Description
ドライパウダー製剤を収容し、当該容器からの計量された用量の薬剤を送達するための設計を有する容器;
前記ドライパウダー薬剤の凝集物を粉砕するためのサイクロン脱凝集器;及び、
マウスピースを通る吸入に誘導される空気流を配向するための送達経路、当該送達経路は前記計量された用量の薬剤に延在する、
を含み、
前記製剤は、d10<1μm, d50=1-3μm, d90=3.5-6μm及びNLT99%<10μmの粒径分布を有する吸入可能なβ2−アゴニスト、及び、ラクトースキャリアを含む。
第一端から第二端へ軸に沿って延在する旋回チャンバーを規定する内壁;
吸入器の送達経路と旋回チャンバーの第一端との間の流体の交通を提供するための、旋回チャンバーの第一端のドライパウダー供給ポート;
脱凝集器の外部領域と旋回チャンバーの第一端との間の流体の交通を提供し、旋回チャンバーの第一端に近接する、旋回チャンバーの内壁の少なくとも1つの入口ポート;
旋回チャンバーの第二端と脱凝集器の外部領域との間の流体の交通を提供する出口ポート;及び、
少なくとも部分的にチャンバーの軸から放射状に外向きに延在する旋回チャンバーの第一端の翼、翼の各々は少なくとも部分的に軸を横断する方向に面した斜めの表面を有している;
を含み、出口ポートでの呼吸に誘導される低圧が、ドライパウダー供給ポートと入口ポートとを通じて、空気を旋回チャンバーに流入させる。
前記分配ポートと交通し、圧開放ポートを含む通路;
密封容器の内部と通路の圧開放ポートとの間の流体の交通を提供するコンジット;及び、
通路に可動的に受けられるカップ組立体、分配ポートと整列した時に薬剤を受けるようにされている凹部、凹部が分配ポートと整列していない時に分配ポートを封じるようにされている第一の封止表面、及び、凹部が分配ポートと整列する時に圧開放ポートを封止し、また、凹部が分配ポートと整列していない時に圧開放ポートを開放する第二の封止表面と、を含む。
フマル酸フォルモテロール二水和物の2つのサンプルが、ジェットミルで微粒化された。2つのバッチは7544MA(従来法のミル)及び7544MO(本発明)というコードを与えられた。微粒化の条件が表1に示される。
薬物動態(PK)臨床研究が行われた。PK研究は、多くの鍵となる製剤パラメータ、計量用量(装置の用量カップの体積)、製剤のブレンドの強度、薬物の粒子サイズ及びラクトースの粒子サイズで、ステップワイズな方法を用いて試験された。PK研究は、中程度の強度の製品(160/4.5μg)で行われた。バッチAはフォルモテロール7544MAを含み、バッチBはフォルモテロール7544MOを含んでいた。バッチA、Bの両方について、吸入器、ブデソニド及びラクトースは同一であった。
Claims (18)
- ドライパウダー製剤を収容する容器;及び、
マウスピースを通る吸入に誘導される空気流を配向するための送達経路
を含むドライパウダー吸入器であって、
前記容器は、計量された用量の薬剤を当該容器から送達するように構成されたものであり、
前記送達経路は、前記計量された用量の薬剤まで及ぶものであり、
前記製剤は、ラクトースキャリアと吸入可能なβ2−アゴニストとを含み、当該吸入可能なβ2−アゴニストが、d10<1μm、1μm≦d50≦3μm、3.5μm≦d90≦6μm及びNLT99%<10μmの粒子サイズ分布を有するものであり、ここで、d10、d50、及びd90はそれぞれ、体積基準累積粒度分布の微粒側から累積10%、50%、90%の粒径を表し、NLT99%は、99%以上の粒子を表す、
ドライパウダー吸入器。 - さらに脱凝集器を含む、請求項1に記載の吸入器。
- 前記脱凝集器が、サイクロン脱凝集器であり、当該サイクロン脱凝集器が、
第一端から第二端へ軸に沿って延在する旋回チャンバーを規定する内壁;
前記吸入器の前記送達経路と前記旋回チャンバーの第一端との間の流体の流通を提供するための、旋回チャンバーの第一端にあるドライパウダー供給ポート;
前記脱凝集器の外部領域と前記旋回チャンバーの第一端との間の流体の流通を提供する、前記旋回チャンバーの第一端に近接する旋回チャンバーの内壁にある、少なくとも1つの入口ポート;
前記旋回チャンバーの第二端と前記脱凝集器の外部領域との間の流体の流通を提供する出口ポート;及び、
少なくとも一部分が前記チャンバーの軸から放射状に外向きに延在する、旋回チャンバーの第一端にある翼、当該翼の各々は前記軸を横断する方向に少なくとも一部分が面する斜め表面を有している;
を含み、
前記出口ポートでの呼吸に誘導される低圧が、前記ドライパウダー供給ポートと前記入口ポートとを通じて旋回チャンバーに入る空気流を起こす、請求項2に記載の吸入器。 - 前記容器が、分配ポートを含む気密容器であり、前記吸入器が、さらに
前記分配ポートと連通し、圧開放ポートを含む通路;
前記気密容器の内部と前記通路の前記圧開放ポートとの間の流体の流通を提供するコンジット;及び、
前記分配ポートと整列した時に薬剤を受けるようになっている凹部と、前記凹部が前記分配ポートと並んでいない時に前記分配ポートをシールするよう設定された第一のシール表面と、前記凹部が分配ポートと整列する時に前記圧開放ポートをシールし、前記凹部が前記分配ポートと並んでいないときには前記圧開放ポートを解放するように設定された第二のシール表面と、を有する、前記通路に可動に受けられるカップ組立体、を含む、請求項1〜3のいずれか1項に記載の吸入器。 - 前記β2−アゴニストが、サルブタモール、レボサルブタモール、テルブタリン、ピルブテロール、プロカテロール、クレンブテロール、メタプロテレノール、フェノテロール、ビトルテロール、リトドリン、イソプレナリン、フォルモテロール、サロメテロール、バンブテロール、インダカテロール、カルモテロール、又は薬学的に許容可能なそれらの塩から選択される、請求項1〜4のいずれか1項に記載の吸入器。
- 吸入可能なβ2−アゴニストがフォルモテロールである、請求項5に記載の吸入器。
- 吸入可能なβ2−アゴニストが、フマル酸フォルモテロールである、請求項1又は2に記載の吸入器。
- 前記製剤がさらに、一又は複数の追加的な吸入可能な活性成分を含む、請求項1〜7のいずれか1項に記載の吸入器。
- 前記追加的な吸入可能な活性成分がコルチコステロイドである、請求項8に記載の吸入器。
- 前記製剤が、フマル酸フォルモテロール、ブデソニド及びラクトースキャリアを含み、前記フマル酸フォルモテロールの粒子サイズ分布がd10<1μm、1μm≦d50≦3μm、3.5μm≦d90≦6μm及びNLT 99%<10μmである、請求項1又は2に記載の吸入器。
- 前記β2−アゴニストが、0.4μm≦d10≦0.6μm、1.5μm≦d50≦2.5μm及び3.6μm≦d90≦5.1μmの粒子サイズ分布を有する、請求項1〜10のいずれか1項に記載の吸入器。
- 前記β2−アゴニストが、0.46μm≦d10≦0.53μm、1.68μm≦d50≦1.92μm及び3.68μm≦d90≦5.07μmの粒子サイズ分布を有する、請求項11に記載の吸入器。
- 呼吸器疾患の治療に用いられる、請求項1〜12のいずれか1項に記載の吸入器。
- 前記呼吸器疾患が、喘息、又は、慢性閉塞性肺疾患である、請求項13に記載の吸入器。
- 前記喘息が、喘息管理に関する国際指針であるGINA(Global Initiative for Asthma)ステージ1,2,3又は4に分類される、軽症、中等症又は重症の喘息である、請求項14に記載の吸入器。
- ドライパウダー吸入器における使用のための吸入可能な医薬製剤であって、ラクトースキャリアと、吸入可能なβ2−アゴニストとを含み、当該吸入可能なβ2−アゴニストが、d10<1μm、1μm≦d50≦3μm、3.5μm≦d90≦6μm及びNLT99%<10μmの粒子サイズ分布を有するものである、吸入可能な医薬製剤。
- 前記β2−アゴニストが、フォルモテロールである、請求項16に記載の吸入可能な医薬製剤。
- 前記β2−アゴニストが、フマル酸フォルモテロールである、請求項17に記載の吸入可能な医薬製剤。
