Description
kDaを超える分子量を持つタンパク質治療薬など、一つ以上の高分子量生理活性薬剤が含まれうる。過去において、身体の天然バリアを突破することが不能であることから、こうした生理活性薬剤の経皮的送達を得ることが困難または不可能であることが実証されてきた。

少なくとも部分的にパッチ300の周辺を越えて延び、使用者が部材をつかみやすくし、希望の方向に引くことができるようにする、タブ部分371(図12〜13)も含みうる。図12〜13に示すとおり、その「非アクティブ」の配置では、パッチ300の薬物送達組立品370は、いかなる有意な程度でもマイクロニードル330内へ流れないように、薬物化合物307をしっかりと保持する。パッチは、パッチから離れるように剥離部材に単に力をかけることにより、「起動」されうる。
310とサポート312の開口部(非表示)との間に予め形成されているシールは破壊される。この様に、薬物化合物107は、薬物送達組立品170からサポート112を経由してマイクロニードル130のチャネル131に流れ始めうる。薬物化合物307が、レザバー306からチャネル331にどのように流れるかの模範的図を、図16〜17に示す。明らかに、薬物化合物307の流れは、受動的に開始され、いかなる能動的置換機構(例えば、ポンプ)も必要としない。
JBX-9300FS EBLシステムを使用して、電子ビームリソグラフィーパターン形成プロセスによりパターンを形成させた。処理条件は以下のとおりであった:

曝露量 = 260 μC/cm2

現像 = 2分間の浸漬の後、30秒間イソプロピルアルコールでリンス
分間で、5 mTorrでの20 sccm Cl2および5 sccm Ar、600 W コイル、50 W RIEおよびDCバイアス96〜102 Vを用いた。シリコンエッチング深さは500ナノメートルであった。
パターンは、NTT Advanced Technology(東京)から購入した平坦な基材の表面である。パターンを、DN1(図24A)、DN2(図24B)、DN3(図24C)、DN4(図24D)およびNTTAT2(図24E)と指定した。型のSEM画像を、図24A、24B、および24Cに示し、膜の画像を図24Dおよび24Eに示す。図8は、図24A(DN1)の型を使用して形成したナノパターン膜を図示する。この特定の膜では、ポリマーの特徴が、前述の温度変化によって引き出された。図24Eのパターンの表面粗さは、34ナノメートルであることが判明した。
8(BB1で指定)は、0.6μmミリポアポリカーボネートフィルターを型として使用して形成した。25μmポリプロピレン膜を、フィルターの上部にかぶせた後、ポリプロピレンがフィルターの細孔に流入できるように加熱して溶かした。次に、型を冷却し、塩化メチレン溶剤を使用してポリカーボネート型を溶かした。
プロセスによりシリコンウェハー上に形成した。各ニードルには、反対方向に配置された2つのチャネルがあり、ニードル(図2では非表示)のベースにある1つの型を通過する穴と整列していた。

Claims (11)
- 皮膚バリアを越えた生理活性薬剤の送達のための装置であって、前記装置が、
マイクロニードルおよび前記マイクロニードルの表面上に作成した複数のナノ構造であって、前記ナノ構造が所定のパターンに配列されている複数のナノ構造と、
前記マイクロニードルと流体連通する組成物であって、前記組成物が、前記生理活性薬剤を含み、前記組成物が5センチポアズを超える粘性を持つ組成物とを含む、装置。 - 前記組成物が、濃度5mg/mLを超える前記生理活性薬剤を含む、請求項1に記載の装置。
- 前記生理活性薬剤が、100kDaを超える分子量を持つ、請求項1または請求項2に記載の装置。
- 前記生理活性薬剤がタンパク質治療薬である、請求項1〜請求項3のいずれか一項に記載の装置。
- 前記組成物が、溶剤、透過促進剤、増粘剤、湿潤剤、潤滑剤、および軟化剤から成る群から選択される医薬品として容認できる賦形剤を含む、一つ以上の医薬品として容認できる賦形剤をさらに含む、請求項1〜請求項4のいずれか一項に記載の装置。
- 前記パターンが、マイクロ構造をさらに含み、前記ナノ構造が前記マイクロ構造よりも小さな断面寸法を持つ、請求項1〜請求項5のいずれか一項に記載の装置。
- 前記マイクロ構造が、500ナノメートルを超える断面寸法を持ち、前記ナノ構造が300ナノメートル未満の断面寸法を持つ、請求項6に記載の装置。
- 前記複数のナノ構造が、
第一のナノ構造と、
前記マイクロ構造の前記断面寸法よりも小さく、前記第一のナノ構造の前記断面寸法よりも大きい断面寸法を持つ第二のナノ構造をさらに備える、請求項6または請求項7に記載の装置。 - 前記ナノ構造の少なくとも一部分が、
a)中心間の間隔が50ナノメートル〜1マイクロメートル、
b)高さが10ナノメートル〜20マイクロメートル、
c)アスペクト比が0.15〜30という特性のうち一つ以上を持つ、請求項1に記載の装置。 - 前記パターンが1を超えるフラクタル次元を持つ、請求項1〜請求項9のいずれか一項に記載の装置。
- 前記複数のナノ構造を含む、前記マイクロニードルの表面が、10ナノメートル〜200ナノメートルの平均表面粗さおよび/または4MPa〜320MPaの有効せん断係数を持つ、請求項1〜請求項10のいずれか一項に記載の装置。
