその塩と、1種又は2種以上の硫黄化合物、あるいはさらに1種又は2種以上のメラニン生成抑制成分を組合わせた成分を含有させることにより、シミ、ソバカスなどの色素沈着を消去し、軽減し、又は防御して、白く美しい肌をつくる、白く美しい肌にかえる、又は白く美しい肌を維持する効果を得るためである。

【発明の実施の形態】
本発明の美白化粧料は、レブリン酸またはその塩と、1種又は2種以上の硫黄化合物、あるいはさらに1種又は2種以上のメラニン生成抑制成分を組合わせた成分を含有することを特徴とする。

本発明におけるレブリン酸またはその塩の配合量は、0.001〜20.0重量%であり、さらに好ましくは0.1〜10.0重量%の範囲である。
配合量が、0.001重量%未満では、本発明の効果に乏しく、また20.0重量%を超えても配合量に見合った効果が望めない。

本発明における硫黄化合物の配合量は、0.001〜10.0重量%であり、さらに好ましくは0.01〜1.0重量%である。
配合量が、0.001重量%未満では、本発明の効果に乏しく、また10.0重量%を超えても配合量に見合った効果が望めない。

本発明におけるメラニン生成抑制成分の配合量は、0.001〜20.0重量%であり、さらに好ましくは0.01〜10.0重量%である。

本発明の美白化粧料としては、洗顔料、化粧水、美容液、乳液、クリーム、ジェル、パック、リポソーム、液状、粘土状、ソリッド粉末状化粧料、エアゾール化粧料、ハップ剤等のスキンケア化粧品、下地クリーム、ファンデーション等のメークアップ化粧品をそのまま使用するか、あるいはエアゾール式、ポンプ式、または押出し式、もしくはこれらに類する機構で噴射する化粧品、さらに浴剤等様々な剤型を提供することができる。

ハップ剤においては上記成分に加えて、基剤(カオリン、ベントナイト等)、ゲル化剤(ポリアクリル酸塩、ポリビニルアルコール等)を本発明の目的を達成する範囲内で0.001〜99.9重量%の範囲で配合することができる。

浴剤においては、硫酸塩、炭酸水素塩、ホウ酸塩、色素、保湿剤を、本発明の目的を達成する範囲内で0.001〜99.9重量%の範囲で適宜配合し、パウダータイプ、液剤タイプに調製が可能である。

以下、実施例により本発明の構成を詳細に説明するが、本発明はこれよって限定されるものではない。実施例に先立ち、各実施例で行った試験について説明する。

(1)チロシナーゼ活性阻害作用
チロシナーゼとしてマッシュルーム由来チロシナーゼ(シグマ社)の活性が125U/mlとなる様酵素溶液を調製し、基質としては、0.1%L−ドーパ溶液を調製して、表1に示すチロシナーゼ反応系で経時的に生じるドーパクロム量を475nmの吸光度として測定(マイクロプレートリーダー:BioRad社)し、その反応速度を求めてチロシナーゼ活性として算出した。

レブリン酸(1.25重量%)の添加は、無添加系(精製水)に比べてチロシナーゼ活性をおよそ1/3にまで低下させた。さらに本発明の成分を組合わせた系では、さらに強くチロシナーゼ活性を低下させており、これらの結果は本発明の成分の組合わせによる相乗効果によるものと思われる(巻末表3)。

(2)メラニン産生細胞内のチロシナーゼ合成抑制作用
メラニン産生細胞としてB16マウスメラノーマ細胞を用い、これを96穴マイクロプレートに3万個/ウェルとなる様に播種して24時間プレ培養の後、本発明のレブリン酸またはその塩と、硫黄化合物の1種又は2種以上とを添加した10%牛胎児血清含有イーグルMEM培地に交換して48時間培養した。次に培地を除去して細胞をPBSで洗浄後、1%Triton X−100含有50mMリン酸緩衝液(pH=6.8)50μl と0.1%L−ドーパ溶液を添加して37℃で6時間インキュベーションを行った。インキュベーション後、475nmにおける吸光度をマイクロプレートリーダー(モデル550:Bio Rad社)で測定し、チロシナーゼ合成量として算出した。また別に、先の操作と同様に48時間培養したB16細胞についてMTT法により細胞の活性を測定し、活性あたりのチロシナーゼ合成量として評価判定を行った。

レブリン酸は添加濃度に依存して、B16細胞内のメラニン生成に主要酵素であるチロシナーゼの合成を抑制する傾向を示した。さらに本発明の成分の組合わせにより飛躍的にチロシナーゼの合成を抑制した。この抑制作用は既存の美白剤であるコウジ酸やアルブチンに比べて強かった。これらの結果は本発明の成分の組合わせによる相乗効果によるものと思われる(巻末表4)。

(3)色素沈着改善作用
シミ、ソバカス等色素沈着が認められた被験者10名を1 群とし、色素沈着部に実施例1〜21、及び比較例1〜21の化粧料については、朝晩2回塗布してもらい、実施例22〜24および比較例22〜24の浴剤については、200リットルの湯に対して浴剤50グラムを溶解し、1 日1回入浴使用してもらい、下記の表2の判定基準に基づいて色素沈着改善作用を評価した。
なお、判定は化粧料および浴剤の使用開始2週間後、1 ヵ月後及び3ヵ月後の合計3回行った。
なお、本試験期間を通して皮膚に異常を訴えた者はなかった。
