Description
有機イオウ、ウロン酸、固有粘度、ヒアルロニダーゼ阻害効果、及び旋光度は、日本薬局方1997に従って測定した。
ホルムアミド中のコンドロイチンの10%w/v溶液60Lを、撹拌下に反応器に入れた。この反応器は、ダブルジャケット中を循環する水によって冷却される。5Lのクロロスルホン酸を18.5ml/minの速度で添加した。添加の間、溶液の温度を約25℃に保った。4時間30分後、添加を完了した。得られたコンドロイチンポリサルフェートの分析値を表1に載せた。
ホルミアミド中のコンドロイチン硫酸の10%w/v溶液600mlを、撹拌下に反応器に入れた。この反応器は、ダブルジャケット中を循環する水によって冷却される。50mLのクロロスルホン酸を、18.5ml/minの速度で添加した。添加の間、溶液の温度を約25℃に保った。3分後に添加を完了した。得られたコンドロイチンポリサルフェートの分析値を表1に載せた。
さらに遅いクロロスルホン酸添加速度(0.2ml/min)で例2を繰り返した。添加時間は4時間10分であり、溶液の温度は15℃だった。得られたコンドロイチンポリサルフェートの分析値を表1に載せた。
例1を繰り返したが、クロロスルホン酸の量を増加し(7.5L)、かつ、反応器内の温度を40℃に調節した。得られたコンドロイチンポリサルフェートの分析値を表1に載せた。
例1を繰り返したが、クロロスルホン酸の添加速度を速くし(25ml/min)、かつ、反応器内の温度を40℃に調節した。得られたコンドロイチンポリサルフェートの分析値を表1に載せた。
Claims (5)
- コンドロイチンのホルムアミド溶液に、前記ホルムアミド溶液の温度が40℃より低く且つ15℃より高く保たれる速度でクロロスルホン酸を添加することを特徴とする、0.09〜0.18の固有粘度及び25.8〜37.3%の有機イオウ含有量を有するコンドロイチンポリサルフェートの製造方法
- 前記ホルムアミド溶液の温度が20〜35℃に保たれる、請求項1に記載の製造方法。
- 前記ホルムアミド溶液の温度が20〜30℃に保たれる、請求項1に記載の製造方法。
- 前記ホルムアミド中のコンドロイチン濃度が5〜20%w/vである、請求項1〜3のいずれか一項に記載の製造方法。
- ヒアルロニダーゼ阻害に対する前記得られたコンドロイチンポリサルフェートの活性が、14〜29である、請求項1〜4のいずれか一項に記載の製造方法。
