Description
抗菌ペプチド(anti-microbial peptide)は、生体内で自然免疫メカニズムを担当する天然抗菌物質であって、細菌、真菌、原虫、ウィルスを含む各種微生物に対して抗菌力を有しており、局所的な感染抑制と全身免疫反応を誘導する特性を有するペプチド物質である。通常、抗菌ペプチドは、両親媒性の構造を有しており、抗菌作用メカニズムとして抗菌ペプチドのカチオン部位が微生物細胞膜が有しているアニオン性リン脂質に結合して微生物の細胞膜を破壊する原理によって抗菌活性を示す。
抗生物質や合成抗菌剤など種々の薬剤による従来の抗菌剤では、抵抗性微生物が出現し、耐性菌に対する対策は新たな抗菌剤を開発するしかない。
しかし、抗菌ペプチドの産生を促進する方法であれば、上記のような問題はなく、自己生体防御機構を増強できる。
又は治療(アテローム性動脈硬化症、冠状動脈性心臓病、脳卒中、動脈閉塞性疾患または
潰瘍)の用途が知られている。(特許文献7〜9)
ヒト表皮角化細胞を3×105個/ウェルとなるように6ウェルプレートに播種し、5%FBSを含むDMEM培地にて一晩培養した。南天実エキス(調製例1)もしくは南天葉エキス(調製例3)を溶解した培地に交換し、37℃、5% CO2インキュベーター内で24時間培養した。細胞からのTotal RNA抽出はQuickGene RNA cultured cell HC Kit Sを用い、倉敷紡績社のプロトコールに従って調製した。cDNA合成後に下記表のプライマーを使用してリアルタイムPCRにより遺伝子発現を確認した。内部標準として下記表1のGAPDHを使用した。また、南天実エキスもしくは南天葉エキスの培地への添加量は、細胞毒性を示さない濃度を添加した。
Claims (1)
- ナンテン(Nandina domestica)の実の乾燥粉砕物1kgに水を10L添加し、90℃にて6時間還流して抽出した南天エキスを有効成分とするβ−ディフェンシン1及び/又はカテリシジン遺伝子発現促進剤。
