Description
本発明は、HIVに対して有用なプロドラッグ化合物の製造方法、特に新規のアルキル化、アミド化、塩素化、およびホスフェート導入戦略を用いてプロドラッグ、1−ベンゾイル−4−[2−[4−メトキシ−7−(3−メチル−1H−1,2,4−トリアゾール−1−イル)−1−[(ホスホノオキシ)メチル]−1H−ピロロ[2,3−c]ピリジン−3−イル]−1,2−ジオキソエチル]−ピペラジンおよびその特定の中間体の製造方法に関する。本発明はまた、本明細書に記載の方法によって得られた化合物に関する。
1−ベンゾイル−4−[2−[4−メトキシ−7−(3−メチル−1H−1,2,4−トリアゾール−1−イル)−1−[(ホスホノオキシ)メチル]−1H−ピロロ[2,3−c]ピリジン−3−イル]−1,2−ジオキソエチル]−ピペラジンとして同定され、そして、構造式:
第1の実施態様において、本発明は、化学名が2−アミノ−2−(ヒドロキシメチル)プロパン−1,3−ジオール−(3−(2−(4−ベンゾイルピペラジン−1−イル)−2−オキソアセチル)−4−メトキシ−7−(3−メチル−1H−1,2,4−トリアゾール−1−イル)−1H−ピロロ[2,3−c]ピリジン−1−イル)メチル ホスフェートである、式I:

(c)化合物3のアミン基をメトキシ基に変換して、化合物:

(f)化合物6から二環式構造を形成して、

(h)その後、トリアゾリル部分を化合物8に加えて、

(i)TMG、NMPおよびNaIまたはK2CO3、MeCNおよびTBAIの存在下において、化合物:

をアセチル化および臭素化して、化合物:
を製造すること;および
(b)化合物2をニトロ化して、化合物:
を製造すること;および
(c)化合物3のアミン基をメトキシ基に変換して、化合物:
を製造すること;および
(d)次いで、化合物4を化合物:
に変換すること;および
(e)化合物5を化合物:
に変換すること;および
(f)化合物6から二環式構造を形成して、
を製造すること;および
(g)次いで、化合物を塩素化して、
を製造すること;および
(h)その後、トリアゾリル部分を化合物8に加えて、
を製造すること;および
(i)化合物9を構造:
に変換すること;および
(j)化合物10を修飾して、化合物:
を製造すること;および
(k)化合物11を反応させて、化合物:
を製造すること;および
(l)次いで、化合物12を化合物:
に変換すること;
(m)次いで、化合物13を化合物:
に変換すること;および
(n)次いで、化合物14を反応させて、化合物:

(o)次いで、化合物15を式Iで示される化合物に変換すること

- 工程(b)が酸性条件下で行われる、請求項1記載の方法。
- 工程(b)が硝酸を用いて行われる、請求項4記載の方法。
- 工程(b)が硝酸および硫酸を用いて行われる、請求項5記載の方法。
- 工程(c)がアルコール中でNaNO2および塩化トリメチルシリル(TMS−Cl)を用いて行われる、請求項1記載の方法。
- 前記アルコールがメタノールである、請求項7記載の方法。
- 工程(iv)がジクロロメタンを用いて行われる、請求項2記載の方法。
- 工程(vi)が水中アセトンおよびトロメタミンを用いて行われる、請求項2記載の方法。
