Abstract

The present invention relates to subcutaneous formulations of anti-CD38 antibodies and their uses. In particular, pharmaceutical formulations are provided comprising about 100-120 mg/mL of daratumumab and about 50-5,000 U/ml hyaluronidase. Further provided are unit dose forms comprising: a) daratumumab in an amount of about 1,200-1,800 mg; b) rHuPH20 in an amount of about 30,000-45,000 U; c) histidine at a concentration of about 5-15 mM; d) sorbitol at a concentration of about 100-300 mM; e) PS-20 at a concentration of about 0.01-0.04% w/v; and f) methionine at a concentration of about 1-2 mg/mL at a pH of about 5.5. Also disclosed is the use of an anti-CD38 antibody and hyaluronidase in the manufacture of a medicament for treating CD38-expressing cancers in a subject, wherein a) the medicament is to be administered subcutaneously to the subject, b) the medicament comprises 1,800 mg of the anti-CD38 antibody that is an IgG1 isotype and has the variable heavy and light chain regions of SEQ ID NO: 4 and 5 (i.e. the variable regions of daratumumab), and c) the medicament has a hyaluronidase activity of about 30,000 U to about 45,000 U.

Claims (5)

What is claimed is:
1. Use of an anti-CD38 antibody and a hyaluronidase in the manufacture of a medicament for the treatment of a CD38-expressing cancer in a subject, wherein: a) the medicament is to be administered subcutaneously to the subject for a time sufficient to treat the cancer; b) the medicament comprises about 1,800 mg of an anti-CD38 antibody, wherein the antibody is of an IgG1 isotype and comprises a heavy chain variable region (VH) of SEQ ID NO: 4 and a light chain variable region (VL) of SEQ ID NO: 5; and c) the medicamenthas a hyaluronidase enzymatic activity of about 30,000 U to about 45,000 U.
2. The use of claim 1, wherein the medicament further comprises a pharmaceutically acceptable carrier.
3. The use of claim 1 or 2, wherein the anti-CD38 antibody comprises a heavy chain of SEQ ID NO: 12 and a light chain of SEQ ID NO: 13.
4. The use of claim 1 or 2, wherein the anti-CD38 antibody is daratumumab.
5. The pharmaceutical composition of any one of claims 1-4, wherein the medicament comprises: a) from about 20 mg/mL to about 160 mg/mL of the anti-CD38 antibody; b) from about 20 mg/mL to about 140 mg/mL of the anti-CD38 antibody; c) from about 20 mg/mL to about
