<試験例>

乳酸菌としては、粉末状の有胞子性乳酸菌(学名:Bacillus coagulans)を用いた。
ピペリンとしては、粉末状のピペリンを用いた。
<Test Example>
The peat used was paste-like peat from Northern Europe.
Powdered sodium D-pantothenate was used as the vitamin.
As the vitamin, powdered vitamin D3 was used.
As the lactic acid bacteria, powdered spore-forming lactic acid bacteria (scientific name: Bacillus coagulans) was used.
As the piperine, powdered piperine was used.
<細胞賦活試験>
(1)細胞培養
37℃、5%CO2インキュベーター内で、75cm2フラスコを用いて、ヒト肝癌由来細胞(HepG2、)を培養した。培地としては、P/S(ペニシリン/ストレプトマイシン)を1%添加したDMEMに、10%ウシ胎児血清(FBS)となるように調製したものを使用した。次いでトリプシン処理により浮遊させた細胞をFBS含有培地に懸濁し、75cm2フラスコからコラーゲンコートした96well plateの各wellに1×104cells/wellの細胞密度で播種した。その後、37℃、5%CO2インキュベーター内で24時間前培養した。各wellより培地を除去後、所定濃度に調製した被験物質含有の調製用培地を100μL/well添加し、37℃、5%CO2インキュベーター内で24時間培養した。なお、調製用培地としては、P/Sを1%添加したDMEMに、10%FBS、0.5%DMSO、0.5mM H202となるように調製したものを使用した。
<Cell activation test>
(1) Cell culture Human hepatoma-derived cells (HepG2, ) were cultured in a 75 cm2 flask in a 37°C, 5% CO2 incubator. The medium used was DMEM supplemented with 1% P/S (penicillin/streptomycin) and adjusted to 10% fetal bovine serum (FBS). Next, cells suspended by trypsin treatment were suspended in FBS-containing medium, and seeded at a cell density of 1 x 104 cells/well from the 75 cm2 flask into each well of a collagen-coated 96-well plate. Then, the cells were pre-cultured for 24 hours in a 37°C, 5% CO2 incubator. After removing the medium from each well, 100 μL/well of a preparation medium containing the test substance prepared to a predetermined concentration was added, and the cells were cultured for 24 hours in a 37°C, 5% CO2 incubator. The medium used for preparation was prepared by adding 1% P/S to DMEM, 10% FBS, 0.5% DMSO, and 0.5 mM H2O2 .
(2)被験試料の調製
各被験物質を、調製用培地を用いて、表1乃至4の濃度となるように調製した。泥炭と、他素材(ビタミン、乳酸菌及びピペリンから選ばれる少なくとも1種)の組み合わせ(実施例1乃至4)は、泥炭と他素材を1:1の割合で混合したものを使用した。

上記の24時間培養後、培地を除去し、無血清DMEM200μL/wellで洗浄した。その後、30容量倍に希釈したCell counting kit-8溶液(同仁化学製社製)を各wellに150μLずつ添加し、37℃、5%CO2インキュベーター内に静置し適度に発色させた後、450nmにおける吸光度を測定した。

得られた結果を図1乃至4に示す。図1乃至4には、3連の平均値及び標準偏差を記載した。数値は、高いほど細胞賦活効果が高いことを示す。図1乃至4に示す結果から明らかな通り、泥炭と、ビタミン、乳酸菌及びピペリンから選ばれる少なくとも1種とを含有する実施例1乃至4は、泥炭、ビタミン、乳酸菌、ピペリンのそれぞれのみからなる比較例1乃至6よりも、細胞賦活の優れた作用を有することが分かる。したがって、泥炭と、ビタミン、乳酸菌及びピペリンから選ばれる少なくとも1種とを含有することにより、細胞を賦活させ、代謝を促進して、その結果、抗肥満効果、ダイエット効果を得られる。 The results are shown in Figures 1 to 4.
