Description
SEQ ID No.1:αS1−カゼインの91〜95番目に相当するペンタペプチド。
完全体のカゼインは、公知の方法に従った酸による沈殿及びアルカリによる中和によって、ミルクから得られる。
高架式十字迷路試験
(原理)
高架式十字迷路試験は、新規の嫌悪状況における探索行動の量を測定することを可能とする(PELLOW等、1985(非特許文献16))。この実験は、壁が無い領域に対する不安と新しい環境を探索する欲望との葛藤を利用する。装置の中央に置かれたラットは、その環境を探索することを強いられる。壁が無いアームは、不安が生じる環境を構成し、一方、壁があるアームでは不安が生じない。全てのアームに入った回数、壁が無いアームに入った回数、壁があるアームに入った回数、壁が無いアームに居た時間、及び最初に壁が無いアームに入るまでの時間が、動物の不安の程度を反映するパラメータとなる。
木製の高架式十字迷路は、互いに90°の角度を有する4つの枝部からなる。2つの壁が無い枝部(45cm×10cm)及び2つの壁がある枝部(45cm×10cm、高さが40cmの木製の板により囲まれている)は、互いに対向して配置されている。中央の四角形の寸法は、10cm×10cmである。壁が無い枝部は、照度が500lxの白色光により照らされている。高架式十字迷路は、地面から50cmの高さに位置する。
観察は、24時間周期のリズムの影響を最小限にするために、朝の9時と11時との間に行う。高架式十字迷路に通す前に、動物の探索行動を刺激するために、動物を壁が無い領域に10分間置く。その後、高架式十字迷路の中央に置き、5分間観察する。十字迷路は、臭いを除去するために、2度の試験の間に95%エタノール(v/v)により洗浄される。
本実験では、45匹のラットを3つの群に分ける。
ウィスターラットの腹腔内に、1%(v/v)グリセロール及び0.2%(v/v)メチルセルロースを含む溶液を2mL/kg(n=15)、ジアゼパムを1mg/kg(n=15)又はペンタペプチドであるCNαS1ーf(91−95)を0.5mg/kg(n=15)で注入した。これらによって、(i)異なるアームに入った全回数、(ii)壁が無いアームに入った回数、及び(iii)壁が無いアームに居た時間に対して生じた効果を、ノンパラメトリック法による分散分析(Kruskall-Wallis)により解析し、それぞれの平均の比較をマン−ホイットニー検定により行った(それぞれの処理の群の平均であるAとBとで有意差がある、p<0.05、図1)。全てのアームに入る回数により反映される動物の探索活性は、それぞれの処理によって有意差は見られない。賦形剤により処理された動物が壁が無いアームに入る回数及び壁が無いアームに居る時間は、ジアゼパムを処理した動物及びαS1−カゼインの91〜95番目の断片を処理した動物の場合よりも有意に少ない。ジアゼパムとαS1−カゼインの91〜95番目の断片との間には、有意差は認められない。ウィスターラットの高架式十字迷路試験において、αS1−カゼインの91〜95番目の断片は、ジアゼパムと類似の作用を有する。すなわち、不安を生じさせる領域への探索が増大する。
(原理)
明暗試験は、齧歯類が非常に明るい環境を先天的に嫌う性質と、新しい環境及び光等の弱いストレス因子に応じた自発的な探索行動とに基づいている。明暗箱装置は、齧歯類の探索行動を、非嫌悪部屋(暗所)において慣らした後に、不慣れな嫌悪部屋(白色光により非常に明るい)に関して実験できる(BOURIN及びHASCOET、2003(非特許文献17))。古典的な抗不安薬は、この装置を用いることにより認められ得る。
明暗箱は、寸法が65cm×49cm×35cm(h×L×l)であり、3つの8×8cmの扉を有する板により2つの同一の部屋に分離されている。部屋の底面はおがくずに覆われている。各ラットはその箱の後側の部屋に24時間置かれ、前側の部屋に通じる扉は、近づき難く形成されている。従って、後側の部屋は慣れた部屋となる。動物は餌及び水を適宜与えられる。試験の日に、処理した後のラットは、慣れた部屋に再び置かれ、前側の部屋(不慣れな部屋)に通じる扉は、動物が環境を自由に探索できるように、近づきやすく形成される。不慣れな部屋は、照度が1500lxである白色光で照らすことにより嫌悪させる。動物を10分間観察し、各部屋に居る時間を測定する。
本実験では、36匹のラットを3つの群に分ける。
ウィスターラットの腹腔内に、1%(v/v)グリセロール及び0.2%(v/v)メチルセルロースを含む溶液を2mL/kg(n=12)、ジアゼパムを1mg/kg(n=12)又はペンタペプチドであるCNαS1−f(91−95)を0.5mg/kg(n=12)で注入した。これらによって、(i)不慣れな部屋に入った回数、(ii)慣れた部屋に居た時間、及び(iii)不慣れな部屋に居た時間に対して生じた効果を、ノンパラメトリック法による分散分析(Kruskall-Wallis)により解析し、それぞれの平均の比較をマン−ホイットニー検定により行った(それぞれの処理の群の平均であるAとBとで有意差がある、p<0.05、図2)。賦形剤で処理された動物が嫌悪する不慣れな部屋に居た時間は、ジアゼパムを処理した動物及びαS1−カゼインの91〜95番目の断片を処理した動物の場合よりも有意に少ない。さらに、賦形剤で処理された動物が慣れた部屋に居た時間は、有意に増大している。ジアゼパムとαS1−カゼインの91〜95番目の断片との間には、有意差は認められない。ウィスターラットの明暗箱試験において、αS1−カゼインの91〜95番目の断片は、ジアゼパムと類似の抗不安作用を示す。
Claims (12)
- SEQ ID No.1に示すペプチド及び/又はSEQ ID No.2に示すペプチドの有効量を活性成分として含むことを特徴とする不安症、睡眠障害及びてんかんを治療するための組成物。
- SEQ ID No.1に示すペプチド及び/又はSEQ ID No.2に示すペプチドの有効量を含むαS1−カゼインの加水分解産物又は加水分解産物の断片を含む、不安症、睡眠障害及びてんかんを治療するための組成物。
- 適当な医薬品賦形剤をさらに含むことを特徴とする請求項1又は2に記載の組成物。
- 食品であることを特徴とする請求項1又は2に記載の組成物。
- 不安症、睡眠障害及びてんかんの治療に用いる薬剤の製造を目的とするSEQ ID No.1に
示すペプチド及び/又はSEQ ID No.2に示すペプチドの使用。 - 不安症、睡眠障害及びてんかんの治療に用いる薬剤の製造を目的とするSEQ ID No.1に
示すペプチド及び/又はSEQ ID No.2に示すペプチドの有効量を含む、カゼイン若しくは
αS1−カゼインの加水分解産物及び/又はカゼイン若しくはαS1−カゼインの加水分解産物の断片の使用。 - SEQ ID No.1に示すペプチド及び/又はSEQ ID No.2に示すペプチドの少なくとも1つの有効量を含むことを特徴とする、不安症、睡眠障害及び/又はてんかんに罹りやすい人々のための食品。
- SEQ ID No.1に示すペプチド及び/又はSEQ ID No.2に示すペプチドの有効量を含むカゼイン又はαS1−カゼインの加水分解産物又は加水分解産物の断片を含む、不安症、睡眠障害及び/又はてんかんに罹りやすい人々のための食品。
- SEQ ID No.1に示すペプチド又はSEQ ID No.2に示すペプチドをコードしていることを特徴とする単離されたポリヌクレオチド。
- 請求項9に記載のポリヌクレオチドを含むことを特徴とする発現ベクター。
- 請求項9に記載のポリヌクレオチド及び/又は請求項10に記載の発現ベクターを用いて形質転換されたことを特徴とするヒトを除く宿主生物。
- SEQ ID No.1に示すペプチド又はSEQ ID No.2に示すペプチドを発現することを特徴とするヒトを除く形質転換された宿主生物。
