肌荒れ、日焼けや乾燥により生じる角層の落屑等の肌状態の悪化に関し、デスモゾーム蛋白質の増加が原因の一つとして知られている。

<脂肪酸含有PBSの調製>
被験試料として、表1に示す脂肪酸を選定した。

電子天秤を用いて、各脂肪酸をエッペンチューブに分注し、各脂肪酸の濃度が2μg/μLとなるようにメタノールを加え調製した。次いで、50mLチューブにPBSを12mL分注し、各脂肪酸を60μL加え、ボルテックスにかけ、脂肪酸含有PBSを調製した。
コントロールについては、50mLチューブにPBSを12mL分注し、メタノールを60μL加え、ボルテックスにかけて調製した。

次いで、スライドガラスをPBSで濯ぎ、0.5%Triton X/PBSに5分間室温で浸漬させ、透過処理を行った。さらに、PBSで濯いだ後、20%ブロックエース(ケー・エー・シー)により、1時間室温でブロッキングを行った。その後、一次抗体(anti-Desmoglein 1 mouse monoclonal, DSG1-P23;PROGEN Biotechnik GmbH)による反応を室温で2時間行った。
<Immunostaining test>

The slide glass was then rinsed with PBS and immersed in 0.5% Triton X/PBS at room temperature for 5 minutes for permeabilization. After rinsing with PBS, blocking was performed with 20% Block Ace (KAC) at room temperature for 1 hour. After that, reaction with a primary antibody (anti-Desmoglein 1 mouse monoclonal, DSG1-P23; PROGEN Biotechnik GmbH) was performed at room temperature for 2 hours.

図1~3に示す通り、リノール酸メチル、リシノール酸メチル、リシノール酸、リノレン酸、メトプレン、ステアリン酸の6成分において、デスモグレイン1量の有意な減少が認められた。
特に、リノール酸メチル、リシノール酸メチル、リシノール酸及びステアリン酸の4成分においては、顕著なデスモグレイン1量の減少効果が認められた。
As shown in Figures 1 to 3, a significant decrease in the amount of desmoglein 1 was observed in six components, namely, methyl linoleate, methyl ricinoleate, ricinoleic acid, linolenic acid, methoprene, and stearic acid.
In particular, the four components methyl linoleate, methyl ricinoleate, ricinoleic acid, and stearic acid were found to have a significant effect of reducing the amount of desmoglein 1.
