Description
カナダのアルバータ大学から提供された健常分離ヒト膵島細胞(カナダ、アルバータ大学、ヒト膵島移植プログラム、ジョナサン レイキー(Jonathan RT. Lakey)博士より、万人が入手可能である)から、球形のきれいな形態を呈しているものを実体顕微鏡(STEMI、Carl Zeiss社、ドイツ)下にhand pickup 法にて10個選別し、これをT25培養フラスコに播いた。培養液は、ウイリアムズ ミディアム E(WILLIAM′S MEDIUM E)(シグマ社、セントルイス、米国)に10%ウシ胎仔血清(FCS、シグマ社)、10-7 mol/l インスリン(シグマ社)、10-6 mol/l デキサメタゾン(シグマ社)、25μg/ml 上皮成長因子(EGF、シグマ社)、10mM ニコチンアミド(シグマ社)、抗生物質のペニシリンG/ストレプトマイシン(シグマ社)の組成のものを基本培養液として用いた。播種から3日目までは、基本培養液を4mlとし、培養液の交換はしなかった。4日目より、培養液の交換の際に、基本培養液を2ml、レトロウイルスベクターSSR#69産生細胞の培養上清とレトロウイルスベクターSSR#197産生細胞の培養上清をそれぞれ0.45ミクロンのフィルターにかけたものをそれぞれ1mlずつ加え、計4mlとして4時間インキュベーションすることにより、遺伝子導入を1日2回行なった。この操作を、4、6、8日目(計6回)に、同様に繰り返した。
実施例1で得られた可逆性不死化ヒト膵島細胞株を10万細胞個/mlで、培地として低グルコースDMEM(ギブコ社、オークランド、ニュージャージー州)に10%FCS、10mM ニコチンアミド、ペニシリン G/ ストレプトマイシンの組成のもので培養した。培養開始から24時間で、培地中にCre組み換え酵素の発現アデノウイルスベクターAxCANCreを1モイ(MOI:multiplicity of infection)添加し、その後48時間持続感染を行なった。得られた細胞をPBSで2回洗浄し、培地の交換を行なった。得られた復帰NAKT−13細胞は、図5に示すように、自然と細胞同士が凝集し、正常膵島構造に類似した形態をとるようになるという特徴を有する。
実施例1および2で得られた細胞(各5万細胞個)を、6ウェルプレートの中にカバースライドを敷いたところに播種し、培地として低グルコースDMEM(ギブコ社、オークランド、ニュージャージー州)に10%FCS、10mM ニコチンアミド、ペニシリン G/ ストレプトマイシンの組成のものを用いて、細胞が60%密集となるまで24時間培養した。ついで、培地を高グルコースDMEM(ギブコ社、オークランド、ニュージャージー州)に10%FCS、10mM ニコチンアミド、ペニシリン G/ ストレプトマイシンの組成のものに交換し、6時間培養した。

- 可逆性不死化ヒト膵島細胞株、NAKT−13(寄託機関 独立行政法人産業技術総合研究所 特許生物寄託センター、あて名 日本国茨城県つくば市東1丁目1番地1 中央第6(郵便番号305−8566)、寄託日 平成15年9月4日、受託番号FERM BP−08461)またはその継代株。
- 請求項1記載の可逆性不死化ヒト膵島細胞株またはその継代株からhTERT遺伝子およびSV40T遺伝子を除去することにより得られるヒト膵島細胞。
- 請求項1記載の可逆性不死化ヒト膵島細胞株またはその継代株からhTERT遺伝子およびSV40T遺伝子を除去することにより得られるヒト膵島細胞を含有する糖尿病用治療剤。
- 請求項1記載の可逆性不死化ヒト膵島細胞株もしくはその継代株、または請求項2記載のヒト膵等細胞を用いることを特徴とするインスリンの製造方法。
