Description
Claims (20)
- バイオチップに結合される複数の分析物の光学的検出用の解析システムであって、
共焦顕微鏡検出器に結合され、かつ検出器に対してx座標、y座標、およびz座標に沿って移動可能なプラットフォームを備え、該プラットフォームが、ユーザの手動介入なしに自動的にバイオチップを収容するように構成され、
バイオチップが、少なくとも3つの位置合わせマーカを有し、また位置合わせマーカに対する所定位置に複数の分析物をさらに有し、前記解析システムがさらに、
位置合わせマーカを照明して位置合わせマーカ信号を生成する第1の光源および個々の分析物を順次に照明して個別の分析物信号をそれぞれ生成する第2の光源であって、第1の光源と第2の光源は異なるスペクトルプロファイルを有しており、第1の光源の波長は分析物の標識によって実質的に吸収されないものである、前記第1の光源および第2の光源と、
前記分析物の各々のx座標およびy座標に関する位置およびz座標に関する焦点位置を、位置合わせマーカ信号によって確定された位置合わせマーカそれぞれの焦点位置から計算されたバイオチップの焦点面と該複数の位置合わせマーカと複数の分析物との所定の関係を用いて、前記第2の光源による分析物の順次の照明に先立って予測的に確定するように構成されたコンピュータとを備える、解析システム。 - 検出器が、プラットフォームをz座標に沿って移動させることなく、分析物信号の検出を可能にするのに十分である開口数を有する対物レンズまたは対物レンズ系を備える、請求項1に記載の解析システム。
- 第1の光源が、複数の分析物の少なくとも1つの光学的に検出可能な標識の吸収極大と異なる最大波長を有する、請求項1に記載の解析システム。
- 第3の光源をさらに備え、該第3の光源が、複数の分析物の少なくとも1つまたは複数の分析物の別の1つを照明して、第2の分析物信号を生成し、第3の光源が、最大波長を有し、該最大波長が、第1の光源の最大波長とは異なり、かつ複数の分析物の少なくとも1つまたは複数の分析物の別の1つの光学的に検出可能な標識の吸収極大とも異なる、請求項1に記載の解析システム。
- 位置合わせマーカおよび分析物の少なくとも1つが、第1および第2の光源によってそれぞれ異なる角度で照明される、請求項1に記載の解析システム。
- 第1の光源が、レーザまたは発光ダイオードであり、第2の光源が、レーザである、請求項1に記載の解析システム。
- 位置合わせマーカが、蛍光染料、発光化合物、リン光化合物、または反射化合物を含む、請求項1に記載の解析システム。
- 分析物信号が、蛍光信号、化学発光信号、またはリン光信号である、請求項1に記載の解析システム。
- 検出器が、光電子増倍管または電荷結合デバイスを備える、請求項1に記載の解析システム。
- 検出器による分析物の検出のためのz座標に関する焦点位置が、位置合わせマーカからの位置合わせマーカ信号を用いて解析システムによって確定される、請求項1に記載の解析システム。
- 分析物信号が、バイオチップ上のポジティブ制御マーカを用いた解析システムによって正規化される、請求項1に記載の解析システム。
- 検出器に電子的に結合されたデータ転送インタフェースをさらに備える、請求項1に記載の解析システム。
- データ転送インタフェースが、遠隔の場所にあるコンピュータにデータを供給する、請求項12に記載の解析システム。
- ユーザの手動介入なしにプラットフォーム内に自動的に収容されるバイオチップの微小光学解析用の解析システムであって、解析システムは第1の光源および第2の光源を有しており、第1の光源と第2の光源は異なるスペクトルプロファイルを有しており、第1の光源の波長は分析物の標識によって実質的に吸収されないものであり、第1の光源が、バイオチップ上の少なくとも3つの位置合わせマーカを照明して位置合わせマーカ信号を供給し、第2の光源が、個々の分析物を順次に照明して個別の分析物信号を供給し、解析システムがさらに、分析物のx座標およびy座標に関する位置およびz座標に関する焦点位置を、位置合わせマーカ信号によって確定された位置合わせマーカそれぞれの焦点位置から計算されたバイオチップの焦点面と複数の位置合わせマーカと分析物との既知の関係を用いて確定するように構成されたコンピュータを有し、共焦顕微鏡を用いる分析物信号を検出するための各分析物の焦点位置が、位置合わせマーカ信号によって確定された位置合わせマーカそれぞれの焦点位置から計算されたバイオチップの焦点面に基づいて、前記第2の光源による分析物の順次の照明に先立って予測的に確定される、解析システム。
- 分析物信号は、500マイクロメートル以下の径を備える丸い形状を有する、請求項14に記載の解析システム。
- テスト結果が、丸い形状の一部の平均信号値から計算される、請求項15に記載の解析システム。
- 分析物を照明して第2の分析物信号を生成する第3の光源をさらに備える、請求項14に記載の解析システム。
- ユーザの手動介入なしにプラットフォーム内に自動的に収容されるバイオチップの光学的解析用の解析システムであって、バイオチップ上の少なくとも3つの位置合わせマーカを照明する第1の光源を用いる第1の光学サブシステムと、位置合わせマーカ信号を検出する第1の検出器とを備え、システムがさらに、バイオチップ上のプローブまたは分析物を順次照明する第2の光源を用いる第2の光学サブシステムと、プローブまたは分析物信号を検出する第2の検出器とをさらに備え、第1の光源と第2の光源は異なるスペクトルプロファイルを有しており、第1の光源の波長はプローブまたは分析物の標識によって実質的に吸収されないものであり、第1の光学サブシステムが、プローブまたは分析物の各々のx座標およびy座標に関する位置およびz座標に関する焦点位置を、位置合わせマーカ信号によって確定された位置合わせマーカそれぞれの焦点位置から計算されたバイオチップの焦点面と該複数の位置合わせマーカとプローブまたは分析物との既知の関係を用いて、前記第2の光源によるプローブまたは分析物の順次の照明に先立って予測的に確定するように構成されたコンピュータを備え、第2の光学サブシステムが、プローブまたは分析物信号を定量化するのに用いられる、解析システム。
- プラットフォームが、バイオチップを収容し、バイオチップが、プラットフォームをx座標およびy座標に沿って移動させることによって焦点位置に移動させられる、請求項18に記載の解析システム。
- バイオチップが、バイオチップをz座標に沿って移動することなく焦点位置に移動させられる、請求項19に記載の解析システム。
