【発明の属する技術分野】
本発明は、テストステロン5α−レダクターゼ阻害作用及び/又はアンドロゲン受容体結合阻害作用を通じて、テストステロンの活性発現を阻害することができる抗男性ホルモン剤、及びこの抗男性ホルモン剤を含有し、男性ホルモン依存型の疾患である男性型禿頭、多毛症、脂漏症、座瘡、前立腺肥大症、前立腺腫瘍、男児性早熟等の予防・改善に有効な養毛化粧料、皮脂分泌抑制剤、並びに前立腺肥大抑制剤に関するものである。

【従来の技術】
多くのステロイドホルモンは産生臓器から分泌された分子型で受容体と結合してその作用を発現するが、アンドロゲンと総称される男性ホルモンの場合、例えば、テストステロンは標的臓器の細胞内に入ってテストステロン5α−レダクターゼにより5α−ジヒドロテストステロン(5α−DHT)に還元されてから受容体と結合し、アンドロゲンとしての作用を発現する。

前記アンドロゲンは重要なホルモンであるが、それが過度に作用すると、男性型禿頭、多毛症、脂漏症、座瘡、前立腺肥大症、前立腺腫瘍、男児性早熟などの様々な好ましくない症状を誘発する。そこで、過剰のアンドロゲンの作用を抑制することによりこれら好ましくない症状を改善する手段が検討されている。具体的には、テストステロンを活性型5α−DHTに還元するテストステロン5α−レダクターゼの作用を阻害することにより活性な5α−DHTを生じるのを抑制する方法と、テストステロンから生じた5α−DHTが受容体と結合するのを阻害することによりアンドロゲン活性を発現させない方法とが提案されている。

その結果、シプロテンアセテート、オキセンドロン、酢酸クロマジノン等の有効性が確認されたが、これらはステロイドホルモン誘導体であるホルモン様作用等の好ましくない副作用を有するという欠点がある。

また、幾つかの生薬や植物体からの抽出物について、テストステロン5α−レダクターゼ阻害作用(特開昭60−1466829号公報等参照)、5α−DHTとその受容体との結合を阻害する作用(特開昭64−3126号公報等参照)が知られている。

【発明が解決しようとする課題】
しかしながら、安全性、及び生産性に優れ、かつ安価でありながら、高いテストステロン5α−レダクターゼ阻害作用及び/又はアンドロゲン受容体結合阻害作用を有する抗男性ホルモン剤に対する需要者の要望は極めて強く、未だ十分満足し得るものが提供されていないのが現状である。

本発明は、このような状況下、従来における諸問題を解決し、以下の目的を達成することを課題とする。
即ち、本発明は、テストステロン5α−レダクターゼ阻害物質及び/又はアンドロゲン受容体結合阻害物質を有効成分として含む新規、かつ強力で安全な抗男性ホルモン剤、及びこの抗男性ホルモン剤を含有する養毛化粧料、皮脂分泌抑制剤、並びに前立腺肥大抑制剤を提供することを目的とする。

【課題を解決するための手段】
本発明者は、上記課題を解決するため、安全な数多くの植物類の中から、抗男性ホルモン効果を有するものについて鋭意検討を重ねた結果、ムラサキ科チシャノキ属(Ehretia)植物の抽出物が、優れたテストステロン5α−レダクターゼ阻害作用及びアンドロゲン受容体結合阻害作用を有することを知見した。

即ち、ムラサキ科チシャノキ属(Ehretia)植物の抽出物、特にチャングバットの葉部、茎部、枝部又はこれらの混合部位の抽出物が、異なる作用点で男性ホルモン作用の発現を抑制するテストステロン5α−レダクターゼ阻害作用及びアンドロゲン受容体結合阻害作用を有し、男性ホルモン依存型の疾患である男性型禿頭、多毛症、脂漏症、座瘡、前立腺肥大症、前立腺腫瘍、男児性早熟等の予防、改善に有効な骨粗鬆症の予防や治療に有効な養毛化粧料、皮脂分泌抑制剤又は前立腺肥大抑制剤が得られることを見出し、本発明をなすに至った。

従って、発明は、下記の抗男性ホルモン剤、及び養毛化粧料、皮脂分泌抑制剤並びに前立腺肥大抑制剤を提供する。

請求項1の発明は、ムラサキ科チシャノキ属植物から抽出されるテストステロン5α−レダクターゼ阻害物質を有効成分として含むことを特徴とする抗男性ホルモン剤である。

請求項2の発明は、ムラサキ科チシャノキ属植物から抽出されるアンドロゲン受容体結合阻害物質を有効成分として含むことを特徴とする抗男性ホルモン剤である。

請求項3の発明は、ムラサキ科チシャノキ属植物が、チャングバット、マルバチチシャノキ、リュウキュウチシャノキ及びチシャノキから選ばれる1種又は2種以上である請求項1又は2記載の抗男性ホルモン剤である。

請求項4の発明は、ムラサキ科チシャノキ属植物として、チャングバットの葉部、茎部、枝部又はこれらの混合部位を用いる請求項3記載の抗男性ホルモン剤である。

なお、本発明の抗男性ホルモン剤の抽出原料であるムラサキ科チシャノキ属植物(特にチャングバット)は、民間薬、特に茶として飲用されてきた安全性の高いものであるが、異なる作用点であるテストステロン5α−レダクターゼ阻害作用及びアンドロゲン受容体結合阻害作用を有し、男性ホルモン依存型の疾患である男性型禿頭、多毛症、脂漏症、座瘡、前立腺肥大症、前立腺腫瘍、男児性早熟等の予防、改善に有効であることは全く知られておらず、このことは本発明者の新知見である。

【発明の実施の形態】
以下、本発明について更に詳しく説明する。
本発明の抗男性ホルモン剤は、ムラサキ科チシャノキ属植物から抽出されるテストステロン5α−レダクターゼ阻害物質及びアンドロゲン受容体結合阻害物質を有効成分として含むものである。

ここで、ムラサキ科チシャノキ属(Ehretia)植物としては、チャングバット(Ehretia microphylla Lam.)、マルバチチシャノキ(Ehretia dicksonii Hance)、リュウキュウチシャノキ(Ehretia dichotoma Bl.)及びチシャノキ(Ehretia ovalifolia Hassk.)から選ばれる1種又は2種以上が挙げられ、これらの中でも、チャングバットが好ましい。

チャングバット(日本名「フクマンギ」と呼ばれる)は、ムラサキ科チシャノキ属に属する高さ1〜3mの常緑低木で、奄美大島以南の琉球、中国南部、マレーシア、インドに分布する。葉は濃緑色で短枝に束生し、倒卵形で長さ2〜5cmである。

このチャングバットは民間薬、特に茶として飲用されてきたが、テストステロン5α−レダクターゼ阻害作用及びアンドロゲン受容体結合阻害作用を有することは全く知られていない。

本発明において、テストステロン5α−レダクターゼ阻害物質及びアンドロゲン受容体結合阻害物質を抽出するにあたり特殊な抽出方法を採用する必要はなく、室温乃至還流加熱下で、任意の装置を用いて抽出することができる。

具体的には、抽出溶媒を満たした処理槽に、ムラサキ科チシャノキ属植物(特にチャングバットの葉部、茎部、枝部又はこれらの混合部位)を投入し、必要に応じて時々攪拌しながら、30分〜2時間静置して可溶性成分を溶出した後、濾過して固形物を除去し、得られた抽出液から抽出溶媒を溜去し、乾燥することにより抽出物が得られる。抽出溶媒量は通常、抽出原料の5〜15倍量(質量比)であり、抽出条件は、抽出溶媒として水を用いた場合には、通常50〜95℃で1〜4時間程度である。また、抽出溶媒として水とエタノールとの混合溶媒を用いた場合には、通常40〜80℃で30分〜4時間程度である。なお、溶媒で抽出することにより得られる抽出液は、抽出溶媒が安全性の高いものであればそのまま配合して本発明の抗男性ホルモン剤の有効成分として用いることができるが、濃縮液又はその乾燥物としたもののほうが利用しやすい。

〔製造例1〕 チャングバット抽出物
チャングバット(学名:Ehretia microphylla)の各部位(葉、茎及び枝)の乾燥物を細切りしたもの100gに抽出溶媒である50質量%エタノール(水とエタノールとの質量比1:1)1000mLを加え、還流抽出器で80℃、2時間加熱抽出し、熱時濾過した。残渣について同様の抽出処理を行った。得られた2回の抽出液を合わせて減圧下に濃縮し、乾燥して各部位の抽出物を得た。各部位毎の抽出物の収率は表1に示したとおりであった。

〔実施例1〕
製造例1のチャングバットの各部位抽出物について、下記の試験法によりテストステロン5α−レダクターゼ阻害作用を試験した。

テストステロン(東京化成(株)製)4.2mgをプロピレングリコール1mLに溶解し、その20μLに、1mg/mLのNADPH含有5mMトリス塩酸緩衝液(pH7.2)825μLを加えて混合した。

更に、各試料溶液80μL及びS−9(ラット肝臓ホモジネート:オリエンタル酵母(株)製)75μLを加えて混合し、37℃で30分間インキュべ−トした。その後、塩化メチレン1mLを加えて反応を停止させ、激しく振とうした。
その後、遠心分離し、塩化メチレン層を分取して、反応生成物であるジヒドロキシテストステロン、アンドロスタンジオール等の反応生成物をガスクロマトグラフィーにより定量した。

〔実施例2〕
製造例1のチャングバットの各部位抽出物について、下記の試験法によりアンドロゲン受容体結合阻害作用を試験した。

アンドロゲン依存性マウス乳癌細胞SC−3細胞を、2%FBS含有MEM培地(以下、MEM−2と略記する)を用いて1.0×104cells/well/100μLの細胞密度にて96穴マイクロプレートに播種し、37℃、5%CO2−95%airの下で培養した。24時間後、試料及び10−9モル濃度のDHTを添加した0.5%BSA含有Ham F12+MEM培地(以下、HMB培地と略記する)に培地を交換して48時間培養した。

その後、培地を0.97mM MTTを含むMEM−2培地に交換し、2時間培養後、培地をイソプロパノールに交換して細胞内に生成したブルーホルマザンを抽出した。溶出したブルーホルマザンを含有するイソプロパノールについて、ブルーホルマザンの吸収極大点がある570nmの吸光度を測定した。
