Abstract

PURPOSE: To measure the enzyme immunity in a small amount of a sample with an antigen-antibody reaction, by solidifying an antibody on the inner wall of a test tube, introducing beads in which another antibody is solidified on the surfaces thereof, a test solution and an antigen labeled with an enzyme into the test tube to carry out the antigen-antibody reaction, and removing the beads from the test tube to carry out the remaining antigen-antibody reaction.
CONSTITUTION: An antibody A is solidified on the inner wall of a test tube 1, and another antibody B is solidified on the surfaces of beads 2. The beads 2 are then introduced into the test tube 1, and a test solution 3 is added thereto. An antigen B labeled with an enzyme is then added to bond the antigen A in the test solution 3 to the antibody A on the surface of the inner wall in the test tube 1. The antigen B and the antigen B labeled with the enzyme are competitively bonded to the antibody B on the surfaces of the beads 2. The amount of the antigen B labeled with the enzyme linked to the antibody B depends upon the amount of the antibody B to be determined. After the completion of the bonding, the beads 2 are taken out and washed to measure the activity of the enzyme linked to the antibody B on the beads 2. On the other hand, the test tube 1 is washed, and the antigen A labeled with an enzyme is added and linked to the antibody A.
