Description
【発明の詳細な説明】 ［産業上の利用分野］ 本発明は、副腎皮質刺激ホルモン放出因子（Corticotro
pin Releasing Factor,以下CRFと略すことがある。）を
含有する製剤に関し、更に詳しくは長期安定なCRF含有
凍結乾燥製剤に関するものである。

［従来の技術および発明が解決しようとする課題］ CRFは、アミノ酸41個からなるポリペプチドであり、ラ
ット、ヒツジ、ヒト等での存在が明らかになっている視
床下部から分泌されるホルモンの一種である。CRFは副
腎皮質刺激ホルモン（Adrenocorticotropic hornore,AC
TH）産生細胞に作用し、ACTHの分泌を促進する作用があ
り、下垂体、副腎機能等の診断薬及び治療薬としての利
用が期待されている。

り、医薬品として開発する場合、製造してから使用され
るまでの安定性を確保することは、非常に困難なことが

一般にペプチド製剤を安定化する方法としては、グルコ
ース、シュークロース、ラクトース等の糖類やマンニト
ール、ソルビトール、キシリトール等の糖アルコールを
添加して凍結乾燥する方法が知られている。本化合物に
ついても数種の糖類を添加した系について検討を試みた
が、良好な結果は得られなかった。また、分解物の主な
ものは、CRFの分子中に含まれるメチオニンの酸化体で
あるために、抗酸化剤の添加を試みたが良好な効果は得
られなかった。

く偶然にも水溶性高分子および／または塩基性アミノ酸
を添加することにより、CRFの安定性が著しく向上する
ことを見いだし本発明を完成するに至った。

酸、酒石酸、乳酸及びそれらのナトリウム塩、カリウム
塩または水酸化ナトリウムなどが挙げられる。

［実施例］ 以下に実施例及び試験例を挙げて本発明を更に詳細に説

試験例 実施例１〜４並びに比較例１及び２で調製した注射剤を
50℃の恒温槽に保存し、１ヶ月後及び３ヶ月後のCRFの
残存率をHPLCで測定した。結果は表１に示すとおりで、

- 【請求項１】アルギニン、リジンおよびヒスチジンから
選ばれる１以上の塩基性アミノ酸を含有する副腎皮質刺
激ホルモン放出因子含有凍結乾燥製剤。
