フィラグリンとは？ わかりやすく解説

フィラグリン（Filaggrin）とは、表皮の顆粒細胞で産生される塩基性タンパク質の1種である^[1]。フィラグリンのことを指して、ヒスチジン・リッチ・プロテインなどと呼ぶ場合もある。ヒトにおいて皮膚のバリア機能に欠かすことのできない角質層を形成するに当たり、フィラグリンはケラチンと共に重要な役割を担っている。

生合成と代謝

フィラグリンは前駆体のプロフィラグリンとして生合成され、角質層が形成される段階で、リン酸プロフィラグリンが脱リン酸化と加水分解を受けて分解し、フィラグリンが作られる。プロフィラグリンは、フィラグリンが10個から12個繋がった巨大なタンパク質である。フィラグリンは、ケラチンと凝集・結合し、ケラチン・パターンを構成するが、最終的にはフィラグリンは角質上層でアミノ酸などの低分子まで分解される。

皮膚関連疾患との関わり

フィラグリンが作られないと角質に異常が発生し、皮膚のバリア機能が低下する事で、外来物質影響を受け易くなり、皮膚炎の原因ともなる事が判っている。

例えば、アトピー性皮膚炎の患者に、フィラグリンの遺伝子異常が多く見つかっており、アトピー性皮膚炎治療の鍵となる物質として、フィラグリンは注目されている。また、尋常性魚鱗癬の患者は、皮膚におけるフィラグリンの発現が極度に低下していることが判明している。
