Description
また特許文献3は、培養原料細胞と除去対象細胞を含む細胞含有液を多孔質体からなる細胞捕捉材が充填されている容器に導入し、細胞捕捉材に培養原料細胞を捕捉させ、除去対象細胞を容器外に導出した後に容器ごと培養することにより、安価でかつ簡便・短時間操作が可能な細胞分離・培養装置を提案している。しかし、該分離・培養方法はもともとは単球を分離し、樹状細胞へ分化させることを目的としていることから、該装置では、培養原料細胞を細胞捕捉材に捕捉させた状態で容器ごと培養しており、これを成体幹細胞の分離・培養に応用した場合、細胞が増殖できるスペースの関係から、得られる細胞数がある程度制限されるものと考えられる。
・ 少なくとも、
成体幹細胞を含む被処理液から成体幹細胞を選択的に捕捉する手段1と、
手段1の入口側に接続され、成体幹細胞を含む被処理液を手段1に導入する回路2と、
手段1の出口側に接続され、手段1から導出された液体を排液する回路3と、
手段1に回収液を導入する手段4と、
手段1から回収された細胞を培養する手段5と、
から構成される成体幹細胞分離・培養システム。
(2) 手段1が、被処理液導入部と被処理液導出部を有する容器に、被処理液中の白血球及び赤血球を実質的に通過させることが可能である細胞分離材を収納してなる細胞分離器であることを特徴とする(1)の成体幹細胞分離・培養システム。
(3) 細胞分離材が、被処理液中の白血球の60%以上を通過させ、かつ、被処理液中の赤血球の90%以上を通過させることが可能である細胞分離材であることを特徴とする(2)の成体幹細胞分離・培養システム。
(4) 手段4において使用される回収液が、成体幹細胞の培養液であることを特徴とする(1)の成体幹細胞分離・培養システム。
(5) 手段5として、細胞培養バッグまたは細胞培養カセットを用いることを特徴とする(1)の成体幹細胞分離・培養システム。
(6) 手段5として、自動培養装置を用いることを特徴とする(1)の成体幹細胞分離・培養システム。
(7) (1)〜(6)いずれかの成体幹細胞分離・培養システムを用いて、成体幹細胞を含む被処理液から成体幹細胞を分離し増幅する方法。
(8) (7)の方法を用いて調製された細胞組成物。
1)成体幹細胞を含む被処理液から成体幹細胞を選択的に捕捉する手段1と、
2)手段1の入口側に接続され、成体幹細胞を含む被処理液を手段1に導入する回路2と、
3)手段1の出口側に接続され、手段1から導出された液体を排液する回路3と、
4)手段1に回収液を導入する手段4と、
5)手段1から回収された細胞を培養する手段5と、
から構成される成体幹細胞分離・培養システムである。
特に好ましくは耐滅菌性を有する素材であるが、具体的には、ポリプロピレン、ポリ塩化ビニル、ポリエチレン、ポリイミド、ポリカーボネート、ポリスルホン、ポリメチルペンテン等が挙げられる。
図1は本発明の実施態様の一例を示すものであるが、本発明はこれに限定されるものではない。
成体幹細胞を含む被処理液を細胞分離器1−1に通液することにより、成体幹細胞が細胞分離材に捕捉されるが、同時に一部の赤血球や有核細胞が細胞分離材中に留まっている可能性がある。これらの不要な細胞を除去するために、洗浄液を細胞分離器1−1に通液してもよい。この場合の洗浄液流入口は、細胞分離器1−1に通液した被処理液流入口より上流にあることが好ましい。ここで用いられる洗浄液は、成体幹細胞に対して負の影響を与えない液体であれば特に限定されないが、具体例としては、生理食塩水、リンゲル液、血清、血漿、細胞培養に用いられる培地、リン酸緩衝液等の一般的な緩衝液等が挙げられる。成体幹細胞への負の影響が小さいこと、医療用途に用いられている実績があることから、生理食塩水を用いることが好ましい。洗浄液は、成体幹細胞を含む被処理液と同様、回路2を用いて細胞分離器1−1に通液されてもよいし、回路2とは別の回路を設けて細胞分離器1−1に通液されてもよい。いずれの場合にも、洗浄液の通液条件は、被処理液を細胞分離器1−1に通液した場合に準じるものを用いることが好ましい。
細胞分離材に捕捉された成体幹細胞は、手段4に相当する細胞回収液注入装置4−1を用いて、回収液を、ライン4−2を介して細胞分離器1−1に通液することによって、ライン5−2を介して細胞培養器5−1に移送されて回収される。ライン4−2は三方活栓を介して細胞分離器1−1の上流側に位置するライン2−2に連結されてもよいし、下流側に位置するライン3−2に、三方活栓4−3を介して連結されてもよいが、細胞分離器1−1中において成体幹細胞は上流側に偏在する可能性が高いため、下流側に位置するライン3−2に連結されていることが好ましい。
回収された成体幹細胞は、細胞培養器5−1を用いて培養され、必要な細胞数まで増幅されるが、この時、成体幹細胞としての性質を保ったまま増幅されてもよいし、適当な特定の細胞や組織に分化誘導されてもよい。成体幹細胞としての性質を保ったまま増幅された細胞組成物も、特定の細胞や組織に分化誘導された細胞組成物も本発明の範疇に含まれる。また、細胞培養器5−1中で、必要な細胞数まで増幅された成体幹細胞は、その後更に続けて細胞培養器5−1外で培養を続けてもよいし、細胞培養器5−1外で分化誘導を行うこともできる。
本発明の成体幹細胞分離・培養システムに相当する成体幹細胞分離・培養装置を作製し、骨髄液から成体幹細胞の一種である間葉系幹細胞を分離し、増幅させた例を以下に示す。本実施例で用いた装置は、図1に示されるもので、液槽2−1に成体幹細胞を含む被処理液が用意され、細胞分離器1−1とライン2−1の間に細胞培養器5−1が、また細胞分離器1−1の下流に不要細胞回収容器3−1が、細胞分離器1−1と不要細胞回収容器3−1の間に細胞回収液注入装置4−1があるシステムである。細胞分離器1−1としては、出入口を供えた内径1cmの円筒状の筒に、レーヨンとポリプロピレンからなる分割繊維不織布(密度=1.3×105g/m3、繊維径=3〜12μm、目開き=5〜50μm(短径〜長径)、厚み5.5×10−5m)を12枚積層し、不織布の上下を外形1cm、内径7mm、高さ5mmのストッパーにて挟み込むことにより、不織布を固定化した細胞分離器を用いた。
培地に軟骨分化誘導因子(上記)を添加しない以外は実施例1と同様の方法にて、軟骨基質形成能を評価した。その結果、軟骨基質が紫色に染まる異染色性(メタクロマジー)は観察されなかった(図2)。
実施例1と同様の方法で採取した骨髄液2mlを、リン酸緩衝液PBS(−)2mlと混合(2倍希釈)した。次に15ml遠沈管にFicoll paque plus(アマシャム)を3ml添加し、該溶液の上層に先に調整した希釈骨髄液4mlを積層した。1400rpm、30min室温にて遠心分離することにより、成体幹細胞を含む単核球画分層を回収した。PBS(−)を約10ml入れ、1300rpm、5minで細胞の洗浄を行った。次に同様にPBS(−)を約10ml入れ、1200rpm、5minで細胞の再洗浄を行った。再洗浄液を2mlのPBS(−)に懸濁し、血球数の測定を行い、細胞回収率を求めた。また実施例1と同様の方法でコロニー出現率、異染色性の観察を行った。その結果、赤血球の回収率は1%、血小板の回収率は8%、白血球の回収率は83%であった。また出現したコロニー数は、79個/カセットであった。

Claims (7)
- 少なくとも、
成体幹細胞を含む被処理液から成体幹細胞を選択的に捕捉する手段1と、
手段1の入口側に接続され、成体幹細胞を含む被処理液を手段1に導入する回路2と、
手段1の出口側に接続され、手段1から導出された液体を排液する回路3と、
手段1の出口側に配置され、手段1に回収液を導入する手段4と、
手段1の入口側に配置され、手段1から回収された細胞を培養する手段5と、
から構成される成体幹細胞分離・培養システム。 - 手段1が、被処理液導入部と被処理液導出部を有する容器に、被処理液中の白血球及び赤血球を実質的に通過させることが可能である細胞分離材を収納してなる細胞分離器であることを特徴とする請求項1記載の成体幹細胞分離・培養システム。
- 細胞分離材が、被処理液中の白血球の60%以上を通過させ、かつ、被処理液中の赤血球の90%以上を通過させることが可能である細胞分離材であることを特徴とする請求項2記載の成体幹細胞分離・培養システム。
- 手段4において使用される回収液が、成体幹細胞の培養液であることを特徴とする請求項1記載の成体幹細胞分離・培養システム。
- 手段5として、細胞培養バッグまたは細胞培養カセットを用いることを特徴とする請求項1記載の成体幹細胞分離・培養システム。
- 手段5として、自動培養装置を用いることを特徴とする請求項1記載の成体幹細胞分離・培養システム。
- 請求項1〜6いずれか1項に記載の成体幹細胞分離・培養システムを用いて、成体幹細胞を含む被処理液から成体幹細胞を分離し増幅する方法。
