<Caco-2細胞におけるロイシン透過率の評価>
1.サンプルの調製
被験物質として、以下のものを用いた。
(1)大麦の茎葉として、出穂前に刈り取ったものを洗浄後、裁断、乾燥後に粉砕して得られた大麦茎葉の粉砕末(目開き75μmの篩を90%以上通過するもの;株式会社東洋新薬社製)を用いた。チューブに24mgの大麦茎葉の粉砕末を入れ、バッファー1mLを加えた後にボルテックスを行った。その後、15,000×gで5分間室温にて遠心を行い、上清を回収した。これをフィルター滅菌後、希釈して被験物質とした。
(2)分枝鎖アミノ酸として、L-ロイシン(ナカライテスク社製:20327-46)を用いた。チューブに13.1mgのL-ロイシンを入れ、バッファー1mLを加えて溶解した(100mM)。これをフィルター滅菌後、希釈して被験物質とした。
各被験物質の終濃度を下記に示す。
<Evaluation of leucine permeability in Caco-2 cells>
1. Sample preparation
The following were used as test substances.
(1) Ground powder of barley stems and leaves obtained by washing, cutting, drying, and crushing barley stems and leaves that are harvested before heading (more than 90% passes through a 75 μm sieve; Toyo Co., Ltd. (manufactured by Shinyakusha) was used. 24 mg of ground barley stem and leaf powder was placed in a tube, 1 mL of buffer was added, and the tube was vortexed. Thereafter, centrifugation was performed at 15,000×g for 5 minutes at room temperature, and the supernatant was collected. After filter sterilization, this was diluted and used as a test substance.
(2) L-leucine (manufactured by Nacalai Tesque: 20327-46) was used as the branched chain amino acid. 13.1 mg of L-leucine was placed in a tube, and 1 mL of buffer was added to dissolve it (100 mM). After filter sterilization, this was diluted and used as a test substance.
The final concentration of each test substance is shown below.

2.細胞培養および被験物質の添加
(1)37℃、5%COインキュベーター内で、75cmフラスコを用いて、ヒト大腸癌由来細胞(Caco-2)を培養した。培地は10%FBS-DMEMを用いた。
(2)24-wellプレートに培地を600μl添加し、インサートウェルを移した。
(3)トリプシン処理により浮遊させた細胞を、75cmフラスコからインサートウェルに4.0×10cells/wellの細胞密度となるように100μl播種した。
(4)37℃、5%COインキュベーター内で14日間前培養し、その間は2-3日毎に培地交換を実施した。サンプル添加前の培地交換の際は、インサートウェル外側(基底膜側ウェル)を1000μl、インサートウェル内側(管腔側ウェル)を350μlの10%FBS-DMEMに置換した。
(5)Millicell ERS-2にて電気抵抗値を測定し、1000Ω以上になっているウェルを使用した。また、被験物質を添加する各群の電気抵抗値の平均が同一となるように群分けした。
(6)1回あたり1000μLのバッファーにてインサートウェルの外側を3回、内側を3回洗浄後、1000μLのバッファー入った24-wellプレートに移し、インサートウェルに350μLのバッファーを加えた。
(7)COインキュベーター内で30分間培養し、細胞をバッファーに順化させた。
(8)インサートウェル内側のバッファーを175μL除いた後、2倍濃度に調製した被験物質溶液を175μLずつ添加し、COインキュベーター内で2時間培養した。
(9)培養後、基底膜側ウェルのサンプルを1.5mLチューブに全量回収した。

When replacing the medium before sample addition, the outside of the insert well (well on the basement membrane side) was replaced with 1000 μl, and the inside of the insert well (well on the luminal side) was replaced with 350 μl of 10% FBS-DMEM.
(5) The electrical resistance value was measured using Millicell ERS-2, and wells with a resistance of 1000Ω or more were used. Furthermore, the test substances were divided into groups such that the average electrical resistance value of each group to which the test substance was added was the same.
(6) After washing the outside of the insert well three times and the inside three times with 1000 μL of buffer each time, it was transferred to a 24-well plate containing 1000 μL of buffer, and 350 μL of buffer was added to the insert well.
(7) Cultured in a CO 2 incubator for 30 minutes to acclimatize the cells to the buffer.
(8) After removing 175 μL of the buffer inside the insert well, 175 μL each of the test substance solution prepared at twice the concentration was added and cultured in a CO 2 incubator for 2 hours.
(9) After culturing, the entire sample of the basement membrane side well was collected into a 1.5 mL tube.

3.高速液体クロマトグラフィーによるロイシン濃度の測定
上記2(9)で回収した基底膜側ウェルのサンプルをチューブに500μL分注後、遠心エバポレーターを用いてサンプルを乾固した。さらに、400μLのサンプルを同じチューブで乾固した後、200μLの100mM HClを添加し、10分間超音波をかけて溶解した。得られた濃縮液を高速液体クロマトグラフィーによりロイシン濃度を測定した。測定結果を下記の計算式にあてはめ、ロイシン透過率を算出した。
