Description
本国際出願特許請求は、2001年1月24日出願の米国仮特許出願第60/263894号の利益を主張する2002年1月23日出願の米国特許出願第10/057554号の一部継続出願に基づく。本出願は、2004年1月16日出願の米国仮特許出願第60/536899号、および2004年9月15日出願の米国特許出願第10/941443号の優先権を主張する。この親明細書はそれぞれ、その全体が参照により本明細書に組み込まれている。
この開発は、NIDCR(国立歯科頭蓋顔面研究所)からアメリカ歯科協会基金へのUSPHS(アメリカ公衆衛生局)研究補助金DE11789号により一部援助された。アメリカ合衆国またはその一政府機関は、本特許請求発明に対してある種の権利を有し得る。
本発明の種々の実施形態は、自硬性のリン酸カルシウム含有、および/またはカルシウム含有ペーストならびにセメント組成物に関する。この組成物を使用して、骨および歯の修復ならびに類似用途のペーストを形成でき、その際このペーストは修復部位まで送達された後所望の時間内に硬化する。
本発明の種々の実施形態は、包装において安定であり、ウォッシュアウトへの抵抗性があり、また欠損部またはインプラント部位に送られた後、所望の時間内に硬化するリン酸カルシウムセメントペーストを混合するための組成物および手段を含む。リン酸カルシウムセメント粉末とブレンドすると、リン酸カルシウムセメントの急速な凝結を助長する溶液が提供される。この溶液および粉末は安定であり、一緒にブレンドした場合に、硬化時間が短縮されたセメントスラリをもたらす。
周囲温度(すなわち室温または体温)で自己凝結(self−set)してヒドロキシアパタイト(HA)となる生成物をもたらすリン酸カルシウムセメントを調製する。このリン酸カルシウムセメント組成物は、液体溶液および粉末組成物を含む。ある種のリン酸カルシウム前駆体セメントスラリ組成物を使用すると、確実にかつ急速に凝結してHAとなるセメントがもたらされる。種々の最終用途向けに凝結速度を調節することができ、必要に応じて極めて急速とすることができる。得られたヒドロキシアパタイトセメントは、生体適合性および再吸収性(生分解性)の両方を有し、それにより生体骨と接触した場合骨が置換されると考えられる。
下記の実施例は、本発明の好ましい実施形態をさらに例示しているが、添付する特許請求範囲に示している本発明の範囲を決して限定するものではないと解釈すべきである。
当分野の技術者に知られている下記の従来の方法を使用して、TTCP、α−TCP、およびβ−TCPを調製する。DCPA、DCPD、HA、CaCO3、およびCa(OH)2は、J.T.Baker Chemical Co.、Phillipsburg、NJから入手可能なものなどの、市販の試薬級薬品である。
示した化合物を適正な量で蒸留水に溶解して下記に示した濃度を得ることにより、種々のセメント溶液を調製する。実施例1および2の溶液は、当分野の技術者に知られており、また文献中に記載されている従来の方法を使用して調製する。全ての他の溶液は、示した化合物を適正な量で蒸留水に溶解して、下記の表に示した濃度を得ることにより調製する。
選択した溶液および粉末を約0.25〜約0.5の質量比でブレンドして、均質なペースト組成物を形成させる。このペーストを型枠に入れ、この型枠を温度約37℃および湿度100%に保持して、欠損部位における予想される条件を模している。
この実施例では、セメント液としてpH約1、カルシウム濃度約0.66M、およびリン濃度約7.6Mを有するMCPM−形成溶液を使用する。この溶液は、H3PO4溶液(約6.3M)中で過剰量のMCPMを平衡するまで攪拌し、続いて濾過することにより調製する。比較用凝結時間を提供するため、粉末は水とも混合する。
この実施例では、セメント液としてpH約2.1、カルシウム濃度約1.5M、およびリン濃度約4.4Mを有するDCPD−形成溶液を使用する。この溶液は、H3PO4溶液(約2.9M)中で過剰量のDCPDを平衡するまで攪拌し、続いて濾過することにより調製する。比較用凝結時間を提供するため、粉末は水とも混合する。
この実施例は、凝結中のセメント液のリン酸塩濃度に及ぼす、実施例4の溶液の影響を例証する。
Claims (15)
- 歯科用修復ならびに骨インプラントおよび修復向けの物質の組成物であって、
前記組成物が、
無水リン酸二カルシウム、リン酸二カルシウム二水和物、α−リン酸三カルシウム、β−リン酸三カルシウム、リン酸四カルシウム、およびこれらの組合せからなる群から選択される1種または複数のリン酸カルシウム塩を含む粉末状カルシウム化合物と、
前記粉末状カルシウム化合物と混合するための、リン酸一カルシウム一水和物、無水リン酸一カルシウム、またはこれらの組合せに関して飽和している別個の酸性リン酸カルシウム溶液
とを含み、
前記酸性リン酸カルシウム溶液と混合した後の前記粉末状カルシウム化合物が、前記粉末状カルシウム化合物とのペーストを形成する際にpH 0〜1.9の範囲の初期pHを有し、混合後にpH 4.5〜14の範囲の上昇したpHを有し、これにより、ヒドロキシアパタイトセメントの形成を可能にする、
組成物。 - 前記粉末状カルシウム化合物が、リン酸八カルシウム、非晶質リン酸カルシウム、リン酸一カルシウム一水和物、無水リン酸一カルシウム、酸化カルシウム、水酸化カルシウム、炭酸カルシウム、グリセロリン酸カルシウム、グルコン酸カルシウム、硫酸カルシウム、乳酸カルシウム、およびこれらの組合せからなる群から選択される1種または複数のカルシウム塩を含む、請求項1に記載の組成物。
- 薬物、充填材料、結晶調整剤、粘度調節剤、ゲル化剤、増孔剤、再吸収性および非再吸収性繊維およびメッシュ、骨誘導因子、骨形態形成タンパク質、ならびに他のタンパク質からなる群から選択される1種または複数の添加剤を含む、請求項1に記載の組成物。
- 前記1種または複数の添加剤が、ヒドロキシプロピルメチルセルロース、カルボキシメチルセルロース、キトサン、キトサン誘導体、コラーゲン、ガム、ゼラチン、およびアルギン酸塩、ならびにこれらの組合せからなる群から選択されるゲル化剤である、請求項3に記載の組成物。
- 前記1種または複数の添加剤が充填材料である、請求項3に記載の組成物。
- 前記1種または複数の添加剤が増孔剤である、請求項3に記載の組成物。
- 歯科用修復ならびに骨インプラントおよび修復向けの物質の組成物であって、
前記組成物が、
無水リン酸二カルシウム、リン酸二カルシウム二水和物、α−リン酸三カルシウム、β−リン酸三カルシウム、ヒドロキシアパタイト、リン酸四カルシウム、およびこれらの組合せからなる群から選択される1種または複数のリン酸カルシウム塩を含む粉末状カルシウム化合物と、
前記粉末状カルシウム化合物と混合するための、リン酸二カルシウム二水和物、無水リン酸二カルシウム、またはこれらの組合せに関して飽和している別個の酸性リン酸カルシウム溶液
とを含み、
前記酸性リン酸カルシウム溶液と混合した後の前記粉末状カルシウム化合物が、前記粉末状カルシウム化合物とのペーストを形成する際にpH 1.9〜4.3の範囲の初期pHを有し、混合後にpH 4.5〜14の範囲の上昇したpHを有し、これにより、ヒドロキシアパタイトセメントの形成を可能にする、
組成物。 - 前記粉末状カルシウム化合物が、リン酸八カルシウム、非晶質リン酸カルシウム、リン酸一カルシウム一水和物、無水リン酸一カルシウム、酸化カルシウム、水酸化カルシウム、炭酸カルシウム、グリセロリン酸カルシウム、グルコン酸カルシウム、硫酸カルシウム、乳酸カルシウム、およびこれらの組合せからなる群から選択される1種または複数のカルシウム塩を含む、請求項7に記載の組成物。
- 薬物、充填材料、結晶調整剤、粘度調節剤、ゲル化剤、増孔剤、再吸収性および非再吸収性繊維およびメッシュ、骨誘導因子、骨形態形成タンパク質、ならびに他のタンパク質からなる群から選択される1種または複数の添加剤を含む、請求項7に記載の組成物。

- 前記1種または複数の添加剤が充填材料である、請求項9に記載の組成物。
- 前記1種または複数の添加剤が増孔剤である、請求項9に記載の組成物。
- 骨および歯の欠損部を修復するために用いる請求項1および7のいずれか1項に記載の組成物であって、前記組成物が硬化する前に前記欠損部が前記組成物で充填される、組成物。
- 骨および歯の欠損部を修復するためのセメントの製造における、請求項1および7のいずれか1項に記載の組成物の使用。
- リン酸カルシウム骨セメントであって、周囲温度で自己硬化しまた生体骨と接触させてインプラントする場合再吸収可能であり、請求項1および7のいずれか1項に記載の組成物を含む骨セメント。
