本発明は口腔用組成物に関し、苦み成分による悪い使用感が改善されている口腔用組成物に関する。

【従来の技術】
薬効剤或いは特徴成分として無機系化合物、有機系化合物及び抽出物は、口腔用組成物に数多く配合されている。しかし口腔用組成物にこれらの成分を配合すると、中には苦みを有する成分があり、口に含んだ際に苦みを感じ、使用感を損ねることがある。そのため使用性から、これらの苦味を有する薬効剤或いは特徴成分を配合し難いことが問題であった。

本発明は、苦み成分を含みながらも使用感が改善された口腔用組成物を提供することを目的とする。

本発明者らは上記課題を解決するために鋭意検討した結果、苦み成分に対して重曹を1:1若しくはそれ以上の配合比率で配合することによって、苦みがマスキングされ、目的が達成されることを知見した。
従って、本発明は苦み成分として下記の(a)成分、(b)成分及び(c)成分からなる群から選ばれる少なくとも1種を含有し、該苦み成分1質量部に対して重曹を1質量部以上含有する口腔用組成物に関する。

【発明の実施の形態】
本明細書中でいう口腔用組成物とは、練歯磨剤、液状歯磨剤、粉歯磨剤、歯肉塗布剤(口腔用パスタ剤)などを含む。
本発明の口腔用組成物に用いる、苦み成分(a)の無機系化合物の具体例として塩化カリウム、硝酸カリウム、塩化亜鉛、結晶セルロース及び塩化マグネシウム等が挙げられる。中でも特に塩化カリウム、硝酸カリウム及び塩化亜鉛が挙げられる。
塩化カリウムは,天然にはカリ岩塩として,塩化ナトリウムや塩化マグネシウムと混在して産出される。口腔用組成物においては,収斂剤等の目的で配合される。
硝酸カリウムは硝石として天然にインド、エジプト、ベンガル地方に産出する。口腔用組成物においては収斂剤、象牙質知覚過敏症予防等の目的で配合される。

塩化亜鉛は、亜鉛又は酸化亜鉛を塩酸に溶かした溶液を蒸発して得られる白色粉末、粒状の結晶であり、口腔用組成物においては収斂剤、象牙質知覚過敏症予防等の目的で配合される。
このような塩化カリウム、硝酸カリウム若しくは塩化亜鉛は、医薬品や化粧品をはじめ、食品、雑貨品、農芸化学品など広く一般的に使用されるものであり、本発明においてはこれらに使用されている天然品、合成品等のいずれの市販品も使用することができる。
その他、結晶セルロース、塩化マグネシウムも市販品を使用することができる。上記成分(a)を1種単独で、又は2種以上を使用することができる。
本発明の口腔用組成物における苦み成分(a)の含有量は、口腔用組成物の全質量に対しては1〜30質量%程度が適当であり、好ましくは3〜20質量%である。この量が1質量%未満であると期待される効果が発揮されず、一方30質量%を越えると含有量に見合った効果が得られず使用性を損ねる場合がある。

本発明の口腔用組成物に用いる、苦み成分(b)の有機系化合物の具体例としてはポリエチレングリコール、1,3−ブチレングリコール、プロピレングリコール、ラウロマクロゴール、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル等が挙げられる。中でもポリエチレングリコール及び1,3−ブチレングリコールが挙げられる。
ポリエチレングリコールは、水またはエチレングリコールに酸化エチレンを付加させた重合体で、H(OCH2CH2)nOHで表される。この重合体は酸化エチレンの付加量によって液状の低分子量のものから固体の高分子量のものまで各種分子量の重合物ができ、その生成物は種々の重合度のものが混合している。
1,3−ブチレングリコールは、C4H10O2で表される無色透明の粘性の液体である。

このようなポリエチレングリコール、1,3−ブチレングリコールは医薬品や化粧品をはじめ、繊維・ゴム・接着剤・金属・窯業・石油・印刷・インキ工業などに広く一般的に使用されるものであり、本発明においてはこれらに使用されている市販品も用いることができる。この他、プロピレングリコール、ラウロマクロゴール、ショ糖脂肪酸エステル、グリセリン脂肪酸エステル等も市販品を用いることができる。上記成分(b)を1種単独で、又は2種以上を使用することができる。
本発明の口腔用組成物における苦み成分(b)の含有量は、口腔用組成物の全質量に対して5〜50質量%が適当であり、好ましくは5〜30質量%である。この量が50質量%を越えると、口腔用組成物の剤形を保つことが難しく、使用性を損ねる場合がある。

本発明の口腔用組成物に用いる、苦み成分(c)の抽出物には、植物からの抽出物やプロポリスが挙げられる。更に具体的には、ティートリーオイル、プロポリス、ユーカリ油、茶エキス、ユッカエキス、チョウジ油、黄連末及びウーロン茶エキス等が揚げられる。中でも特にティートリーオイル、プロポリス、ユーカリ油、茶エキス及びユッカエキスが挙げられる。
ティートリーオイルは、学名がメラルカアルターニフォリア(Melaleuca alternifolia)というオーストラリア原産のフトモモ科の木から取れる精油分のことである。口腔用組成物には、天然由来の殺菌剤、抗菌剤、矯味剤として配合される。

プロポリスは、花や樹皮のヤニと蜜蜂の唾液を混ぜ合わせた物質若しくは抽出物のことであり、その主要成分はミネラルやアミノ酸、フラボノイド・ジテルペン、ケルセチン等である。口腔用組成物には抗炎症剤、矯味剤として配合される。
ユーカリ油は、ユーカリプトール(Eucalyptus globulusLabillardiere)又はその他近縁植物(Myrtaceae)の葉を水蒸気蒸留して得た精油分のことである。口腔用組成物には、天然由来の殺菌剤、香味剤、矯味剤等として配合される。
茶エキスは、チャノキ(Theaceae)の葉から製したもの(緑茶)若しくは乾燥させたもの、或いは水、エタノール、プロピレングリコール溶液にて抽出して得られるエキスのことである。口腔用組成物には、天然由来の殺菌剤、消臭剤、矯味剤等として配合される。

ユッカエキスとはユッカより得られる抽出物のことであり、原料であるユッカ(Yucca)としては、Yucca arizonica、Yucca brevifoia、Yucca schidigera、Yucca elata、Yucca intermdia、Yucca recurvifolia Salisb、Yucca smalliana Fernald、Yuccaaloifolia Linn、Yucca gloriosa Linn、Yucca mohavensis、Yucca peninsularis、Yucca schottii、Yucca whipplei等があり、これらの一種以上を用いることができる。
ユッカは地上茎、地上部(種子を含む)とも抽出原料として使用することができる。口腔用組成物には、天然由来の殺菌剤、抗菌剤、抗う蝕剤等として配合される。
市販品としては、例えばミツバ貿易(株)から入手可能なユッカフォーメーション50、ユッカエキスパウダー、ユッカサラサポニン50−M等が挙げられる。

このようなティートリーオイル、プロポリス、ユーカリ油、茶エキス若しくはユッカエキスは医薬品や化粧品をはじめ、食品、農芸化学品などに広く一般的に使用されるものであり、本発明においてはこれらに使用されている市販品を用いることができる。この他、チョウジ油、黄連末、ウーロン茶エキス等も市販品を使用することができる。上記成分(c)を1種単独で、又は2種以上を使用することができる。
本発明の口腔用組成物における苦み成分(c)の含有量は、口腔用組成物の全質量に対して0.01〜10質量%が適当であり、好ましくは0.1〜5質量%である。この量が0.01質量%未満であると期待される効果が発揮されず、一方10質量%を越えると、含有量に見合った効果が得られず使用性を損ねる場合がある。
本発明の口腔用組成物には、上記苦み成分(a)〜(c)から1種単独又は2種以上を使用することができる。

本発明の口腔用組成物に用いる重曹は、医薬品や化粧品をはじめ、食品、雑貨品、農芸化学品など広く一般的に使用されるものであり、本発明においてはこれらに使用されている市販品を用いることができる。重曹は炭酸水素ナトリウム、重炭酸ソーダとも言う。
本発明の口腔用組成物における重曹の含有量は、重量で苦み成分に対して1:1若しくはそれ以上の配合比率で配合することができる。すなわち本発明の口腔用組成物に苦み成分1質量部に対して、重曹を1質量部以上、好ましくは1〜10質量部で含有させる。口腔用組成物の全質量に対して重曹の含有量は、0.1〜50質量%程度が適当であり、好ましくは1〜30質量%である。上記の範囲にあると、充分な効果が発揮されるとともに使用性も適当である。

本発明の口腔用組成物は、上述の配合比率において成分を常法に従って配合し、練歯磨剤、液状歯磨剤、粉歯磨剤、歯肉塗布剤(口腔用パスタ剤)等の剤型とすることにより調製される。本発明の口腔用組成物はその剤型に応じて各種の添加剤を配合することができる。例えば歯磨剤とする場合には、水不溶性研磨剤の他、粘結剤、湿潤剤、界面活性剤、香料、甘味料、殺菌剤、防腐剤、色素、水、水溶性フッ化物、シリコーン、その他の有効成分等が適宜用いられる。

水不溶性研磨剤としては、歯磨用リン酸水素カルシウム、重質炭酸カルシウム、軽質炭酸カルシウム、ピロリン酸カルシウム、不溶性メタリン酸ナトリウム、メタリン酸カリウム、無水ケイ酸、含水ケイ酸、ケイ酸アルミニウム、ケイ酸カルシウム、ケイ酸ジルコニウム、ベントナイト、ゼオライト、酸化アルミニウム、水酸化アルミニウム、アルミナ、レジン及びこれらの混合物等が挙げられる。これらのうち、無水ケイ酸、含水ケイ酸、歯磨用リン酸水素カルシウム、ピロリン酸カルシウム、ケイ酸アルミニウム、重質炭酸カルシウム、軽質炭酸カルシウム、水酸化アルミニウム、アルミナが貯蔵安定性の良い歯磨剤を得るためには特に好ましい。

粘結剤としては、カルボキシメチルセルロースナトリウム、メチルセルロース、ヒドロキシエチルセルロース、キサンタンガム、アルギン酸塩、カラギーナン、アラビアゴム、ポリビニルアルコール、トラガントガム、デンプン、ポリアクリル酸ナトリウム等が挙げられる。これらのうち、カルボキシメチルセルロースナトリウム、カラギーナン、キサンタンガム等が特に好ましい。
湿潤剤としては、ソルビトール、グリセリン、マルチトール、キシリトール等が挙げられる。これらのうち、グリセリン、ソルビトール等が特に好ましい。

界面活性剤は、発泡剤又は油状物質の安定化剤として用いられるものであり、各種界面活性剤が用いられる。好ましい界面活性剤の例としては、アルキル硫酸塩、アルキルベンゼンスルホン酸塩、モノアルキルリン酸エステル塩、N−アシルザルコシン酸塩、N−アシルグルタミン酸塩、ポリオキシエチレン硬化ヒマシ油、ポリオキシエチレンポリオキシプロピレンブロック共重合体(プルロニック型)、アルキルグルコシド類、ソルビタン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル、アルキルジメチルアミンオキサイド、カルボベタイン、ヒドロキシカルボベタイン、ホスホベタイン、ヒドロキシホスホベタイン、スルホベタイン、ヒドロキシスルホベタイン等が挙げられる。これらのうち、アルキル硫酸塩、ポリオキシエチレン硬化ヒマシ油、アルキルグルコシド類、ソルビタン脂肪酸エステル、ポリオキシエチレンソルビタン脂肪酸エステル等が特に好ましい。

上記香料としては、スペアミント油、ペパーミント油、ウインダーグリーン油、サッサフラス油、セージ油、マヨナラ油、肉桂油、タイム油、レモン油及びオレンジ油等の天然香料;及び1−メントール、アネトール、カルボン、オイゲノール、チモール、サリチル酸メチル等の合成香料が挙げられる。
甘味料としては、サッカリン、サッカリンナトリウム、ステビオサイド、ネオヘスペリジルジヒドロカルコン、ベルラルチン、p−メトキシシンナミックアルデヒド、グリチルリチン酸塩、アスパルテーム(アスパルチルフェニルアラニンメチルエステル)等が挙げられる。

殺菌剤としては、クロルヘキシジンの塩類、塩化セチルピリジニウム、塩化ベンザルコニウム、塩化ベンゼトニウム、イソプロピルメチルフェノール、トリクロサン等が挙げられる。
防腐剤としては、安息香酸、安息香酸ナトリウム、パラヒドロキシ安息香酸、パラヒドロキシ安息香酸エステル、塩酸アルキルジアミノエチルグリシン液等が挙げられる。
水溶性フッ化物としては、フッ化ナトリウム、モノフルオロリン酸ナトリウム等が挙げられる。
その他の有効成分としては、クロロフィル化合物、ε−アミノカプロン酸、トラネキサム酸、ビタミンC、ビタミンE、ニコチン酸エステル、アラントインクロルヒドロキシアルミニウム、アズレン、塩化リゾチーム、ヒノキチオール、β−グリチルレチン酸、グリチルリチン酸塩類、プロテアーゼ、生薬抽出物等が挙げられる。

本発明の口腔用組成物を、練歯磨剤や液状歯磨剤等のペースト状の歯磨剤組成物とする場合には、基本的には、少なくとも水不溶性研磨剤を10〜75重量%、粘結剤を0.5〜5重量%、湿潤剤及び水を10〜85重量%含有させるのが好ましい。ここで、上記水不溶性研磨剤の含有量は、練歯磨剤の場合には20〜75重量%、液状歯磨剤の場合には10〜30重量%が好ましい。また、粉歯磨剤のような固形組成物とする場合には、上記の歯磨剤用の添加剤の成分のうち任意の固形成分を配合できるが、基本的には、水不溶性研磨剤を60〜99重量%配合するのが好ましい。また、香料及び甘味料は合計で0.01〜5重量%配合するのが好ましい。

また、本発明の口腔用組成物を歯肉塗布剤とする場合には、油性成分又は水溶性高分子の他、湿潤剤、界面活性剤、香料、甘味料、殺菌剤、防腐剤、色素、水、水溶性フッ化物、シリコーン、その他の有効成分等が適宜用いられる。
上記油性成分としては、ワセリン、ラノリン、流動パラフィン、天然油脂、ミリスチン酸イソプロピル、ミリスチン酸オクチルドデシル等の高級脂肪酸エステル類、パルミチン酸、イソステアリン酸等の高級脂肪酸類、ミリスチルアルコール、セチルアルコール等の高級アルコール類、スクワラン、シリコーンオイル等が挙げられる。
また上記水溶性高分子としては、前述の粘結剤として例示したものが使用できるが、これらのうちでもヒドロキシエチルセルロースが特に好ましい。また、ポリオキシエチレンポリオキシプロピレンブロック共重合体のような水溶性高分子も好適に使用できる。上記歯肉塗布剤には、上記の油性成分又は水溶性高分子を該塗布剤中に10〜50重量%、湿潤剤及び水を20〜70重量%含有させるのが好ましい。

【発明の効果】
本発明の口腔用組成物は、苦み成分を含みながらも使用感が改良されていて、また、硬さ、色、外観が経時的に良好に保持されている。

以下、実施例により、本発明を詳細に説明するが、本発明は下記の実施例に制限されるものではない。なお、下記の例に示す%は質量%を意味する。
表1の組成(単位:質量%)に従って常法に準じて練歯磨剤を調製した。これらの練歯磨剤を以下の評価に供した。結果を表1に併せて示す。

調製した実施例及び比較例の練歯磨剤について、15名にて官能評価を行った。評価方法は歯磨き中及び歯磨き後について行った。
「評価基準」
1)歯磨き中:歯みがきを口に含み、磨いている間の使用感評価

3点・・・・・ほとんど苦みを感じない。
2点・・・・・僅かに苦みを感じる。

2)歯磨き後:磨き終わって、口をすすいだ後の評価

<安定性評価>
調製した実施例及び比較例の練歯磨剤について、ラミネートチューブに充填して、40℃の恒温器に保存した。保存期間1ヶ月〜6ヶ月について、定期的に性状(外観、例えば水の分離の有無、硬さ、色)を官能評価し、最終の安定性評価を6ヶ月保存品にて確認した。
「評価基準」
○・・・・・全く変化なし。
△・・・・・硬さが僅かにゆるくなり、色が僅かに褐変している。

以上の実験結果から、本発明の口腔用組成物は、苦み成分による悪い使用感が改善されていることが判った。また、本発明の口腔用組成物は硬さ、色、外観といった性状が経時的に良好に保持されている。
さらに、常法に従って調製した下記実施例6〜9の練歯磨剤について上記の実験を行ったところ、上記と同様な結果が得られた。
