軟体動物の遺伝子工学発明の技術分野
本発明は、遺伝子導入軟体動物(transgenic mollusk)、特に貝類に関し、さ
らにアワビを含む軟体動物の遺伝子工学の方法に関する。さらに本発明は、アワ
ビのアクチンの遺伝子配列の単離と特性解析およびその使用に関する。参考文献
発明の背景
海産食物に対する米国の市場は、大きく、かつ成長しつつあり、一人当りの海
産食物の消費量は、過去10年間で23%増加している。この期間に、海産食物
の消費者物価指数は、244%急騰したが、赤肉の価格は、その半分上昇したに
過ぎない。収量維持のレベルで野生の魚類と貝類の個体数を管理しようとする努
力がなされているにもかかわらず、米国は、その漁師から産出される量より一層
多量を消費し資源を涸渇させている。全世界の海洋収穫量は、西暦2000年の
推定需要量の114,000,000メートルトンに対して、僅か90,000
,000メートルトンしか満たさないと予想されている(Harveyの文献、
1990年)。
この世界的な実状は、カリフォルニアのアワビ産業に反映している。1931
年から1968年まで、アワビの商業的漁獲量は、1年間で平均して約900メ
ートルトンであった。1968年以後、漁獲量は、上記レベルの約15%まで減
少した。この減少は、いく
つもの要因が原因であるとされているが、そのうち最も重要な要因は、恐らく、
極東からの需要によってもたらされた魚獲圧力であろう(Tegneyの文献、
1989年)。
漁場を回復させようとする the California Departm
ent of Fish and Game の集中的な努力にもかかわらず、
現在の漁獲量は、その1968年の水準のままである。カリフォルニアのアワビ
漁場の衰退は、メキシコ(Gutzman del Prooの文献、1992
年)、南アフリカ(Tarrの文献、1992年)およびオーストラリア(Pr
inceとShepherdの文献、1992年)における類似の衰退と符合し
ている。
アワビの供給が減少すると、そのper limit valueは、増大し
続ける。1973年〜1988年の間で、ダイバーに支払われる代価は、インフ
レ率の2倍を上回る800%増大した(Tegner他の文献、1992年)。
この需要の増大によって、商業養殖の技術が開発されてきた。これらの努力は、
主に日本とカリフォルニアに集中しているが、カリフォルニアは、地上で集約的
な養殖を行うための海水システムの開発において独特である(Ebertの文献
、1992年)。発明の概要
一面において、本発明には、軟体動物に対して非相同(heterologous)のDN
A配列を有する遺伝子導入(transgenic)軟体動物が含まれる。その非相同DN
A配列は、組み込まれてもよいし、または染色体外であってもよい。一実施態様
で、上記DNA配列には調節配列が隣接し、このことは該DNA配列を軟体動物
内で発現させ
るのに有効である(例えば、RNAまたはタンパク質の発現)。これらの調節配
列は、アワビのような軟体動物またはドロソフィラ・メラノガスター(Drosophi
la melanogaster)の遺伝子をはじめとする他の起源から誘導することができる
。本発明の実施態様には、ドロソフィラ・メラノガスターまたはアワビ由来のア
クチン遺伝子の調節配列の使用が含まれる。好ましい実施態様では、本明細書に
記載のアクチン遺伝子のプロモーターが用いられる。
本発明の一実施態様において、上記調節配列は、ドロソフィラ・メラノガスタ
ーのACT−5プロモーターから得られ、そして上記の非相同DNA配列は、ギ
ンザケ成長ホルモンをコードしている。この構造体は遺伝子導入アワビを産出す
るのに用いると、アワビの成長が促進される。
本発明を実施するのに有用な非相同DNA配列としては、成長増大を促進する
産物(例えば、成長ホルモンまたはインスリン様成長因子など)をコードしてい
るDNA配列がある。その外に、軟体動物の組織の味またはテクスチャに影響す
る産物をコードしているDNA配列がある。
また本発明には、遺伝子導入軟体動物を本発明の方法によって製造するのに有
用な発現ベクターが含まれる。発現ベクターは、典型的に、(i)軟体動物内で
配列を発現させるのに有効な調節要素が隣接している第一DNA配列(この調節
要素は、軟体動物の遺伝子から得られる)、および(ii)二次宿主内でこの発
現ベクターを繁殖させる第二DNA配列を含有している。代表的な第二配列とし
ては、複製開始点と選択可能マーカーを有するDNA配列があり、これら両者は
二次宿主内で機能する。二次宿主としては、細菌、酵母および昆虫細胞がある。
上記発現ベクターは、上記の調節要素お
よびDNAコーディング配列を保有することができる。
他の実施態様で、本発明には、対象のあらゆるコーディング配列を挿入し易く
するのに有用なクローニング部位に隣接したアワビアクチン遺伝子プロモーター
領域を含有する発現ベクターが含まれている。
また本発明には、軟体動物にトランスフェクトする方法が含まれる。この方法
で、対象のDNA配列を有するベクターが提供される。このベクターは、例えば
エレクトロポレーションによって、選択された軟体動物中に導入される。トラン
スフェクトされた軟体動物、すなわち対象のDNAを保有する軟体動物は、上記
DNA配列の存在に基づいて、例えばハイブリッド形成分析または該DNA配列
に伴う特性の発現によって確認される(例えば、リポーター遺伝子の存在)。
上記DNA配列は、成長増大を促進する遺伝子、例えば、成長因子またはイン
スリン様成長因子などをコードすることができる。
導入されるDNAは、組み込まれてもよいし、または染色体外であってもよい
。一実施態様において、DNAの導入は、該ベクターを、軟体動物の受精卵中例
えばアワビの受精卵中にエレクトロポレートすることによって行われる。典型的
には、そのDNA配列には、そのDNA配列を遺伝子導入軟体動物中で発現させ
るのに有効な調節配列が隣接している。
本発明の他の実施態様に、軟体動物内でタンパク質を組換え発現させる方法が
含まれている。この方法では、そのタンパク質の発現が可能な条件下で増殖する
タンパク質をコードする非相同DNA配列を有する遺伝子導入軟体動物が用いら
れる。
本発明には、さらに、軟体動物の成長を促進する方法が含まれ、
この方法によって、成長増大を促進する遺伝子をコードする非相同DNA配列を
有する遺伝子導入軟体動物が産出される(例えば成長ホルモンまたはインスリン
様成長因子など)。次いで、その遺伝子導入軟体動物は、成長促進タンパク質を
許容する条件下で養殖される。
また本発明には、ポリペプチドをコードするDNA配列およびこのDNA配列
に隣接し該ポリペプチドの転写を促進するのに有効なプロモーターを含有する二
重らせんDNAフラグメントも含まれており、この場合、そのプロモーターはア
ワビアクチン遺伝子のプロモーターであり、かつそのDNA配列はアワビアクチ
ン遺伝子ではない。この実施態様で、そのポリペプチドのコーディング配列は、
そのプロモーター配列に対して非相同である。一つの実施態様で、そのDNA配
列は、成長増大を促進する遺伝子、例えば、成長ホルモン(ギンザケ成長ホルモ
ンなど)またはインスリン様成長因子などである。
このような二重らせんDNAフラグメントは、例えば、対象のベクター中に挿
入する発現カセットとして有用である。
別の面において、本発明には、倍数体の軟体動物例えばアワビの産生方法が含
まれる。この方法では、アワビの受精卵をサイトカラシンBで処理して倍数性を
誘発させる。次に、この処理された受精卵を、それを倍数体生物に発育させる条
件下で養殖する。一実施態様で、サイトカラシンBで処理すると、極体の生成が
70%以上阻害されるようになる。このような倍数体生物は、成長が促進された
特性を有しているので有用である。
本発明のこれらのおよび他の目的と特徴は、以下の発明の詳細な説明を添付図
面とともに読めば、より一層充分に分かるであろう。図面の簡単な説明

相同DNAとは、組換え法によって宿主生物に導入されたのではないDNAを
意味する
非相同DNAとは、宿主生物中にトランスフェクトされたDNAを意味する。
典型的には、非相同DNAとは、トランスフェクトまたは形質転換された細胞の
ゲノムDNA(例えば、CATおよびβ−ガラクトシダーゼの遺伝子配列)から
は、本来得られないDNAを意味する。しかし、組換え法によって生物中に導入
されたDNAは、非相同DNAと呼ぶ(例えば、アワビ成長ホルモン遺伝子を保
有する発現ベクターのアワビ内への導入など)。
染色体外DNAとは、宿主細胞内に導入されたプラスミドDNAを意味し、そ
のプラスミドDNAは、その宿主の細胞のゲノムDNA中には組み込まれていな
い。
用語「軟体動物」は、本明細書で用いる場合、ビバルビア(Bivalvia)綱(例
えば、カキ類)、ガストロポーダ(Gastropoda)綱(例えば、アワビ)、および
セファロポーダ(Cephalopoda)綱(例えば、イカ)を含めて、モルスカ(Mo
llusca)門の構成員を意味する。
II.遺伝子導入貝類の産出に有用なベクターの開発
A.外因性DNAの導入
遺伝子導入軟体動物を産出する有効な方法を開発するには、これら生物体の何
らかの特異性を考慮する必要がある。本発明の目的のための代表的な軟体動物は
、アワビである。
アワビは、「ブロードキャスト・スポーナー(broadcast spawners)」として
知られている海の無脊椎動物のあいまいなグループ(loose group)に属してい
る。このような生物の場合、受精は体外受精であり、多数の精子と卵子が可妊成
体から放出されて行われる。大きな成体の雌(7インチ)は、10,000,0
00個のオーダーの卵を産出し、ふ化の条件下で受精率は95%以上である。
発育は、迅速かつ同調して進行する。すなわち、減数分裂I(第一極体の出現
)が20分間で完了し、減数分裂II(第二極体の出現)が30分間で完了し、
次いで第一卵割が2時間で完了する。24時間で原腸形成が完了し、次いで面盤
すなわち泳ぐための簡単な動物システムが発生した。
この時点で、卵子がふ化して、ベリジャー幼生を放出する。次の24時間中に
、事実上全ての成体組織の一次分化が起こり、トロコフォア幼生と呼ばれるもの
になる。トロコフォアの発育は、5〜7日間続き、その期間にセツルメント(se
ttlement)が起こる。
セツルメントは、動物が固体の基板に結合することによって行われ、その後、
動物は、その泳ぐ器官を失い、成体の貝殻を発生し、次いでグレージング(graz
ing)を始める。理由は、充分理解されていないが、セツルメントは、死亡率が
高く、ふ化の条件下でも僅かコンピテントベリジャー幼生の2〜5%しかセツル
メントに成功しない。このことは、この生物の高い多産性のため、ふ化を大きく
阻害しないが、効率的な遺伝子導入技術の開発に対する問題点になっている。
外因性DNAを生殖細胞系列に組み込む機会を改善するため、外因性DNAを
第一分裂分割の前に導入する。エレクトロポレーションでDNAを導入すると、
第一分裂分割の比較的短い時間枠(約2
時間)内で多数の卵子を処理することができる。エレクトロポレーションによる
DNAの導入が本発明の好ましい実施態様であるが、外因性DNAを導入する別
の方法、例えばマイクロインジェクション法なども同様に使用できる。
アワビのトランスフェクションを行う場合にさらに考慮すべきことは、動物界
におけるその分類学的位置、および発現ベクター内で使用するのに適切なプロモ
ーターを選択する際の上記位置の影響である。「古典的な」脊椎動物の発現ベク
ターは、通常いくつもの組織内で高いレベルで発現されるプロモーターを使用し
ている。これらのプロモーターのなかで最も普及しているのは、レトロウイルス
類のサイトメガロウイルス類(CMV)およびラウス肉腫ウイルス(RSV)の
長末端反復(LTR)由来のプロモーターである。
しかし、脊椎動物と軟体動物を分ける進化上の距離があるため、脊椎動物のプ
ロモーターは、軟体動物の組織内で発現を行わないようである。本発明を裏付け
るために実施した実験では、脊椎動物より軟体動物に一層密接に関連している生
物のディー・メラノガスターのプロモーター(例えば、ACT5−C、Koel
leの文献、1993年)を利用した。他のディー・メラノガスタープロモータ
ー/発現ベクターにも、本発明を実施するのに有用なものがある(例えば、Ri
o他の文献、1985年。Davies他の文献、1986年。Schwein
fest他の文献、1988年)。
代表的なプラスミドpMK18については、実施例1で述べる。プラスミドp
MK18は、ACT5−Cプロモーターのβ−アクチンプロモーターを含有して
いる。このプロモーターは、遺伝子が大部分の成体組織内ではもとより、発育の
全体を通じて発現される(Fryberg他の文献、1983年)ので、選択し
た。上記プラ
スミドは、大腸菌のβ−ガラクトシダーゼ遺伝子及びubx遺伝子の5´非翻訳
領域からなる「発現カセット」を含有している。
本発明を裏付けるのに実施した実験では、エレクトロポレーションによってア
ワビの受精卵にトランスフェクトするのにプラスミドpMK18を使用した。エ
レクトロポレーション条件の初期の最適化は、ドットブロットハイブリッド形成
法を用いて、受精卵中への外因性DNAの導入を検出して行った(実施例2)。
DNAは、単一の卵子(single egg)または幼生から抽出し、次にニ
トロセルロースに固定し、次いで適当な放射能標識をつけたプラスミドDNAと
ハイブリッドを形成させた。
ドットブロットは、二つの時点で行った。すなわち、(i)2〜4細胞の段階
(two-to-four cell stage)…DNAの取り込みを監視するため、および(ii
)ふ化後直ちに…DNAの保持を監視するため、行った。エレクトロポレーショ
ン条件の最適化を行った後の典型的なドットプロットを図1に示す(実施例3)
。この図で、全エレクトロポレートされた卵子の約80〜100%において、D
NAが取り込まれ、保持されていることが分かる。
遺伝子導入アワビ内での外因性DNAの発現について検査するため、50個づ
つの幼生のプールを、幼生の発育期間中の各種の時点で収集し、β−ガラクトシ
ダーゼの活性について検定した(実施例4)。これら検定の結果を図4に示す。
これらのデータは、pMK18(β−アクチンプロモーター)を使用するとβ−
ガラクトシダーゼが発現することを示している。
トロコフォア幼生(1日目)で活性が観察され、そしてこの活性は、ベリジャ
ー幼生(2日目)で最高に到達する。ドットブロット法によってプラスミドDN
Aが幼生期間全体を通じて保持されてい
ることが明らかになっているので、酵素活性の衰退は、プラスミド配列の喪失が
原因ではない。β−ガラクトシダーゼ活性のピークは、明らかに、トロコフォア
からベリジャーへ変異する間、すなわち細胞分裂が最大の時点で起こる。β−ア
クチンのレベルは、細胞分裂と正の相関関係があるので、ドロソフィラのアクチ
ンプロモーターが発育上正しい具合に作用していると考えられている。
このデータは、また、脊椎動物のレトロウイルスプロモータCMVが活性でな
く見えることを実証している。類似の結果がRSVプロモーターの場合に得られ
た。
プラスミドDNAが保持され、セツルメントの後に発現されたかどうかを検査
するため、pMK18DNAを用いて約6000個の卵子のエレクトロポレーシ
ョンを行った結果、約200個の幼若体を得た。これらの幼若体を3箇月齢、6
箇月齢および9箇月齢でサンプリングを行った(実施例3)。3箇月齢の幼若体
から得た典型的なドットブロットを図2に示す。
検査した9匹の動物のうち、すべてがDNAを保持し、これら動物のうち、6
匹が有意なレベルのβ−ガラクトシダーゼの発現を示した(表1、本質的に実施
例4に記載されているように検定した)。
6箇月齢と9箇月齢の幼生アワビから類似の試験結果が得られた。プラスミド
DNAは、幼若体に存続しているが、安定した遺伝子導入系を樹立するには、ゲ
ノムDNA中に組み込まれていなければならない。未切断のゲノムDNAをDN
Aハイブリッド形成分析に付した結果、そのプラスミド配列は、高分子量のDN
Aであることが明らかになった。このような高分子量は、鎖状体形成または鎖状
体形成と組み込みが原因であろう。
これらの可能性を識別するために、ゲノムDNAをHindIIIで切断した
。この制限酵素は、EcoRIで線状にしてエレクトロポレーションを行うのに
用いたプラスミドを非対称的に切断する(実施例3、図3A)。そのDNAを、
pMK18の200bpフラグメントとハイブリッドを形成させて分析した(図
3A)。
上記ハイブリッド形成分析の結果は、次のとおりであった。可能性のある3種
の鎖状体のうちの2種に対応する強力な8kbのシグナルがすべてのゲノムDN
A試料中に観察された。第三の種類の鎖状体に対応する弱い約16kbのシグナ
ルがいくつかのブロット中に観察された。これらの試験結果は、大部分の鎖状体
が、魚について観察されているような頭−尾マルチマーであることを示唆してい
る(Stuart他の文献、1988年。ChongとVielkindの文献
、1989年)。
結合フラグメント(junction fragment)が、その時点の約70%で観察され
たが、このことは非常に高度の組み込みを示唆している。
組み込まれたDNAと組み込まれていないDNAの両者を示すオートラジオグラ
ムを図3Bに示す(実施例3)。
ACT5−Cプロモーターは、幼生のアワビと幼若アワビの両者で機能するが
、ドロソフィラS2細胞内でのその発現に比べて僅かに約5〜10%の効率で機
能するにすぎない。より効率的な軟体動物発現ベクターの開発について以下に述
べる。
ACT5−Cプロモーターの調節下でギンザケ成長ホルモン(csGH)遺伝
子(Gonzalez−Villansenors他)を有する遺伝子導入アワ
ビも構築した。csGH遺伝子の配列は、配列番号:1で示す。ACT5−Cの
制御下のcsGH遺伝子を含有するベクターは、pLH2と命名した。対照の(
野生型)アワビに対する、pLH2ベクターを保有する遺伝子導入アワビの重量
分布を測定した。図6は、このような分析の一つの結果を示す。図6のデータは
、遺伝子導入アワビの方が非遺伝子導入の対照のアワビより重量分布が広くかつ
平均重量が高いことを示している。これら試験結果は、成長特性が増大した遺伝
子導入軟体動物を産出する効力を実証している。
ジノゲン(gynogen)と三倍体の産生は、倍数性、特に三倍体と四倍体を化学
的に誘発するすることによって達成することができる。化学的誘発方法は、該生
物の生活環の特定の相、例えば減数分裂Iまたは減数分裂IIにおいてサイトカ
ラシンBで処理することによって実施できる。この方法では、アワビの受精卵が
提供される。その受精卵は、典型的には二倍体卵であるが、四倍体のような異な
る倍数性であってもよい。
次に、その受精卵を、倍数性を誘発するのに有効な濃度のサイトカラシンBで
処理する。この濃度は、処理される卵子のタイプに依
存している。有効な濃度は、実験で決定することができる。アワビの場合、サイ
トカラシンBの好ましい濃度の範囲は、約0.1mg/1〜1mg/lである。
処理された卵子は、次に倍数体の生物に発育させられる。受精卵が二倍体の場
合、得られる倍数体生物は一般に三倍体かまたは四倍体である。
サイトカラシンBによる処理は、典型的には、減数分裂I、減数分裂II、ま
たは減数分裂IとIIの両者を阻害するために行われる。好ましい実施態様では
、このような処理を行うと極体の生成が70%以上阻害される。また、このよう
な処理は、倍数体生物が典型的に染色体の正常補体(normal complement)を有
する同じ生物(通常二倍体)に比べて成長特性が増大しているので、アワビの成
長を促進する方法を提供する。
ジノゲンは、一世代内で自殖(selfing)を行うことができるので、一層古典
的な繁殖法を促進するのみならず、遺伝子導入体を迅速にホモ接合(homogygose
)する機構を提供することができる。例えば、ジノゲンは、F1に使用して導入
遺伝子(transgenes)をホモ接合し、次に三倍体を産生することによってF2が
不稔となる。
さらに、三倍体(ジノゲンを産生する際の1成分を含有している)は、不稔シ
ードストックを産生することができるので、遺伝子工学で処理された生物に対す
る環境上の懸念を緩和できる。
TII.貝類ベクターの開発
本発明は、また「全アワビ」または「全軟体動物」の発現プラスミドの開発に
も関する。誘導プロモーター類、メタロチオネインおよび熱ショックなどのプロ
モーター(すでに魚類に使われているよ
うに)は、外部刺激によって誘導することができる。しかし、このような誘導プ
ロモーター類は、大規模商業生産に対して現実的に適合できない。
アワビの増殖は、海水が連続的に流動するタンク内で起こる。工業生産が要求
する規模で水温または化学組成を調節しようとすることは、コスト面で非効率的
で環境面でも賢明なことではない。
A.アワビβ−アクチンのプロモーターの単離
本発明を裏付けるために実施した実験は、β−アクチンプロモーターを使用し
てアワビの発現ベクターを構築することに関する実験である。このプロモーター
は、二つのことを考慮して選択した。第一に、β−アクチンは、いくつもの成体
組織内で高レベルで発現される(Fryberg他の文献、1983年)。第二
に、発現に関与する要素の組合せを変えてベクターを構築することによって、β
−アクチンプロモーターの活性を調節することができる(Liu他の文献、19
90年a。Liu他の文献、1990年c)。この特性は、生物の応答が循環ホ
ルモンのレベルに依存している場合、成長ホルモンのようないくつかのタンパク
質を発現させるのに重要である。成長ホルモン(GH)の場合、生物の成長は、
ある最適濃度まではホルモンの濃度の比例しているが、最適濃度をこえると阻害
作用が全くないかまたはごく僅かとなる(Agellon他の文献、1988年
。Moriyamaの文献、1992年)。
アクチン遺伝子類は、多重遺伝子族である(Vanderkerckhove
およびWeberの文献、1978年a、1978年b)。これらの遺伝子は、
高度に保存されているので、β−アクチンをα−アクチンまたはγ−アクチンか
ら識別するプローブまたは
プライマーを設計することは不可能である。これとは対照的に、3´非翻訳領域
は、広い相同性(extensive homology)を示さない(Fryberg他の文献、
1983年。Liu他の文献、1990年c)ので、特定のアクチンDNA配列
を同定するのに使用できる。
アワビのβ−アクチン遺伝子を単離するため、まずその遺伝子のコーディング
領域の一部をポリメラーゼ連鎖反応増幅法で単離した(実施例5A)。非脊椎動
物のアクチンに対する一組の縮重プライマーを用いて、アワビβ−アクチン遺伝
子のコーディング領域内の約400bpのフラグメントを単離した(実施例5A
、図5A、5Bおよび8A)。
約400bpの上記配列から、アワビβ−アクチン配列特異的プライマーを合
成する。これらのプライマーを用い、該遺伝子の3´と5´の末端の両者を、c
DNA末端のPCR迅速増幅(PCR−RACE)反応(Frohmanの文献
、1988年、1990年)で単離する。
5´と3´の末端を単離した後、配列を決定する。該遺伝子の5´と3´の非
翻訳末端に対して特異的なオリゴヌクレオチドプローブを合成する。これらのオ
リゴヌクレオチドは、λ−gt10(Stratagene社、米国カリフォル
ニア州ラ・ホーヤ所在)中に保有されている、アワビゲノムDNAクローンに対
するハイブリッド形成プローブとして用いられる。積極的にハイブリッドを形成
するクローンを同定し、次いで、制限エンドヌクレアーゼ開裂法によって地図を
作成する。開裂フラグメントを該オリゴヌクレオチドとハイブリッドを形成させ
て、アクチン遺伝子とそのプロモーターおよび3´と5´の非翻訳領域を含有す
る適当な大きさの制限フラグメントを確認する。
上記のアクチン遺伝子含有制限フラグメントを、標準の技法(Sambroo
k他の文献、1989年)を用いて、大きさを選択したライブラリーを構築する
ことによって、クローン化する。次いで、全アクチン挿入断片のDNA配列を決
定する。
5´非翻訳配列を含めてアワビアクチン遺伝子の5´領域を含有する配列を決
定した(実施例5B)。これらの配列を図9に示す(配列番号:5)。図10は
、配列を決定されたアクチン遺伝子の領域の概略図を示す。過渡的発現の検定結
果(実施例5B、表2)は、アクチン遺伝子プロモーターが機能していることを
実証している。
このアワビβ−アクチン遺伝子は、発現ベクターを構築するのに使用できる(
実施例5c)。一実施態様において、アワビアクチンプロモーター配列は、例え
ば成長ホルモン遺伝子が、該プロモーターの制御下にある場合、アワビを形質転
換するするのに有用なベクターを構築するのに使用できる。
他の適切な軟体動物遺伝子由来のプロモーター(例えば、ヘキソキナーゼ、ホ
スホグリセリン酸キナーゼ、ピルビン酸キナーゼなど)は、β−アクチンについ
て本明細書に記載した手続にほぼ従って単離することができる。
B.発現ベクターの構築
アクチンプロモーターに特異的なモチーフは、他のアクチンプロモーターと比
較することによって同定する。類似の比較は、他の単離されたアワビ遺伝子につ
いて行うことができる。プロモーター領域および他の5´と3´の機能領域は、
欠失地図作成によって同定する(実施例5B)。欠失体の配列を決定し、次いで
全アクチン遺伝子クローンと比べてエンドポイントを決定した。プロモーターお
よび5´と3´の非翻訳領域を各種の比率で含有するプラスミドが構築された。
ポリリンカー部位が、迅速クローン化を行うため、典型的に5´と3´の非翻
訳遺伝子の調節領域の間に挿入される。ポリリンカーは、通常、例えばpUC1
8のポリリンカー(Clontech社、米国カリフォルニア州パロアルト所在
)の場合のように、いくつかの独特の酵素エンドヌクレアーゼ部位を持っている
。受精卵への発現ベクターDNAの取り込みおよびDNAの組み込みは、実施例
2と3に記載したようにして測定する。
アクチン遺伝子発現カセット(すなわち、ポリリンカーに隣接する5´/3´
アクチン配列)は、典型的には、大量のベクターDNAの操作と単離を容易に行
うためにシャトルベクター中にクローン化される。このようなシャトルベクター
は、いくつか市販されている(Clontech社、米国カリフォルニア州ラ・
ホーヤ所在)。シャトルベクターは、典型的に、複製開始点(選択された宿主中
でプラスミドが増殖するための複製開始点、例えばその複製開始点はcolE1
タイプでもよい)および宿主細胞内で選択できる選択可能マーカー遺伝子(例え
ば、細菌の場合、β−ラクタマーゼ遺伝子(AmpR))を持っている。
C.発現のレベルの測定
本発明の発現ベクターを用いたときの遺伝子発現のレベルは、遺伝子導入アワ
ビの産出および生きている幼生と幼若体中での非相同DNAの発現の特性解析に
よって測定した。β−ガラクトシダーゼ、クロラムフェニコールアセチルトラン
スフェラーゼ(RAT)およびルシフェラーゼ(de Wet他の文献、198
7年。Gibb
s他の文献、1991年)の遺伝子をはじめとする、いくつかのリポーター遺伝
子が、遺伝子発現を定量するのに有用である。典型的に、リポーター遺伝子は、
発現カセット(例えば、アクチン遺伝子発現カセット)中にインフレーム融合遺
伝子(in-frame fusion)として挿入される。
遺伝子発現のレベルは、選択されたリポーター遺伝子を検出する標準の酵素検
定法によって定量される(例えば、CAT酵素検定システム、Promega社
、米国ウィスコンシン州マディソン所在)。
さらに、発現レベルの定量は、RNAハイブリッド形成分析法(Ausube
l他の文献、Sambrook他の文献)を用い、内因性遺伝子RNA(例えば
、β−アクチンmRNA)のレベルをリポーター遺伝子RNA(例えば、ルシフ
ェラーゼmRNA)のレベルと比較して実施する。
IV.成長に影響する貝類遺伝子の単離
A.成長ホルモンの遺伝子
成長ホルモン(GH)遺伝子を挿入すると、次のようないくつかの商業上重要
な水産物の成長速度が増大する結果が得られている。例えば、ドジョウ(ミスグ
ルヌス・アンギリカウダトゥス=Misgurnus anguillicau
datus)(Zhu他の文献、1986年)、コイ(シプリヌス・カルピオ=
Cyprinus carpio)(Zhang他の文献、1990年)、大西
洋サケ(サルモ・サラール(Salmo salar)(Rokones他の文
献、1989年。Du他の文献、1992年)、ニジマス(オノリンクス・ミデ
ィス=Onorhynchus mydiss)(Guyomard他の文献、
1989年)、カワカマス
(エソックス・ルシウス=Esox lucius)およびウォールアイ(Wa
lleye)(スティゾステジオン・ビトレウム=Stizostedion
vitreum)(Moav他の文献、1992年)がある。遺伝子導入魚類は
、典型的に、その遺伝子を導入されていない同胞より1〜3倍の大きさである。
商業的に重要な貝類のアワビとカキは、外因性GHに応答する(Morse他
の文献、1981年、1984年)。変態中のアワビの幼生をヒトGHとともに
インキュベートすると、4日後に貝殻の長さが約10%大きくなった。最近、M
oriyama(1992年の文献)は、幼若アワビに組換えサケホルモンを注
入するかまたは幼生アワビを該ホルモンに浸漬すると、重量が2〜3倍増大する
ことを示した。
イースターンカキ(eastern oyster)(クラソストレア・バージニカ=Cra
ssostrea virginica)について類似の試験結果を、Payn
terとChenが報告している(1991年の文献)。これらの貝類に対する
GHの正作用は、おそらく、原始的軟体動物中に複合ホルモン系が存在している
ことを反映している(JooseとGaraertsの文献、1983年)。こ
の解釈と一致して、Moriyamaと彼の同僚たちは、アワビから成長ホルモ
ン様ペプチドを単離し、部分的に精製しており、そしてこのペプチドも成長を刺
激する(Moriyama他の文献、1989年、1992年)。
本発明は、軟体動物の成長に影響する遺伝子を導入できる組換えベクターと方
法を示す。本発明の方法によって、アワビのGH遺伝子は、例えば、生成するタ
ンパク質発現産物がアクチン/GH融合タンパク質であるように、アクチンカセ
ット発現ベクター中にクロ
ーン化される。あるいは、アクチン調節配列を用いGH調節配列を置換して、G
H遺伝子を、アクチン遺伝子調節配列の転写と翻訳の制御下に置く。
さらに、本発明の方法を用いて、正常なGH遺伝子調節配列を欠失させ、次に
この配列を高レベルの発現を行わせる調節配列で置換することによって(すなわ
ち遺伝子置換)、GH遺伝子の内生コピーを修飾することができる。
B.他の可能性がある有用な遺伝子
遺伝子導入アワビの成長速度と最終の大きさを増大するためにアワビ成長ホル
モン遺伝子を使用するのに加えて、インスリン様成長因子(IGF1)遺伝子も
GHに対するその正の調節によって成長を促進するのに用いることができる(S
araとHallの文献、1990年)。IGF1は、最近カキ類から単離され
ている。各種の生物体から由来のIGF1様の配列は、アワビ同族体のIGF1
遺伝子の単離に用いる縮重プライマーを産出するのに使用できる(アワビβ−ア
クチン遺伝子の単離について本明細書で述べたのと同様)。
V.用途
本発明は、モルスカ(mollusca)門のメンバーの形質転換法を含んで
いる。この門には、ガストロポーダ綱が含まれ、この綱にアワビ類が含まれてい
る。
A.ベクターと変異品種
本発明は、軟体動物を形質転換するのに適切なベクターを提供す
るものである。このようなベクターは、個々にまたはキットで販売されており、
本発明のトランスフェクション法と形質転換法に用いることができる。また、キ
ットには、形質転換を行うのに有用な緩衝液が入っていることもある。
本発明のベクターと方法は、遺伝子導入アワビなどの遺伝子導入水産生物を産
出するのに使用できる。本発明のベクターと方法は、非相同DNAを導入するか
、あるいは組換え遺伝子法で内生遺伝子を修飾するのに使用できる。このような
遺伝子導入生物は、操作して先に述べたように成長速度を高めることができる。
成長を高めることに加えて、疾病抵抗性、貝殻および/または貝の肉の着色、肉
のテクスチャおよび味などの他の特性を有する品種を作ることができる。この技
術は、アワビのみならず、他の商業上重要な軟体動物にまで広げることができる
。
B.遺伝分析
本発明のトランスフェクション法は、一層古典的な遺伝的方法を補完する。本
明細書に記載の遺伝子発現の効率によって、成長と生物発生の過程にとって重要
な遺伝子発現について詳細な研究を行うことが可能になる。さらに、本発明の遺
伝子導入生物(transformed organism)は、かような過程を妨害するかまたは促
進するのに有効な化合物を(典型的には、リポーター遺伝子、例えばGHプロモ
ーターの制御下にあるリポーター遺伝子の発現に対するかような化合物の作用を
利用することによって)選別する手段を提供するものである。本発明の遺伝子導
入軟体動物も、トランスフェクションで導入されたDNAがコードする非相同タ
ンパク質を発現する伝達体(vehicles)である。
また、本発明のベクターと方法は、遺伝子置換法および遺伝子ノックアウト法
を含めて、軟体動物の組換え遺伝子操作を行う手段を提供するものである。
本発明を実施する好ましい実施態様、使用および方法を詳細に説明してきたが
、本明細書に示したような他の各種の使用、組合せおよび実施方法は、本発明の
意図内にあることが分かるであろう。
材料と方法
大腸菌DNAのポリメラーゼI(クレノウフラグメント)は、Boehrin
ger Mannheim Biochemicals社(BMB)(米国イン
ディアナ州インディアナポリス所在)から入手した。T4DNAリガーゼとT4
DNAポリメラーゼは、New England Biolabs社(米国マサ
チューセッツ州ビバリー所在)から入手し、ニトロセルロースフィルターは、S
chleicher and Schuell(米国ニューハンプシャー州キー
ン所在)から入手した。
合成のオリゴヌクレオチドリンカーとプライマーは、市販の自動オリゴヌクレ
オチド合成器を用いて製造した。あるいは、注文設計の合成オリゴヌクレオチド
は、例えばSynthetic Genetics社(米国カリフォルニア州サ
ンディエゴ所在)から購入できる。cDNA合成キットおよびランダムプライミ
ング標識付けキットは、Boehringer Mannheim Bioch
emical社(BMB、米国インディアナ州インディアナポリス所在)から入
手した。
ペプチド類をコードするオリゴヌクレオチド配列は、オリゴヌクレオチド合成
の標準方法で直接合成するか、またはコーディング配
列が大きい場合そのコーディング配列に対応する多重オリゴヌクレオチドフラグ
メントのタンデムアレーを含む一連のクローン化ステップで合成することができ
る(Crea、Yoshio他、Eaton他)。オリゴヌクレオチドのコーデ
ィング配列は、標準の組換え法(Maniatis他、Ausubel他)によ
って発現させることができる。
標準の分子生物学の方法とクローン化法は、Ausubel他、Sambro
ok他、およびManiatis他の文献に既に記載されているのと本質的に同
様に実施した。
ポリメラーゼ連鎖反応は、本質的にメーカーの支持どおりに行った。ポリメラ
ーゼ連鎖反応法は、Kawasakiの文献、Mullisの文献、およびMu
llis他の文献に記載されている。
受精卵は、Pacific Mariculture Inc.社(米国カリ
フォルニア州95060サンタクルズ、コーストロード5515所在)から入手
した。受精卵の他の出所は、次のとおりである。The Avalone Fa
rm Inc.社(米国カリフォルニア州93430、カーヨコス、私書箱13
6)、およびAb Lab社(米国カリフォルニア州93043−4328、ポ
ート・ヒューニーム、センター・ドライブ560)である。
実施例1
プラスミド類の構築
プラスミドpMK18とpLH2は、pMK26(図7。Koelle他の文
献、1991年。pMK26=pAct/Sv40/BS)から構築した。プラ
スミドpMK26は、ドロソフィラメラノガスターACT5−Cプロモーター、
β−アクチンプロモーター
を含有している。このプロモーターは、発育中全体を通じておよび殆どの成体組
織中において発現される(Fryberg他の文献、1983年)。両者ベクタ
ーは、下記のように修飾されたpMK26から構築した。すなわち、pMK26
をEcoRIで部分的に消化し、pMK26内のKpnI部位の隣のEcoRI
部位(図7)を、クレノウとdNTPを用いて満たした(Ausubel他、M
aniatis他)。獲られたプラスミドをブラント末端連結法によって再び環
状にし、次いで単一のEcoRI部位を有する修飾pMK26を制限地図作成に
よって確認した。
cPbxd6.2の4.8β−ガラクトシダーゼubxカセット(Irvin
e他の文献、1991年)を、そのフラグメントのブラント末端連結によって修
飾pMK26のSmaI部位に挿入することによって、pMK18を構築した。
エレクトロポレーションを行う場合、プラスミドは、ユニークEcoRI部位で
線状にした。
pLH2を構築するため、csGH遺伝子ならびにすべての5´−非翻訳リー
ダーおよび373BPの3´−非翻訳領域を含有する1070KBのHindI
IIフラグメントをpRSVCSGHから得た(Gonzalez−Villa
nsenor他)。上記HindIIIフラグメントを、修飾pMK26ポリリ
ンカーのユニークHindIII部位中に連結した。このプラスミドは、エレク
トロポレーションを行う場合は、EcoRIによって線状にした。csGHのD
NA配列は配列番号:1として示す。csGHの全翻訳配列は、配列番号:2と
して示す。
pLUCベクターは、以下のようにして生成させた。ルシフェラーゼ遺伝子と
SV40調節配列を含有する、pXP2の2.0kbXkoI/StyIフラグ
メント(Nordeen,Biote
chniques,6巻、454〜457貞、1988年)を単離した。このフ
ラグメントを、xhoI/StyIで消化したpBLCAT6中にクローン化し
て(Boshart他、Gene)110巻、129〜130頁、1992年)
、プロモーターなしのルシフェラーゼ含有ベクターを得た。
上記のpCMVtkLUCベクターは、CMVエンハンサーを有するHSVチ
ミジンキナーゼプロモーターの制御下にあるルシフェラーゼ遺伝子を含有してい
た。
実施例2
アワビへのDNAの導入
DNAを導入するため、4000から6000卵/200μlの濃度のアワビ
受精卵200μlを、Baekon2000エレクトロポレーターキュベットの
各々に入れた。各キュベットには、2mg/mlの濃度の線状化プラスミドDN
A100μlと人工海水400μlが入っていた。
典型的な人工海水の構成成分は、次のとおりである。すなわち、24.72g
/lのNaCl、0.67g/lのKCl,1.36g/lのCaCl2・2(
H2O)、4.66g/lのMgCl2・4(H2O)、6.29g/lのMgS
O4・7(H2O)、0.18g/lの炭酸ナトリウム、pH7.8である。
エレクトロポレーションの条件は、次の通りであった。10KV、6サイクル
、パルス時間:160μsおよびバースト時間:1.6秒である。エレクトロポ
レーションを行った後、これらの卵をペニシリンとストレプトマイシンを各々1
00μg/lずつ含有する海水1l(pen/strep、Life Tech
nologie
s社、米国メリーランド州ガイサーズバーグ所在)中に入れた。その卵/幼生を
通気しながら14℃で培養した。海水/pen/strepは毎日取り替えた。
実施例3
遺伝子導入アワビのゲノムDNAハイブリッド形成分析
A.DNAの取り込みと保持
上記ベクターでトランスフェクトしたアワビ卵を、トランスフェクトしてい
るDNAの存在について評価した。受精してから2時間後(第一卵割期(first
cleavage division))と24時間後(トロコフォア幼生)に、エレクトロポレ
ーションされた卵からDNAを抽出した(Hahnの文献、1989年)。上記
の最初の時点では、DNAの取り込みを観察し、第二の時点では、DNAの保持
を観察した。
各時点で、5個の卵または幼生のプールを、「エッペンドルフ」微量遠心単離
管中のTE(Maniatis他)10μl内で、ホモジナイザーを用いて摩砕
した。破片を除いてから、核酸を沈澱させた。再懸濁させたDNA試料を、ドッ
トブロットキャプチャーシステム(dot blot capture system)を用いてニトロ
セルロースに移した(Ausubel他、Schleicher and Sc
huell社)。
このようなドットブロット分析の代表的な結果を図1に示す。図1は、プラス
ミドDNAがエレクトロポレートされたアワビ卵中に取込まれ保持されているこ
とを示す。上記ニトロセルロースフィルターに結合させたDNAを放射能標識を
つけたプラスミドpMK18 DNAとハイブリッドを形成させた(ランダムプ
ライマー、B
oehringer−Mannheim社、米国インディアナ州インディアナポ
リス所在)。図1において、2時間後の試料の最下行の左から2本のスロットは
、エレクトロポレートされていない卵から抽出したDNAである。標準は、図1
のトップパネルに示してあり、左から右へそれぞれ600、300、100、6
0および30ピコグラムのpMK18 DNAである。
このようにして得たデータは、エレクトロポレートされた卵全部の約80〜1
00%にDNAが取込まれ保持されていることを示唆した。
B.トランスフェクトされたDNAの保持
pMK18ベクターでトランスフェクトしたアワビを成長させた。3箇月齢の
幼若アワビからDNAを抽出し、放射標識をつけたプラスミドpMK18 DN
Aを用いドットブロット法で分析した。図2は、遺伝子導入アワビと非遺伝子導
入の対照アワビについてのドットブロットハイブリッド形成データを示す。濃度
標準は(左から右へ)、600、400、100、60、30、10、6および
3ピコグラムのpMK18 DNAである。
図2の結果は、エレクトロポレートされたDNAが3箇月齢の幼若アワビ中に
保持されていることを実証している。
C.3箇月齢の遺伝子導入幼若アワビのDNAハイブリッド形成分析
上記3箇月齢の遺伝子導入幼若アワビから単離したDNA試料を、アガロース
ゲルに負荷し、サイズ分画を行い、ニトロセルロースフィルターに移した(Au
subel他、Sambrook他)。こ
れらのフィルターを放射能標識をつけたpMK18とハイブリッドを形成させた
。これらフィルターのオートラジオグラムは、pMK18相同配列が高分子量D
NA中に存在していることを実証した。これら試験結果を図3Bに示す。図3B
において、二つのレーンのDNA試料は、二つの異なる動物から単離された。左
側パネルのDNAが組み込まれている。右側のパネルのDNAは組み込まれてい
ない。
pMK18の鎖状体と組込みを識別するため、ゲノムDNA試料を制限エンド
ヌクレアーゼHindIIIで消化した。この制限エンドヌクレアーゼは、エレ
クトロポレーションを行うのに用いた、EcoRIで線状にしたプラスミドを非
対称的に切断する(図3A参照)。ニトロセルロースフィルター上の上記DNA
を、pMK18の8kb HindIII/EcoRIフラグメント由来で20
0bpのポリメラーゼ連鎖反応(Perkin−Elmer/Cetus、米国
コネティカット州ノーウォーク)で増幅したフラグメントとハイブリッドを形成
させた(図3A参照)。
3種の可能性のある鎖状体のうちの2種に対応する強力な8kbのシグナルが
観察された(図3B)。ゲノムDNAの全試料中に、各試料中に鎖状体が存在し
ていることを示す8kbのシグナルが観察された。
上記DNA試料のいくつかには、約16kbのシグナルも観察された。この1
6Kbのシグナルは、第三のタイプの鎖状体に対応するシグナルであった(図3
A)。これらの試験結果は、これら鎖状体の大部分が頭尾マルチマーであること
を示唆している。
遺伝子導入アワビ由来のDNA試料の約70%に結合フラグメントが観察され
た(図3B)例えば、8kb未満の分子量で現れるシ
グナル)。この試験結果は、極めて高度に取り込まれていることを示唆している
。
実施例4
遺伝子導入アワビにおける遺伝子発現の分析
ベクターpMK18は、β−ガラクトシダーゼの発現カセットを持っていた。
このベクターを有する遺伝子導入アワビのβ−ガラクトシダーゼの発現について
、時間の経過とともに検定した。
50個の幼生からなるプールを、幼生の発育期間中、各種の時間間隔をおいて
抽出し、次いでβ−ガラクトシダーゼのレベルを、Koelle他の1991年
の文献にすでに記載されているのと本質的に同様にして、蛍光基質4−メチルウ
ンベリフェリル−β−D−ガラクトシド(Sigma Chemical Co
.社、米国ミズーリ州セントルイス所在)を用いて測定した。
図4は、エレクトロポレートされた幼生内でのβ−ガラクトシダーゼの発現に
ついての代表的な蛍光試験から得たデータを示す。蛍光単位をY軸に示し、時間
(日数)(受精後)をX軸に示す。酵素活性は、任意の蛍光単位で示してある。
図4に示すデータは、プラスミドpMK18については、5個の別個のエレク
トロポレーション試験結果の平均値であり、対照のプラスミドCMVLacにつ
いては、2個の別個の実験結果の平均値である。CMVlacは、β−ガラクト
シダーゼ遺伝子に隣接したサイトメガロウイルス プロモーターを保有するベク
ターである(例えば、「pCMVβ」、Clontech社、米国カリフォルニ
ア州パロアルト所在)。標準誤差は、計算しなかったが、試験間の変動は、5%
未満であった。
このデータは、pMK18(β−アクチンのプロモーター)を用いたときβ−
ガラクトシダーゼが発現されることを示唆している。ドットブロット試験の結果
が、幼生期を通じてプラミドDNAが保持されていることを示しているので、酵
素活性の減少は、プラスミド配列の損失が原因ではない。
脊椎動物のレトロウイルスのプロモーターCMVは、遺伝子導入アワビ内では
活性でないようである。RSVのプロモーターの場合も類似の結果が得られた。
実施例5アワビアクチンの遺伝子の同定と単離および軟体動物発現ベクターの構築
A.アワビアクチン遺伝子の約400塩基対のフラグメントの分離
非脊椎動物アクチンに対する一組の縮重プライマー(ディー.メラノガスター
(5CX、8F、87E)、ビー.モリイ(B.Moryi)(A1、A2、A
3)のアクチン遺伝子に対する「GENBANK」配列に基づいて)を設計した
。これらプライマーの配列を配列番号:3および配列番号:4として示す。これ
らのプライマーは、標的コーディング領域内で約400bpのフラグメントを増
幅する(図5A)。この約400bpのフラグメントの単離を示す概略図を図3
Aに示す。
RNAは、アワビの卵巣組織から単離した(L.GrossmanおよびK.
Moldave編集「Methods in Enzymology」、12巻
、パートB、120−129貞のR.A.Cox Iの報告(Academic
Press社、米国フロリ
ダ州オーランド所在)、1988年発行。Tel−Test Inc.社(米国
テキサス州フレンズウッド所在)から入手できる「RNAZOL」キット)。
10μlの逆転写酵素(Invitrogen社、米国カリフォルニア州サン
ディエゴ所在)を用い、最終容積20μlで、全卵巣RNA5μgからcDNA
を合成した。各cDNA試料1μlを用い、20μMのプライマーを加えて10
0μlポリメラーゼ連鎖反応(PCR)を行った。ポリメラーゼ連鎖反応の条件
とプロトコルは、市販のキツト(Perkin−Elmer社、米国コネティカ
ット州ノーウォーク所在)に見られるのとほぼ同じにした。
増幅産物を、電気泳動を利用し大きさに基づいて分割した。増幅後、各反応産
物混合物の約7%を、1%アガロースゲルの電気泳動によって単離した(FMC
Bioproducts社、米国メイン州ロックポート所在)。これらのゲル
を臭化エチジウムで染色し、紫外線照射で眼に見えるようにした。
このような分析の一つの試験結果を図5Bに示す。図5Bの第1レーンは、縮
重アクチンプライマーのセットと卵巣由来のDNAを利用した増幅で得られた増
幅産物を示す。第2レーンは、縮重アクチンプライマーのセットと全アワビ卵巣
RNAを利用した増幅で得られた増幅産物を示す。約400bpの予想増幅産物
