Abstract
Description
도 2는 본 발명에서 사용될 수 있는 생물 반응기의 개략도이다.
도 3a는 세포 성장 챔버의 중공 섬유 세포 성장 챔버 실시양태의 측면도를 도시한다. 도 3b는 도 3a의 실시양태의 중공 섬유 세포 성장 챔버의 절취된 측면도를 도시한다.
도 4 - 블록 다이아그램 시스템 및 모든 백 연결부를 나타내는 세포 확장 시스템(CES) 유체 회로.
도 5 - 골수로부터 또는 세포 은행으로부터 멀티스템(MultiStem)을 확장시키는 데 사용되는, 카리디안 비씨티(Cardian BCT, 현재는 테루모(Terumo)가 소유함)로부터의 퀀텀 세포 확장 시스템(Quantum Cell Expansion System)의 흐름도.
도 6 - WJS 골수 흡인물을 골수 단핵 세포(BMMNC)의 사전 선발 없이 퀀텀에 접종하고, 동일 공여체 유래의 BMMNC를 보통의 세포 배양 플라스틱 상에 접종하였다. 이를 3가지의 독립적인 공여체에 대하여 행하였다. T75 내의 세포를 3회 이상 계대하고, 세포 수를 계수하여 집단 배가(population doubling; PD)를 계산하였다. 퀀텀 내의 세포를 수확하고, 계수하고, 새로운 생물 반응기 상에 로딩하고, 6일 후 다시 수확하였다. 퀀텀에서의 골수 흡인물로부터 시작되는 멀티스템의 확장 속도는 세포 배양 플라스틱 상에서의 확장과 유의하게 상이한 것은 아니다.

Claims (13)
- a) 세포를 중공 섬유 기재 상에 접종하여 세포를 기재에 유착시키는 단계; b) 기재 상의 유착된 세포를 확장시키는 단계; 및 c) 확장된 세포를 기재로부터 제거하는 단계를 포함하는, 생체외에서 세포를 확장시키는 방법으로서, 상기 세포는 비배아 줄기, 비생식 세포이고, 텔로머라아제를 발현하며, 형질전환되지 않고, 정상 핵형을 갖는 방법.
- 제1항에 있어서, d) 제거된 세포를 동일하거나 상이한 기재 상에 재접종하는 단계; 및 e) 요망되는 수의 확장된 세포에 도달할 때까지 단계 b) - d)를 반복하는 단계를 추가로 포함하는 방법.
- 제2항에 있어서, 제거된 세포를 대상체에게 투여하는 단계를 추가로 포함하는 방법.
- 제1항에 있어서, 중공 섬유 기재가 폐쇄식 연속 관류 생물 반응기 내에 있는 것인 방법.
- 제3항에 있어서, 세포는 약 10-100배 확장되는 것인 방법.
- 제1항 내지 제5항 중 어느 한 항에 있어서, 중공 섬유가 코팅되는 것인 방법.
- 제6항에 있어서, 코팅이 피브로넥틴인 방법.
- 제7항에 있어서, 섬유 중 벽 재료가 PA/PAES/PVP인 방법.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 단계 (a)에서의 세포가 실질적으로 균질한 집단을 포함하는 것인 방법.
- 제1항 내지 제8항 중 어느 한 항에 있어서, 단계 (a)가 골수, 제대 혈액, 제대 매트릭스, 말초 혈액, 태반, 태반 혈액, 근육, 뇌, 신장 및 다른 실질 기관(solid organ)으로부터 선택되는 조직 유래의 세포를 접종하는 것을 포함하는 것인 방법.
- 제1항에 있어서, 비배아 줄기, 비생식 세포가 oct-4, rex-1, rox-1 또는 sox-2 중 하나 이상을 추가로 발현하는 것인 방법.
- 제1항 또는 제11항에 있어서, 비배아 줄기, 비생식 세포가 내배엽, 외배엽 및 중배엽 배아 계통 2가지 이상 중 1가지 이상의 세포 유형으로 분화될 수 있는 것인 방법.
- 제12항에 있어서, 비배아 줄기, 비생식 세포가 내배엽, 외배엽 및 중배엽 배아 계통 각각 중 1가지 이상의 세포 유형으로 분화될 수 있는 것인 방법.
