Description
DBU:1,8−ジアゾビシクロ[5.4.0]ウンデカ−7−エン
eq.:当量
TFA:トリフルオロ酢酸
THF:テトラヒドロフラン
分取キラルクロマトグラフィーによる、(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリルの鏡像異性体の分割
式(I)の化合物480mgを、メタノール5mlに溶解し、Chiracel OJ相300gを25cmに充填したProchrom 50cmx50mmのカラムに80ml/分の流速で注入し、ヘプタン/イソプロパノール(70/30)の混合物中にその流速で溶出させた。式(Ia)の鏡像異性体((S)の立体配置)を、45.6%の収率および97.6%の鏡像異性体純度で得た。また、式(Ib)の鏡像異性体((R)の立体配置)を、42.2%の収率および99.3%の鏡像異性体純度で得た。
NaBH4の存在下での(S)−(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリルの還元による、[(7S)−3,4−ジメトキシビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル]メタンアミン
窒素掃気下に置き、冷却器、磁気撹拌子、窒素吸気口にCaCl2ガード、および温度計を備えた125ml入り三ツ口フラスコに、NaBH4(3eq.)およびTHF(10ml/g)を導入した。TFA(2.97eq.)中に20〜25℃で滴加した。THF(4ml/g)中の(S)−(3,4−ジメトキシビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリル(1eq.)の溶液を滴加した。20〜25℃で一晩撹拌し、次いで、反応混合物を0.3MHCl水溶液(0.5eq.)に注入し、20〜25℃で1時間撹拌した。減圧下のフリット越しにろ過し、THFで濯いで、溶媒を減圧下で留去した。未精製の反応生成物をジクロロメタン(20ml/g)に溶解し、10ml/gの水および水酸化ナトリウム溶液(2ml/g)を加えた。15分間撹拌し、次いで分離するのを許し、有機相を水洗し、MgSO4上で乾燥し、溶媒を減圧下で留去して、標記生成物を78.8%の収率および94.4%の鏡像異性体純度で得た。
炭素担持パラジウムの存在下での(S)−(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリルの還元による、[(7S)−3,4−ジメトキシビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル]メタンアミン
(S)−(3,4−ジメトキシビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリル1eq.、1.12NHClメタノール溶液1eq.、および0.1重量%の5%炭素担持パラジウムを、125mlのオートクレーブに導入した。メタノール(10ml/g)で濯いだ。窒素、次いで水素で掃気し、20℃で撹拌し、30バールの下、その温度で5時間水素化した。オートクレーブの圧力を解き、反応混合物をろ過し、溶媒を減圧下で留去した。得られた塩酸塩をジクロロメタン(20ml/g)に取って、10ml/gの水および水酸化ナトリウム溶液(2ml/g)を加えた。15分間撹拌し、次いで分離するのを許し、有機相を水洗し、MgSO4上で乾燥し、溶媒を減圧下で留去して、標記生成物を90%の収率および95.5%の鏡像異性体純度で得た。
(R)−(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリルのラセミ化による、ラセミ化合物の(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリル
(R)−(3,4−ジメトキシ−ビシクロ[4.2.0]オクタ−1,3,5−トリエン−7−イル)ニトリル100mg(0.53mmol)、イソプロパノール5mlおよびDBU121mg(1.5eq.)を、冷却器および磁気撹拌装置を備えたフラスコに導入した。65℃で2時間加熱し、次いで環境温度に戻るのを許した。ろ過して、標記化合物を得た。
