本発明は、バチルス・セレウス種に属する新規微生物およびそれを用いた脱臭方法に関する。

【従来の技術】
バチルス・セレウス種に属する菌としては、食品腐敗菌や食中毒菌の研究が進んでいるが、有用微生物としての研究は全く行われていないといっても過言ではない。

【発明が解決しようとする課題】
そこで、本発明は、バチルス・セレウス種に属する有用新規微生物、およびそれを用いた脱臭方法を提供することを目的とするものである。

【課題を解決するための手段】
上記の課題は、本発明の下記(1)または(2)の構成により達成される。
(1) 下記の菌学的特徴を有するバチルス・セレウス種に属する新規微生物。
(2) 上記(1)のバチルス・セレウス種に属する新規微生物を発臭源に散布することにより脱臭を行う脱臭方法。

【発明の実施の形態】
本発明によるバチルス・セレウス種に属する新規微生物は、Bacillus cereus CCC-b(寄託番号:FERM P-16176)として独立行政法人産業技術総合研究所特許生物寄託センターに寄託されている。

以下、このBacillus cereus CCC-bの菌学的性質を説明する。

以上の菌学的性質の全体から、本菌は、バチルス属に属し、通性嫌気性であり、卵黄反応:+からバチルス・セレウス種に属する菌であると判定したが、このBacillus cereus CCC-bは、デンプンの分解を行わないという非典型性状を示したので新菌であると判定した。
このBacillus cereus CCC-bは、一般栄養培地であるならいかなる培地でも良好に成育する。

培養は、好ましくは15〜45℃程度で良好に増殖する。

このBacillus cereus CCC-bは、水や液体培地に分散させ分散液の状態で各種用途に用いることができるが、好ましくは微生物群を担体に固定し、取扱を容易にした担体固定化微生物群として用いる。

微生物群を固定する担体としては、微生物の保持容量が大きく、また微生物の活性化が容易であることが好ましい。

担体としては、岩石(例えば、真珠岩、珪藻土)またはその粉砕物、砂利、砂、プラスチックス、セラミックス(例えば、アルミナ、シリカ、天然ゼオライト、合成ゼオライト)、タルク等も好ましく、特に、多孔質セラミックス、多孔質プラスチックスのような連続通気孔を有する多孔質材料が好ましい。例えば、真珠岩の粉砕物を高温高圧で保持し熱処理した後、急激に減圧して多孔質にした微細粉(商品名:パーライト)は、菌類を着生させる性質があるため、好適である。このような多孔質材料において、通気孔の径は、2〜10μm程度であることが好ましい。また、このような担体の性状としては、塊状物、粒体、粉体、微粉末または板状物、針状物等いかなるものでもよいが、好ましくは平均粒径が2mm以下、特に50μm〜1mm程度の粉粒体がよい。なお、このような担体固定化微生物群は、必要に応じて、布やネット等の通水性や通気性のよい容器に収納して使用しても良い。

上記担体としては、これらの他、リボン状やシート状の織布や不織布を用いることができる。

このような担体に微生物群を担持させるには、担体と本発明の微生物群分散液とを混合した後、乾燥すればよいが、担体で直接微生物群を培養してもよい。

担体の微生物群の担持量は、担持させるときの条件によっても異なるが、好ましくは、5個〜200億個/cm3、好ましくは10個〜100億個/cm3である。

このBacillus cereus CCC-bを用いることにより・養豚所、公衆便所等に用いて、脱臭効果が確認された。また、このBacillus cereus CCC-bは、排水中の有機物を有効に分解しえ、更には、リン等の金属の回収機能が確認された。

なお、上記Bacillus cereus CCC-bは、マウスによる経口投与試験によりその安全性が確認された。

【実施例】
次に、本発明の実施例について説明する。
まず、次のような組成の培地を作成した。
培地
ペプトン 5g
肉エキス 5g
Nacl 2.5g
水 500ml(pH7.0)
以上の組成の培地を、1000ml容器に入れ、これにBacillus cereus CCC-bを接種し、よく撹拌した後、静置して、30℃で1日間培養した。
得られた培養液を遠心分離機にかけ、菌体を分離し、これをよく洗浄し、担体に付着させてパウダーとした。担体としては珪藻土を焼いて、粉砕したものを用いた。

このパウダー状のBacillus cereus CCC-bを、1本のままの腐った鰯に振りかけ2時間経過後にその臭いをかいだところ、明かに臭気が減少していた。
参考実験
上記パウダー状のBacillus cereus CCC-bを、計量対象へ接種してのCOD、全窒素量および全リン量の経時変化を計量した。試験方法は、CODはJIS 0102−17、全窒素量はJIS 0102−45.2、そして全リン量はJIS 0102−46.2に従って行った。計量対象としては、海水での牛乳100倍液に、1リットル当たり0.2gの澱粉を加えたものを用いた。そのままのものを比較例とし、これにBacillus cereus CCC-bを接種したものを実施例とした。試験は、20℃に保って行った。また、各量の測定にあたっては、計量対象液を2時間静置し、その上澄み液を試験液とした。
試験の結果を表1に示した。なお、試験は、それぞれサンプルを2つずつ作製し、その平均を測定値とした。

表1から分かるように、Bacillus cereus CCC-bは、水の浄化機能を有し、金属を除去する作用があることが判明した。
このように、Bacillus cereus CCC-bは、上記の実験で確認された脱臭機能、有機物分解機能、金属除去機能を備えた極めて有用な新規微生物である。[0001]

DETAILED DESCRIPTION OF THE INVENTION
A novel microorganism belonging to the species Bacillus cereus according to the present invention is deposited as Bacillus cereus CCC-b (deposit number: FERM P-16176) at the National Institute of Advanced Industrial Science and Technology Patent Organism Depositary.

The obtained culture broth was centrifuged to separate the cells, washed thoroughly, and attached to a carrier to obtain a powder. The carrier used was baked and pulverized diatomaceous earth.

Reference experiment The powdery Bacillus cereus CCC-b was inoculated into the object to be weighed, and the changes over time in COD, total nitrogen amount and total phosphorus amount were measured.
