Description

本発明は、金属サレン錯体化合物に係り、自己磁性を有すると共に、軟膏組成物に適用可能な金属サレン錯体化合物、この金属サレン錯体化合物を有する局所麻酔薬剤、及びこの金属サレン錯体化合物を有する抗悪性腫瘍薬剤に関する。
一般に薬剤は生体内に投与され患部に到達し、その患部局所において薬理効果を発揮することで治療効果を引き起こすが、薬剤が患部以外の組織（つまり正常組織）に到達しても治療にはならない。したがって、いかにして効率的に患部に薬剤を誘導するかが重要である。
薬剤を患部に誘導する技術は、ドラッグ・デリバリと呼ばれ、近年研究開発が盛んに行われている分野である。このドラッグ・デリバリには少なくとも二つの利点がある。一つは、患部組織において十分に高い薬剤濃度が得られることである。薬理効果は、患部における薬剤濃度が一定以上でないと生じないため、低い薬剤濃度では十分な治療効果が期待できない。二つめは、薬剤を患部組織のみに誘導して、正常組織への副作用を抑制することができることである。
このようなドラッグ・デリバリが最も効果を発揮するのが、抗がん剤によるがん治療である。抗がん剤は、細胞分裂の活発ながん細胞の細胞増殖を抑制するものが大半であるため、正常組織においても細胞分裂の活発な組織、例えば骨髄あるいは毛根、消化管粘膜等の細胞増殖を抑制する。このため抗がん剤の投与を受けたがん患者には貧血、抜け毛、嘔吐等の副作用が発生する。これら副作用は患者にとって大きな負担となるため、投薬量を制限しなければならず、抗がん剤の薬理効果を十分に得ることが出来ないという問題がある。
このような抗がん剤（抗悪性腫瘍薬）中で、アルキル系抗悪性腫瘍薬は、核酸蛋白等にアルキル基（−ＣＨ２−ＣＨ２−）を結合させる能力を持つ抗がん剤の総称であり、ＤＮＡをアルキル化してＤＮＡ複製を阻害し、細胞死をもたらす作用がある。この作用は、細胞周期に無関係に働きＧ０期の細胞にもおよび、増殖が盛んな細胞に対する作用が強く、骨髄、消化管粘膜、生殖細胞、毛根等に障害を与えやすい。
また、代謝拮抗系抗悪性腫瘍薬は、核酸や蛋白合成過程の代謝物と類似の構造をもつ化合物であり、核酸合成を阻害する等して細胞を障害し、分裂期の細胞に特異的に作用する。
抗腫瘍性抗生物質は、微生物によって産生される化学物質であり、ＤＮＡ合成抑制、ＤＮＡ鎖切断等の作用をもち抗腫瘍活性を示す。
また、微小管阻害薬は、細胞分裂の際に紡錘体を形成したり、細胞内小器官の配置や物質輸送等、細胞の正常機能の維持に重要な役割を果たしている微小管に直接作用することで抗腫瘍効果を示す。微小管阻害剤は細胞分裂が盛んな細胞や神経細胞等に作用を及ぼす。
白金製剤は、ＤＮＡ鎖又は鎖間結合あるいはＤＮＡ蛋白結合を作ってＤＮＡ合成を阻害する。シスプラチンが代表的薬剤であるが腎障害が強く、多量の補液が必要とされる。
また、ホルモン類似薬系抗悪性腫瘍薬は、ホルモン依存性の腫瘍に対して有効である。男性ホルモン依存性の前立腺がんに対して女性ホルモンを投与したり抗男性ホルモン剤を投与したりする。
分子標的薬は、それぞれの悪性腫瘍に特異的な分子生物学的特徴に対応する分子を標的とした治療法である。
また、トポイソメラーゼ阻害薬は、ＤＮＡに一時的に切れ目を入れてＤＮＡ鎖のからまり数を変える酵素である。トポイソメラーゼＩは、環状ＤＮＡの一方の鎖に切れ目を入れ、もう一方の鎖を通過させた後、切れ目を閉じる酵素であり、トポイソメラーゼ阻害薬ＩＩは環状ＤＮＡの２本鎖両方を一時的に切断し、その間を別の２本鎖ＤＮＡを通過させ、再び切れ目をつなぎ直す酵素である。
さらにまた、非特異的免疫賦活薬は、免疫系を活性することによってがん細胞の増殖を抑制する。
また、ドラッグ・デリバリの利点は、局所麻酔剤でも存在する。局所麻酔剤は、痔疾患、口内炎、歯周病、虫歯、抜歯、あるいは手術等による粘膜や皮膚等の居所的なかゆみや疼痛を処理ずるために用いられている。代表的な居所麻酔剤としては、リドカイン（商品名：キシロカイン）が知られているが、このリドカインは即効性に優れているものの、抗不整脈作用を有する。また、脊椎麻酔を行う際、脊髄液の中に麻酔薬であるリドカインを注入すると脊髄液中を拡散し、最悪の場合は頸部の脊髄に達することで呼吸機能が停止し重篤副作用をもたらす懸念がある。
ドラッグ・デリバリの具体的な手法としては、例えば、担体（キャリア）を用いたものがある。これは患部に集中しやすい担体に薬剤を載せて、薬剤を患部まで運ばせようというものである。担体として有力視されているのが磁性体であり、薬剤に磁性体である担体を付着させ、磁場によって患部に集積される方法が提案されている（例えば、特許文献１参照）。
しかしながら、磁性体を担体（キャリア）として使用する場合、経口投与が困難なこと、担体分子が一般に巨大であること、担体と薬剤分子との間の結合強度、親和性に技術的な問題がある等の理由から、実用化が困難であった。
そこで、有機化合物の基本骨格に対して、正又は負のスピン電荷密度を付与する側鎖が結合され、全体として外部磁場に対して磁気共有誘導される範囲の適性を持ち、人体や動物に適用された際に、体外からの磁場によって局所的に磁場が与えられている領域で保持され、元来保有している医薬効果を前記領域において発揮するようにした局所治療薬が紹介されている。この技術では、薬剤として、鉄サレン錯体が挙げられている。（例えば、特許文献２参照）。

しかしながら、例えば、舌がんや、後眼部組織等の治療のように、比較的患者への負担がかかる治療では、より優れた非侵襲性を有する薬物を疾患部位へ効率よく移行できる薬剤やドラッグ・デリバリ・システム等の開発が望まれている。
本発明は、このような事情に鑑みなされたものであり、優れた非侵襲性を有し、疾患部位へ効率よく移行できる金属サレン錯体化合物、この金属サレン錯体化合物を有する局所麻酔薬剤、及びこの金属サレン錯体化合物を有する抗悪性腫瘍薬剤を提供することを目的とする。
この目的を達成するために、本発明は、金属サレン錯体又は当該金属サレン錯体の誘導体の複数分子がそれぞれの金属原子の部分で水を介して多量体化されており、基剤に混合されて軟膏となる金属サレン錯体化合物を提供するものである。
また、本発明に係る金属サレン錯体化合物は、前記複数分子が２分子であり、当該２分子がそれぞれの金属原子の部分で水を介して２量体化されていることがさらに望ましい。
この金属サレン錯体化合物は、基剤に混合されて軟膏とすることができるため、軟膏として患部に投与することができる。したがって、優れた非侵襲性を有し、疾患部位へ効率よく移行させることができる。
なお、本発明でいう「軟膏」とは、例えば、油脂性基剤を用いたものの他、日本薬局方で規定されているように、乳剤性基剤を用いたクリーム剤等も含まれるものである。軟膏を作成するための「基剤」は、皮膚に付着し、有効成分を長く皮膚にとどめる働きをするものであり、塗りやすく、皮膚に対する刺激性が無く、有効成分の安定性に影響しないものが求められる。
また、本発明に係る金属サレン錯体化合物は、口腔内に使用可能な基剤に混合することで、例えば、舌や歯茎、ほほの内側等に適用することもできる。
このような基剤としては、疎水性基剤（油脂性基剤）、親水性基剤（乳剤性基剤、水溶性基剤、懸濁性基剤）、特殊な剤型（糊膏（リニメント）、泥膏（パスタ）、硬膏、ローション・スプレー等）、口腔用軟膏剤、眼軟膏剤等が挙げられる。より具体的には、例えば、ワセリン（黄色ワセリン、親水ワセリン、白色ワセリン）、ケナログ、流動パラフィン、ポリオキシエチレン硬化ヒマシ油、マクロゴール、ゲル化炭化水素、等の軟膏基剤等が挙げられる。
本発明の好適な形態は、下記式（Ｉ）、(II)、（III）で示される自己磁性金属サレン錯体及びその誘導体である。

（実施例１）
本発明の金属サレン錯体化合物の製造を次のように行った。
Step 1:
４−ニトロフェノール（4-nitrophenol；化合物１（compound 1））；２５ｇ、０．１８ｍｏｌ、ヘキサメチレンテトラミン（hexamethylene tetramine）；２５ｇ、０．１８ｍｏｌ、ポリリン酸（polyphosphoric acid）；２００ｍｌの混合物を１００℃で１時間攪拌した。その後、その混合物を５００ｍｌの酢酸エチルと１Ｌ（リットル）の水の中に入れ、完全に溶解するまで攪拌した。さらにその溶液に４００ｍｌの酢酸エチルを追加で加えたところ、その溶液は２つの相に分離した。次に、この２つの相に分離した溶液から水の相を取り除き、残りの化合物を塩性溶剤で２回洗浄し、無水ＭｇＳＯ４（硫酸マグネシウム）で乾燥させた結果、化合物２（compound 2）が１７ｇ（収率５７％）合成できた。
Step 2:
化合物２（compound 2）；１７ｇ、０．１０ｍｏｌ、無水酢酸（acetic anhydride）；２００ｍｌ、Ｈ２ＳＯ４；少々、を室温で１時間攪拌させた。得られた溶液は、氷水；２Ｌの中に０．５時間混ぜ、加水分解を行った。得られた溶液をフィルターにかけ、大気中で乾燥させたところ白い粉末状のものが得られた。酢酸エチルを含む溶液を使ってその粉末を再結晶化させたところ、化合物３（compound 3）の白い結晶を２４ｇ（収率７６％）得ることができた。
Step 3:
化合物３（compound 3）；２４ｇ、７７ｍｏｌと、メタノール；５００ｍｌに１０％のパラジウムを担持したカーボン；２．４ｇの混合物を一晩、１．５気圧の水素還元雰囲気で還元した。終了後、フィルターでろ過したところ茶色油状の化合物４（compound 4）が２１ｇ合成できた。
Step 4, 5:
無水ジクロメタン(DCM)；２００ｍｌに化合物４（compound 4）；２１ｇ、７５ｍｍｏｌ、二炭酸ジ-tert-ブチル（di(tert-butyl) dicarbonate）；１８ｇ、８２ｍｍｏｌを窒素雰囲気で一晩攪拌した。得られた溶液（化合物５（compound 5））を真空中で蒸発させた後、メタノール；１００ｍｌで溶解させた。その後、水酸化ナトリウム；１５ｇ、３７４ｍｍｏｌと水５０ｍｌを加え、５時間還流させた。その後冷却し、フィルターでろ過し、水で洗浄後、真空中て乾燥させたところ茶色化合物がえられた。得られた化合物は、シリカジェルを使ったフラッシュクロマトグラフィーを２回行うことで、１０ｇ（収率５８％）の化合物６（compound 6）が得られた。
Step 6:
無水エタノール；４００ｍｌの中に化合物６（compound 6）；１０ｇ、４２ｍｍｏｌを入れ、加熱しながら還流させ、無水エタノール；２０ｍｌにエチレンジアミン；１．３ｇ、２１ｍｍｏｌを０．５時間攪拌しながら数滴加えた。そして、その混合溶液を氷の容器に入れて冷却し１５分間かき混ぜた。その後、エタノール；２００ｍｌで洗浄し、フィルターをかけ、真空で乾燥させたところ化合物７（compound 7）が８．５g（収率８２％）で合成できた。
Step 7:
通常のメタノール（昭和化学製メタノール、純度９９．５％以上）；５０ｍｌの中に化合物７（compound 7）；８．２ｇ、１６ｍｍｏｌ、トリエチルアミン（triethylamine）；２２ｍｌ、１６０ｍｍｏｌを入れ、メタノール；１０ｍｌの中に、Ｆｅサレン錯体化合物の場合は、ＦｅＣｌ３・４Ｈ２Ｏ（塩化鉄（III）四水和物）；２．７ｇ、１６ｍｍｏｌを、Ｍｎサレン錯体化合物の場合は、ＭｎＣｌ３・４Ｈ２Ｏ（塩化マンガン（III）四水和物）を、Ｃｒサレン錯体化合物の場合は、ＣｒＣｌ３・４Ｈ２Ｏ（塩化クロム（III）四水和物）；２．７ｇ、１６ｍｍｏｌを、それぞれ加えた溶液を窒素雰囲気下で混合した。室温窒素雰囲気で１時間混合したところ茶色の化合物が得られた。その後、真空中或いはマグネシウムを使う等して十分に水を乾燥或いはマグネシウムに吸着除去させた。得られた化合物をジクロロメタン；４００ｍｌで希釈し、塩性溶液で２回洗浄し、Ｎａ２ＳＯ４で乾燥させ、真空中で乾燥させたところ、水分子を含む２量体の金属サレン錯体化合物が得られた。得られた化合物は、ジエチルエーテルとパラフィンの溶液中で再結晶させ高速液化クロマトグラフィーで測定したところ純度９５％以上の水分子を含む２量体の金属サレン錯体化合物であった。
得られた水分子を含む２量体は下記の化学構造式の通りである。
なお、金属と酸素との間の結合は、共有結合と金属結合とが融合したものであると考えられる。得られた水分子付２量体の元素分析をしたところ、Ｃ；５７．７３％、Ｈ；４．４２％、Ｆｅ；１７．２％、Ｎ；８．４９％、Ｏ；１２．１６％となり、計算値と実験値の違いはすべて絶対誤差の±０．４％以内に入っていた。
次に、水分子混入の存在を明らかにするためＴＧ−Ｍａｓｓ分析を行った。その結果、室温から２６０℃までの範囲で、水分子が検出されることが分かった。これは、水分子が結晶に取り込まれているからである。ＴＧ−Ｍａｓｓ分析の結果を図１及び図２に示す。
なお、ＴＧ−Ｍａｓｓ分析の実験条件は次の通りである。

Ｍｎサレン錯体化合物の３７℃（３１０Ｋ）における磁場−磁化曲線を、カンタムデザイン社のＭＰＭＳ７を用いて測定したところ常磁性であった。その結果を、図３に示す。

（実施例５）
Ｆｅサレン錯体化合物の３７℃（３１０Ｋ）における磁場−磁化曲線を図６に示す。図３、図５及び図６から、Ｆｅサレン錯体化合物と比較して、Ｃｏサレン錯体化合物は、磁場が１００００Ｏｅ（エルステッド；（１Ｔ（テスラ）））以上で磁化が多くなることが分かる。また、Ｍｎサレン錯体化合物は、Ｆｅサレン錯体化合物と比較して、磁場が３００００Ｏｅ（３Ｔ）以上で磁化が多くなることが分かる。したがって、Ｆｅサレン錯体化合物は、磁場が１００００Ｏｅ（１Ｔ）以下で最も磁化が大きく、ネオジウム永久磁石等を用いる磁場誘導ドラッグ・デリバリ・システムに適している。また、磁場が１００００Ｏｅ（１Ｔ）を超える場合は、Ｃｏサレン錯体化合物、Ｍｎサレン錯体化合物の磁化が大きく、超伝導磁石を用いる磁場誘導ドラッグ・デリバリ・システムに最適となる。
（実施例６）
ラットＬ６細胞が３０％のコンフルエントの状態の時に、既述の方法によって得られたＦｅサレン錯体化合物、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについて、金属サレン錯体化合物の粉末を磁石に引き寄せられるのが目視できる程度の量を培地に散布して４８時間後に培地の状態を写真撮影した。なお、図７は、ラットＬ６細胞の培地がある角型フラスコに棒磁石を接触させた状態を示している。
次いで、４８時間後、角型フラスコ底面の一端から他端までを撮影し、細胞数を算出した。この結果のうち、Ｆｅサレン錯体化合物の結果を図８に示す。なお、図８において、「磁石から近位」とは、角型フラスコ底面における磁石端面の投影面積内を示し、「磁石から遠位」とは、角型フラスコ底面において磁石端面と反対側にある領域を示す。
図８に示すように、磁石から近位では、Ｍｎサレン錯体が引き寄せられてＦｅサレン錯体の濃度が増しＦｅサレン錯体のＤＮＡ抑制作用によって細胞数が遠位よりも極端に低いことが分かる。また、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても、細胞数は、磁石から近位が遠位よりも極端に低い結果が得られた。この結果、本発明による、磁性を持った薬剤と、磁気発生手段とを備えたシステムによって、個体の目的とする患部や組織に薬剤を集中して存在させることが可能となる。
次に、誘導装置を用いた誘導例について説明する。この誘導装置は、図９に示すように、重力方向に互いに向き合う一対の磁石２３０及び２３２がスタンド２３４とクランプ２３５とによって支持されており、磁石２３０と磁石２３２との間には金属板２３６が配置されている。一対の磁石２３０と磁石２３２との間に金属板２３６、特に鉄板を配置することにより、局所的に一様で強力な磁界を作り出すことができる。この誘導装置は、磁石の代わりに電磁石を用いて発生磁力を可変にすることができる。また、ＸＹＺ方向に一対の磁力発生手段を移動できるようにして、テーブル上の固体の目的とする位置に磁力発生手段を移動させることができる。この磁界の領域に固体の組織を置くことにより、この組織に薬剤を集中させることができる。
より具体的には、例えば、体重約３０グラムのマウスに既述のＦｅサレン錯体化合物（薬剤濃度５ｍｇ／ｍｌ（１５ｍｍｏｌ））を静注して開腹し、右の腎臓を前記一対の磁石２３０及び２３２の間に配置されるようにマウスを金属板２３６の上に置く。なお、使用した磁石は、信越化学工業株式会社製 品番：Ｎ５０（ネオジウム系永久磁石）、残留磁束密度；１．３９〜１．４４Ｔである。このとき、右側の腎臓に与えられた磁場は、約０．３Ｔであり、左側の腎臓に与えられる磁場は、その約１／１０である。
左の腎臓及び磁界を適用しない腎臓（ｃｏｎｔｒｏｌ）と共に、マウスの右腎に磁界を加えて１０分後、ＭＲＩでＳＮＲをＴ１モード及びＴ２モードで測定した。その結果、図１０に示すように、磁界を加えた右腎（ＲＴ）が左腎（ＬＴ）及びコントロールに比較して薬剤を組織内に留め置くことができることが確認された。
また、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても、前記と同様のマウスに静注して開腹し、図９に示す誘導装置により同様に磁場を与えたところ、磁界を加えた右腎（ＲＴ）が左腎（ＬＴ）及びコントロールに比較して薬剤を組織内に留め置くことができることが確認された。
図１１にマウスにおけるメラノーマ成長に対するＦｅサレン錯体化合物の効果を示す。メラノーマは、培養メラノーマ細胞（クローンＭ３メラノーマ細胞）の局所的移植によって、マウス尾腱においてインビボ（in vivo）に形成されたことが分かる。なお、図１１（１）は、Ｆｅサレン錯体化合物の代わりに、塩水を注入した塩水グループ（saline）、図１１（２）は、磁場を適用せずにＦｅサレン錯体化合物を注入したグループ（ＳＣ）、図１１（３）は、磁場を適用しつつ（ｎ＝７〜１０）Ｆｅサレン錯体化合物を注入したグループ（ＳＣ＋Ｍａｇ）の効果を示す写真である。
Ｆｅサレン錯体化合物を尾腱の静脈から静脈投与（５０ｍｇ／ｋｇ）し、市販の棒磁石（６３０ｍＴ、円筒状ネオジウム磁石；長さ１５０ｍｍ、直径２０ｍｍ）を用いて、局所的に磁場を印加した。棒磁石の適用は、Ｆｅサレン錯体化合物を１０〜１４日注入した直後に、メラノーマサイトに３時間穏やかに接触させることにより行った。
棒磁石の適用は、磁場強度が、メラノーマ延長が予想される部位に最大強度となるように、１５０ｍｍ以下のマウス尾腱に対して２週間の成長期間行った。Ｆｅサレン錯体化合物の初回注入の１２日後に、メラノーマ染色された部位を評価することによって、メラノーマの延長を評価した。
図１２に示すように、に示すように、Ｆｅサレン錯体化合物の代わりに、塩水を注入した塩水グループ（ｓａｌｉｎｅ）では、メラノーマ拡張は最大であった（１００±１７．２％）。一方、磁場を適用せずにＦｅサレン錯体化合物を注入したＳＣグループでは、メラノーマ拡張は緩やかに減少した（６３．６８±１６．３％）。これに対して、磁場を適用しつつ（ｎ＝７〜１０）Ｆｅサレン錯体化合物を注入したＳＣ＋Ｍａｇグループでは、ほとんどのメラノーマが消失した（９．０５±３．４２％）。
なお、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても同様の結果が得られた。
図１３に示すように、ヒストロジカル試験を、組織部の腫瘍増殖マーカーであるａｎｔｉ−Ｋｉ−６７抗体及びａｎｔｉ−Ｃｙｃｌｉｎ Ｄ１抗体を用いて、ヘマトキシリン−エオジン染色及び免疫組織染色により行った。その結果、Ｆｅサレン錯体化合物を注入した場合（ＳＣ）においてメラノーマの腫瘍拡張が減少し、さらにＦｅサレン錯体化合物に磁場の適用が組み合わされた場合にはほとんどが消失することが分かった。
なお、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても同様の結果が得られた。
また、薬剤（Ｆｅサレン錯体化合物；９．２５ｍｍｏｌ）に磁場強度；２００Ｏｅ、周波数；５０ｋＨｚ〜２００ｋＨｚの交流磁場を印加したところ、薬剤の温度が２℃〜１０℃上昇した（図１４）。これは、体内投与時の温度に換算すると３９℃〜４７℃に相当し、がん細胞を殺傷することが可能な温度域であることが確認された。なお、図１４（１）は、薬剤に交流磁場をかけたときの、時間に関する温度の変化であり、図１４（２）は、周波数を固定して、磁場のみを変化させたときの最大温度であり、図１４（３）は、磁場を固定して、周波数のみを変化させたときの最大温度である。
なお、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても同様の結果が得られた。
（実施例７）
金属サレン錯体に結合する化合物の電子の移動は第一原理計算で求めることができる。このコンピュータシミュレーションを実現するシステムは、コンピュータとしての公知のハードウエア資源を備えるものであって、メモリとＣＰＵ等の演算回路を備える演算装置と、演算結果を出力する表示手段とを備えている。
メモリは、既存の有機化合物または３次元構造を特定するデータと、コンピュータシミュレーションを実現するソフトウエア・プログラムとを備えている。このソフトウエア・プログラムは、各化合物の側鎖を追加・変更・削除し、所定の側鎖間で架橋し、記述のスピン電荷密度の高い領域を計算し、構造全体としてのスピン電荷密度を決定可能なものである。このプログラムとして、例えば、市販品(Ｄｍｏ１３、アクセルリス社製)を利用することができる。
ユーザは化合物について、側鎖を追加する位置を入力し、または側鎖を変更し、あるいは削除するものを選択し、さらに、架橋を形成すべき箇所をメモリの支援プログラムを利用して演算装置に指定する。演算装置はこの入力値を受けて、スピン電荷密度を演算してその結果を表示画面に出力する。また、ユーザが既存の化合物の構造データをコンピュータシステムに追加することによって、既知の化合物についてのスピン電荷密度を得ることができる。
金属サレン錯体に他の化合物が結合したものの電荷移動は、求めた上向きと下向きのスピン電荷密度を三次元空間で積分すると求めることができる。前述した化学式（Ｉ）のｅ、ｂ、ｋ、ｈ、あるいはｅ、ｈに電荷移動した際の計算結果を次の各表に示す。なお、各表において、マイナス（−）は電子が増加していることを示し、プラス（＋）は電子が減っていることを示す。
（実施例８）
前述したＦｅサレン錯体化合物を、軟膏用の調整剤であるワセリンを基剤として１００ｍｍｏｌと２００ｍｍｏｌの濃度で各々混合した軟膏を作成した。次に、これらの軟膏に対し、２５８Ａ、４００ｋＨｚ、５１．７４ｍＴの条件で交流磁場を印加し、時間と温度上昇との関係を測定した。この結果を図１５に示す。図１５から、濃度が１００ｍｍｏｌ及び２００ｍｍｏｌの軟膏は、測定直後（０秒）の温度は３５℃であり、濃度が２００ｍｍｏｌの軟膏は、３００秒後に約５℃の温度上昇が見られたことが分かる。
次に、前述した式（Ｉ）で示される化合物を、口内炎用の軟膏であるケナログを基剤として１００ｍｍｏｌと２００ｍｍｏｌの濃度で各々混合した軟膏を作成した。次いで、これらの軟膏に対し、２５８Ａ、４００ｋＨｚ、５１．７４ｍＴの条件で交流磁場を印加し、時間と温度上昇との関係を測定した。この結果を図１６に示す。図１６から、濃度が１００ｍｍｏｌ及び２００ｍｍｏｌの軟膏は、測定直後（０秒）の温度は３５℃であり、濃度が２００ｍｍｏｌの軟膏は、３００秒後に約５℃の温度上昇が見られたことが分かる。
次に、比較として、ワセリン２００ｍｇと、ケナログ２００ｍｇの各々に対し、２５８Ａ、４００ｋＨｚ、５１．７４ｍＴの条件で交流磁場を印加し、時間と温度上昇との関係を測定した。この結果を図１７に示す。図１７から、ワセリン、ケナログ共に、測定直後（０秒）の温度は３０℃であり、交流磁場を３００秒間印加しても、温度上昇は２℃未満であったことが分かる。
以上から、金属サレン錯体化合物を混入した軟膏は、ワセリン単独及びケナログ単独に比べ、測定直後の温度が５℃高く、抗がん剤として適していることが分かる。また、特に金属サレン錯体化合物の濃度が２００ｍｍｏｌの軟膏は、交流磁場印加による温度上昇が顕著であり、抗がん剤としてより適していることが分かる。
なお、Ｍｎサレン錯体化合物、Ｃｒサレン錯体化合物、Ｃｏサレン錯体化合物のそれぞれについても同様の結果が得られた。
また、本発明に係る他の金属サレン錯体化合物についても、本実施形態と同様の実験を行ったところ、上記に準じた良好な結果が得られた。
（実施例９）
次に、２価のＦｅサレン錯体化合物（ＣＡＳ＃ １４１６７−１２−５；東京化成製）を、アガロースに１００ｍｍｏｌとなるように溶解したものを試験管に入れたサンプルを作成した。比較として、試験管に純水を入れたサンプルを作成した。次いで、これらのサンプルに対し、東京医研株式会社製の医療用近赤外線（波長６００ｎｍ〜１６００ｎｍ）を出力１０ＷのスーパーライザーＰＸ ＴｙｐｅＩで各々照射した。この時の時間と温度上昇の関係を図１８に示す。
図１８から、２価のＦｅサレン錯体化合物を含有するサンプルは、測定直後（０秒）の温度は約２６℃であり、３００秒後には約５３℃まで温度上昇したことが分かる。これに対し、純水を入れたサンプルは、測定直後（０秒）の温度は約１９℃であり、３００秒後の温度は約２３℃であり、殆ど温度上昇していないことが分かる。
以上から、２価のＦｅサレン錯体化合物をアガロースに溶解させたサンプルは、近赤外線照射に対する温度上昇が顕著であり、抗がん剤として適していることが分かる。

Claims (2)

  1. 金属部分を鉄とするサレン錯体化合物を有効成分として基剤に質量当たり０．０１％〜１０％混合させた軟膏であって、患部に塗布された後、交流磁場が印加されることにより、当該患部に対して発熱させる軟膏。
  2. 前記サレン錯体化合物が、下記式で示される請求項１に記載される軟膏。
    但し、上記式において、Ｍは、Ｆｅであり、ａ〜ｆ、Ｙのそれぞれは、水素である。
