〔実験1:動物試験(ラット外傷性関節症モデルにおける滑膜の遺伝子発現解析)〕
実験には、10週齢の雄のWisterラット(Crij:WI)を用いた。Sham手術(偽手術、皮膚および筋層の切開のみ実施)によるOA非発症モデルと、内側側副靭帯および内側半月板切除手術(MTT手術)によるOA発症モデルとを作製した。手術は全個体で右膝のみに実施し、左膝は無処置膝とした。手術から2日後に動物飼育室に導入し、1週間の予備飼育の後、各試験群の平均体重に偏りがないように割付を行った。ラットの飼育条件は12時間明暗周期、室温24±2℃とした。飼料は、一般飼育用固形飼料(ラボMRストック、日本農産工業社製)を用いて、常時給餌・給水条件で飼育した。
[Experiment 1: Animal study (gene expression analysis of synovial membrane in a rat traumatic arthropathy model)]
For the experiment, 10-week-old male Wister rats (Crij:WI) were used. An OA-free model was prepared by sham surgery (sham surgery, only incision of the skin and muscle layer was performed) and an OA-onset model was prepared by medial collateral ligament and medial meniscus resection (MTT surgery). Surgery was performed only on the right knee of all rats, and the left knee was left untreated. Two days after surgery, the rats were introduced into the animal breeding room, and after one week of preliminary breeding, they were assigned so that there was no bias in the average weight of each test group. The rats were kept under a 12-hour light-dark cycle and at a room temperature of 24±2°C. They were kept using solid feed for general breeding (Labo MR Stock, manufactured by Nippon Nosan Kogyo Co., Ltd.) and were fed and watered at all times.
試験条件を表1に示す。果実由来ポリフェノールとしては、リンゴ未完熟果実由来ポリフェノール抽出物「アップルフェノン(登録商標)」(AP)(BGG JAPAN社製)を投与した。N-アセチルシステイン(NAC)は陽性対照として投与した。Sham群およびControl群には滅菌水を投与した。被験物質は胃ゾンデにより試験期間中1日1回毎日経口投与した。 The test conditions are shown in Table 1. 8週間の投与終了後に解剖を行った。セボフレン(登録商標)吸入麻酔液(丸石製薬株式会社)による麻酔下で下大静脈採血を行ってラットを安楽死させ、右膝の膝蓋骨下の滑膜組織および右後肢膝関節を採取した。右後肢膝関節の関節軟骨周辺部の組織切片を作製し、変形性ひざ関節症が発症していることを確認した。 After 8 weeks of treatment, the rats were dissected. 滑膜の遺伝子発現解析のために、右膝関節より滑膜組織の採取を行った。単離した滑膜組織はPBSで洗浄後、RNAlater(Invitrogen社製)に4℃で一晩浸漬した後、TRIzol(登録商標)reagent(Invitrogen社製)で処理し、totalRNAを精製した。これを鋳型として、PrimeScript RT reagent Kit(Perfect Real Time、タカラバイオ株式会社製)を用いてcDNAを合成した。このcDNAを鋳型として、TB Green Premix Ex Taq II(タカラバイオ株式会社製)およびThermal Cycler Dice Real Time System TP800(タカラバイオ株式会社製)を用いて、定量的リアルタイムPCRを行った。用いたプライマーの塩基配列を表2および配列番号1~4に示す。 For gene expression analysis of synovial membrane, synovial tissue was collected from the right knee joint. The base sequences of the primers used are shown in Table 2 and SEQ ID NOs: 1 to 4.

滑膜におけるIL-10の遺伝子発現解析結果を図1に示す。Control群ではSham群に比べてIL-10の遺伝子発現量が比較的低値となった。AP投与群ではControl群に比べてIL-10の遺伝子発現量が顕著に増加した。IL-10は抗炎症性サイトカインのひとつとして知られている。果実由来ポリフェノールはIL-10の遺伝子発現を強く促進し、関節の炎症、特に滑膜の炎症を軽減する可能性が示唆された。 The results of gene expression analysis of IL-10 in the synovium are shown in Figure 1. In the control group, the gene expression level of IL-10 was relatively low compared to the sham group. In the AP administration group, the gene expression level of IL-10 was significantly higher compared to the control group. IL-10 is known as an anti-inflammatory cytokine. It has been suggested that fruit-derived polyphenols strongly promote the gene expression of IL-10, and may reduce joint inflammation, especially inflammation of the synovium.

滑膜におけるTNF-αの遺伝子発現解析結果を図2に示す。Control群ではSham群に比べてTNF-αの遺伝子発現量が有意に高値となった。一方で、AP投与群ではControl群に比べてTNF-αの遺伝子発現量が有意に低値となり、Sham群と同等であった。TNF-αは代表的な炎症性サイトカインのひとつである。果実由来ポリフェノールはTNF-αの遺伝子発現を抑制し、関節の炎症、特に滑膜の炎症を軽減する可能性が示唆された。 The results of gene expression analysis of TNF-α in synovium are shown in Figure 2. It has been suggested that fruit-derived polyphenols may suppress TNF-α gene expression and reduce joint inflammation, particularly inflammation of the synovium.
滑膜におけるMMP-13の遺伝子発現解析結果を図3に示す。Control群ではSham群に比べてMMP-13の遺伝子発現量が有意に高値となった。一方で、AP投与群ではControl群に比べてMMP-13の遺伝子発現量が低値となり、陽性対照であるNAC投与群とほぼ同等であった。MMP-13は軟骨の主成分のひとつであるコラーゲンを分解する働きがある。果実由来ポリフェノール抽出物はMMP-13の遺伝子発現を抑制し、軟骨分解の抑制に作用する可能性が示唆された。 The results of gene expression analysis of MMP-13 in the synovium are shown in Figure 3. MMP-13 has the function of breaking down collagen, one of the main components of cartilage. It was suggested that fruit-derived polyphenol extract may suppress MMP-13 gene expression and act to inhibit cartilage degradation.
滑膜におけるSOD1の遺伝子発現解析結果を図4に示す。Control群ではSham群に比べてSOD1の遺伝子発現量が有意に高値となった。一方で、AP投与群ではControl群やNAC投与群に比べてSOD1の遺伝子発現量が有意に低値となり、Sham群と同等であった。炎症により亢進された滑膜の酸化ストレスが、APが有する抗酸化作用によって軽減されて、SOD1の遺伝子発現量が低下したと推測される。 The results of gene expression analysis of SOD1 in the synovium are shown in Figure 4. It is speculated that the antioxidant effect of AP reduces oxidative stress in the synovium, which is enhanced by inflammation, resulting in a decrease in SOD1 gene expression levels.
NACおよびAPの抗酸化作用(ORAC値)をCao G et al., Free Radical Biology and Medicine, vol. 14, Issue 3, 303-311, March 1993に記載の方法に基づいて測定した。NACおよびAPの抗酸化作用は標準物質として用いたビタミンC(L-アスコルビン酸)よりも高いこと、ならびにNACおよびAPの抗酸化作用の程度と図4で示されたSOD1の抑制作用の程度との間に相関あることが示された(図5)。 The antioxidant activity (ORAC value) of NAC and AP was measured based on the method described in Cao G et al., Free Radical Biology and Medicine, vol. 14, Issue 3, 303-311, March 1993.
