Description
ステップ1:チダミドを2mol/L希塩酸溶液に添加し、室温で撹拌することによって溶解させ、水で希釈し、結果として生じる溶液に2mol/L NaOH溶液を滴下して添加し、30分間撹拌し、次いで濾過する;ここでステップ1におけるチダミドと希塩酸溶液との重量比は、1:4.2〜1:4.4の範囲内であり、チダミドと水との重量比は、1:25〜1:30の範囲内であり、チダミドとNaOH溶液との重量比は、1:2.5〜1:2.7の範囲内である;
ステップ2:結果として生じる固体を収集し、水に添加し、混合物に2mol/L NaOH溶液を滴下して添加し、60分間撹拌し、次いで濾過し、結果として生じる固体を収集し、水で洗浄して5〜7のpH値にし、次いで乾燥させる;ここでステップ2におけるチダミドと水との重量比は、1:15〜1:25の範囲内であり、チダミドとNaOH溶液との重量比は、1:1.5〜1:2.0の範囲内である
を含む。
X線粉末回折の試験条件:計測器:D/MAX−1200(日本);線源:Cu−Kα(40kV、40mA)。
チダミド(約95%の純度)4.84kgを、2mol/L希塩酸溶液20.91kgに添加し、室温で撹拌によって溶解させた。引き続いて、水137.8kgを添加し、5分間撹拌した。得られた溶液に、2mol/L NaOH溶液21.35kgを滴下して添加し、30分間撹拌し、濾過した。結果として生じる固体を水96.70kgに添加し、引き続いて2mol/L NaOH溶液8.39kgを滴下して添加した。混合物を60分間撹拌し、濾過し、水で洗浄して5〜7のpH値にした。結果として生じる固体を真空下で80℃にて24時間乾燥させてチダミド結晶形A 4.32kgを99.2%の純度で得た。この結晶形のX線粉末回折パターン、赤外線スペクトル、および示差走査熱量測定パターンを、それぞれ図3、図4、および図5に示した。
チダミド結晶形A 1gをジメチルスルホキシド10mLに添加し、室温で撹拌することによって溶解させた。結果として生じる溶液に水100mLを滴下して添加し、30分間撹拌し、濾過した。結果として生じる固体を真空下で80℃にて24時間乾燥させてチダミド結晶形Bを99.2%の純度で得た。この結晶形のX線粉末回折パターン、赤外線スペクトル、および示差走査熱量測定パターンを、それぞれ図6、図7、および図8に示した。

Claims (7)
- そのX線粉末回折パターンが4.18°、6.61°、8.42°、12.69°、17.85°、18.34°、19.27°、20.10°、20.59°、21.58°、23.70°、23.96°、25.52°、27.00°、27.90°、29.59°、および29.94°の2θに特徴的なピークを有し、その赤外線スペクトルが3412、3282、3199、3043、1654、1615、1524、1514、1497、1442、1418、1332、1296、1234、1198、1183、1166、および1027cm −1 に特徴的な吸収ピークを有し、その示差走査熱量測定パターンが239.4℃に吸熱ピークを有することを特徴とするチダミド結晶形A。
- ステップ1:チダミドを2mol/L希塩酸溶液に添加し、室温で撹拌することによって溶解させ、水で希釈し、結果として生じる溶液に2mol/L NaOH溶液を滴下して添加し、30分間撹拌し、次いで濾過する;ここでステップ1におけるチダミドと該希塩酸溶液との重量比は、1:4.2〜1:4.4の範囲内であり、チダミドと水との重量比は、1:25〜1:30の範囲内であり、チダミドとNaOH溶液との重量比は、1:2.5〜1:2.7の範囲内である;
ステップ2:結果として生じる固体を収集し、水に添加し、混合物に2mol/L NaOH溶液を滴下して添加し、60分間撹拌し、次いで濾過し、結果として生じる固体を収集し、水で洗浄して5〜7のpH値にし、次いで乾燥させる;ここでステップ2におけるチダミドと水との重量比は、1:15〜1:25の範囲内であり、チダミドとNaOH溶液との重量比は、1:1.5〜1:2.0の範囲内である
を含むことを特徴とする、請求項1に記載のチダミド結晶形Aを調製するための方法。 - 細胞分化増殖関連疾患を処置するための医薬の調製における請求項1に記載のチダミド結晶形Aの使用。
- そのX線粉末回折パターンが4.18°、8.43°、12.65°、16.90°、17.83°、19.47°、20.13°、20.58°、21.22°、21.54°、21.92°、23.94°、25.55°、26.98°、27.92°、29.58°、および29.90°の2θにピークを有し、その赤外線スペクトルが3412、3327、3272、3196、3043、1653、1619、1570、1515、1496、1444、1419、1331、1357、1296、1278、1267、1232、1198、1184、1166、および1038cm −1 に特徴的な吸収ピークを有し、その示差走査熱量測定パターンが214.1℃および241.2℃に吸熱ピークを有することを特徴とするチダミド結晶形B。
- チダミド結晶形Aをジメチルスルホキシドに添加し、室温で撹拌することによって溶解させ、結果として生じる溶液に水を滴下して添加し、30分間撹拌し、次いで濾過し、結果として生じる固体を収集し、乾燥させて化合物を得、ここで、該チダミド結晶形Aとジメチルスルホキシドとの重量比は、1:10〜1:20の範囲内であり、該チダミド結晶形Aと水との重量比は、1:100〜1:200の範囲内であることを特徴とする、請求項4に記載のチダミド結晶形Bを調製するための方法。
- 細胞分化増殖関連疾患を処置するための医薬の調製における請求項4に記載のチダミド結晶形Bの使用。
- 細胞分化増殖関連疾患を処置するための医薬製剤であって、請求項1に記載のチダミド結晶形Aまたは/および請求項4に記載のチダミド結晶形B、ならびに薬学的に許容される賦形剤を含むことを特徴とする医薬製剤。
