Description
(1) ポリリン酸を有効成分として含有する抗真菌剤。
(2) ポリリン酸が、下記一般式:
(4) ポリリン酸がポリリン酸塩である、(1)から(3)のいずれか記載の抗真菌剤。
(5) ヒノキチオールをさらに含む、(1)から(4)のいずれかに記載の抗真菌剤。
(6) 皮膚真菌症の予防及び治療のための、(1)から(5)のいずれかに記載の抗菌剤。
(7) 皮膚真菌症が水虫である、(6)に記載の抗菌剤。
本発明において使用されるポリリン酸は、代表的にはオルトリン酸の脱水縮合によって2個以上のPO4四面体が頂点の酸素原子を共有して直鎖状に連なった構造を有する直鎖状ポリリン酸であるが、側鎖に有機基が導入された側鎖状ポリリン酸、環状ポリリン酸(メタリン酸)、枝分かれ状のリン酸重合体構造のポリリン酸、ウルトラリン酸、あるいはこれらの混合物であってもよい。
(製造例1)ポリリン酸(中鎖ポリリン酸)の製造
食品添加物規格のヘキサメタリン酸ナトリウム20gを精製水200mlに溶解し、これに96%のエタノール32mlを徐々に加えた。これをよく攪拌し室温で30分ほど放置した後,遠心分離(10,000×g、20分、25℃)を行い、水溶液成分と沈殿物とを分離した。水溶液成分を廃棄し、回収した沈殿物に70%エタノールを加えて洗浄し、真空乾燥した。このようにして、9.2gの中鎖(平均鎖長40〜60)ポリリン酸塩を沈殿物として得た(収量46.0%)。
50gの不溶性長鎖ポリリン酸ナトリウム(シグマ社製)を1リットルの0.2N塩酸水溶液に懸濁し、37℃で20分間攪拌しながら加水分解した。その後、5Nの水酸化ナトリウム40mlを加え中和した。中和後の液を遠心し、分解されていない不溶性の長鎖ポリリン酸ナトリウムを沈殿として分離した。加水分解後の水溶性長鎖ポリリン酸ナトリウムを含む上清に208mlのエタノールを加えて再び遠心し、水溶性の長鎖ポリリン酸(平均鎖長約300)を沈殿として回収した。
(実施例1)ポリリン酸ナトリウムによる白癬菌の増殖阻害
Trichophyton mentagrophytes IFO 7522株を用い、ポリリン酸ナトリウムによる菌体増殖阻害効果を調べた。各最終濃度(0, 0.015, 0.03, 0.06, 0.125%)のポリリン酸ナトリウム(平均鎖長40)を含むポテトデキストロース寒天プレート(直径10 cm)を作製した。ポテトデキストロース寒天培地上で充分に生育したT. mentagrophytes IFO 7522株の菌糸をコルクポーラーを用いて型抜きし、直径8ミリメートルのディスク状に成形した。このディスクを上記の寒天プレートの中央に接種して25℃で培養を行い、菌体増殖の様子を視覚的に確認した。
T. mentagrophytes IFO 5466株を用いて実施例1と同様の方法により、各種鎖長をもつポリリン酸ナトリウムの菌体増殖阻害効果について調べた。はじめに各最終濃度(0, 0.03, 0.06, 0.12%)の各種鎖長をもつポリリン酸ナトリウム [トリポリリン酸ナトリウム (P3)、平均鎖長15 (P15)、40 (P40)、60 (P60)、300 (P300)]を含むポテトデキストロース寒天プレート(直径6 cm)を作製した。比較群として同じ濃度のオルトリン酸 (P1)、及びピロリン酸 (P2) を含むプレートを作製した。次に、直径6ミリメートルのディスク状に成形したT. mentagrophytes IFO 5466株の菌糸を上記のプレート中央に接種して25℃で培養を行い、菌体増殖の様子をコロニーの直径を測定することにより経時的に観察した。同時にスキャナを用いて菌体の増殖を視覚的に確認した。
T. mentagrophytes IFO 5466株を用い、菌体増殖阻害におけるポリリン酸ナトリウムと既存の抗菌性物質との相乗効果を調べるため、実施例1と同様の方法により菌体増殖阻害実験を行った。本実施例ではバクテリアのほかCandidaやTricophytonなどの真菌に対して抗菌活性をもつ物質として知られるヒノキチオールを用い、ポリリン酸ナトリウムと併用した場合のT. mentagrophytesの増殖阻害に対する相乗効果について調べた。各最終濃度(0, 0.00025, 0.0005, 0.001%)のヒノキチオール及び各種濃度(0, 0.03, 0.06%)のポリリン酸ナトリウム(平均鎖長40)を含むポテトデキストロース寒天プレート(直径10 cm)を作製し、直径6ミリメートルのディスク状に成形したT. mentagrophytes IFO 5466株の菌糸をプレート中央に接種して25℃で培養を行い、菌体増殖の様子をコロニーの直径を測定することにより経時的に観察した。同時にデジタルカメラ及びスキャナを用いて菌体の増殖を視覚的に確認した。
Candida albicans IFO 0579株を用い、ポリリン酸ナトリウムによる増殖阻害効果を調べた。各最終濃度(0, 0.06, 0.12%)のポリリン酸ナトリウム(平均鎖長40)を含むポテトデキストロース液体培地を作製した。ポテトデキストロース液体培地中で一晩培養を行い充分に生育したCandida albicans IFO 0579株を、上記のポリリン酸ナトリウムを含む液体培地に1/100希釈で接種して23℃で24時間振盪培養を行い、菌体増殖の様子を660 nmにおける培養液の濁度を経時的に測定することにより確認した。
出芽酵母(Saccaromyces cerevisiae)YPH499株を用い、ポリリン酸ナトリウムの増殖阻害効果について調べた。各最終濃度(0, 0.03, 0.06, 0.12, 0.25 %)のポリリン酸ナトリウム(平均鎖長40)を含むYPD液体培地を作製した。YPD液体培地中で一晩培養を行い充分に生育したYPH499を、上記のポリリン酸ナトリウムを含む液体培地に一定量接種して30 ℃で24時間振盪培養を行い、菌体増殖の様子を660 nmにおける培養液の濁度を経時的に測定することにより確認した。
出芽酵母(Saccaromyces cerevisiae) YPH499株を用い、ポリリン酸ナトリウム(平均鎖長40)による抗真菌剤アンフォテリシンB(ファンジソン)の抗菌効果の増強に関して調べた。YPH499株の一晩培養液を10-7倍までYPD液体培地で希釈し、その希釈液(0.1 ml)をファンジソン、ポリリン酸ナトリウム、ファンジソン及びポリリン酸ナトリウムを含むYPD寒天培地(プレート)上に塗布することによって菌体を播種した。30℃で一晩培養後、プレート上のコロニー数を計測し、薬剤を含まないYPD寒天培地上で生育したコロニー数を100%として、それぞれの薬剤を含むプレート上での菌の生存率を算出した。結果を表2に示した。
Claims (5)
- 下記一般式:

(式中、nは40〜300の整数を表す)で表される直鎖状リン酸の1種又は2種以上の混合物からなるポリリン酸を有効成分として含有し、その含有量が0.1〜5重量%である、抗白癬菌剤。 - ポリリン酸がポリリン酸塩である、請求項1に記載の抗白癬菌剤。
- ヒノキチオールをさらに含む、請求項1または2に記載の抗白癬菌剤。
- ポリリン酸とヒノキチオールの配合比が濃度比で1:0.004〜1:0.02である、請求項3に記載の抗白癬菌剤。
- 足白癬(水虫)、頭部白癬(シラクモ)、体部白癬(ゼニタムシ)、股部白癬(インキンタムシ)、爪白癬、および手白癬から成る群から選択される皮膚真菌症の予防及び治療のための、請求項1から4のいずれか1項に記載の抗白癬菌剤。
