Description
(1)信号を発生させることを意図した成分又は材料、例えば付着した標識をそのまま残すこと、および
(2)付着しない(すなわち遊離の)成分又は材料をできるだけ完全に除去すること、によって制御される。
(a)容器に付着した被検体および標識を有する表面コートされた容器を提供する工程、
(b)前記容器に付着した前記被検体および標識の少なくとも一部と接触させるのに十分な第1の液面高さまで前記容器中に洗浄液を分配する工程、
(c)付着しない標識を洗浄液中へ除去するために、分配と吸引とが同じノズル又は分配と吸引とが異なるノズルのいずれかを上下に動かして容器中の前記洗浄液の前記液面を上下に揺動させる工程、および
(d)表面コートされた前記容器から、前記洗浄液を除去する工程、
を含む、アナライザーの表面コートされた容器の壁に付着した被検体および標識を洗浄する方法の発明により達成される。
試料を入れたスプレプトアビジンでコートされた容器にビオチン試薬を加えて、ビオチン化抗cTnl抗体と、スプレプトアビジンでコートされた容器と、試料中に存在するcTnlとの間の反応を開始させた。またHRP接合試薬を加えて、HRP接合型抗cTnl抗体と試料中のcTnlとの間の反応を開始させた。次いで試料および試薬を8分間、37℃でインキュベーションした。インキュベーション後に試料を入れた容器を本発明および周知の洗浄方法によって洗浄した。洗浄後、ルミノール誘導体、過酸塩、および置換アセトアニリド電子伝達剤を含有する信号試薬を加えて発光を生じさせ、これを照度計で読取った。この結果を図5のグラフに示す。このグラフが示すように、本発明の洗浄方法を用いた結果は、周知の洗浄方法を用いた結果よりもずっと安定しており、且つ異常値が少なかった。
前述のように信号試薬吸引を用いた8回の洗浄および信号試薬吸引を用いない8回の洗浄を行った。これらの結果を図6のグラフに示す。このグラフが示すように、本発明の揺動洗浄方法および信号試薬吸引を用いた結果は周知の洗浄方法を用いた結果よりもずっと安定しており且つ異常値が少なかった。
信号試薬吸引および揺動洗浄を用いて一連の洗浄を行った。信号試薬吸引又は揺動洗浄を用いない他の一連の洗浄を行った。これらの結果を図7のグラフに示す。このグラフが示すように、本発明の洗浄方法を用いた結果は周知の洗浄方法を用いた結果よりもずっと安定しており且つ異常値が少なかった。
本発明は、また、HBsAg(B型肝炎表面抗原)の分析性能を改善するのに有用である。マウス単クローン性抗HBsAg抗体をコートされた容器、HRPで標識したマウス単クローン性抗HBsAg抗体の複合体、および試料中のHBsAgを反応させた。次いでこの試料を29分間、37℃でインキュベーションした。インキュベーション後、試料を入れた容器を、本発明の揺動の実施形態および周知の洗浄方法によって洗浄した。洗浄後、ルミノール誘導体、過酸塩、および置換アセトアニリド電子伝達剤を含有する信号試薬を加え、続いて信号試薬をインキュベーションして発光を生じさせ、これを照度計で読取った。信号を読取る前に、本発明の表面除去の実施形態を用いて表面の膜を除去した。上記洗浄方法(周知および本発明の両方)を用いていくつかの作業を行い、HBsAgのレベルを求めた。
Claims (8)
- (a)容器に付着した被検体および標識を有する表面コートされた容器を提供する工程、
(b)前記容器に付着した前記被検体および標識の少なくとも一部と接触させるのに十分な第1の液面高さまで前記容器中に洗浄液を分配する工程、
(c)付着しない標識を洗浄液中へ除去するために、分配と吸引とが同じノズル又は分配と吸引とが異なるノズルのいずれかを上下に動かして容器中の前記洗浄液の前記液面を上下に揺動させる工程、および
(d)表面コートされた前記容器から、前記洗浄液を除去する工程、
を含む、アナライザーの表面コートされた容器の壁に付着した被検体および標識を洗浄する方法。 - 工程(b)〜(d)に2.5秒以下を要する、請求項1に記載の方法。
- 前記第1の液面高さよりも低く、且つ前記付着した被検体および標識の少なくとも一部と接触させるのに十分な液面高さまで、容器中に洗浄液を分配する後続の工程、および
容器から洗浄液を除去する後続の工程、
を更に含み、前記揺動工程(c)をいずれかの前記分配及び除去の工程の間に行う、請求項1に記載の方法。 - 前記アナライザーが免疫診断用検定アナライザーである、請求項1に記載の方法。
- 前記容器がカップ型であり、且つ抗体を塗布されている、請求項4に記載の方法。
- 測定される前記被検体がトロポニンIである、請求項4に記載の方法。
- 前記後続の洗浄液除去の工程の後に、追加の液を加える工程及び表面部分除去の工程を更に含み、
前記追加の液が信号試薬を含有し、その液面部分を、分析される付着した被検体に先立って吸引により除去し、
前記アナライザーが免疫診断用検定アナライザーであり、前記容器がカップ型で且つ抗体を塗布されており、且つ測定される前記被検体が肝炎である、
請求項3に記載の方法。 - (a)コートされた容器中に被検体を保持する試料を提供する工程、
(b)前記容器中に試薬を提供する工程、
(c)随意に、一緒にした前記試料及び試薬をインキュベーションする工程、
(d)請求項1に従って洗浄を行う工程、
(e)随意に、信号試薬を加える工程、および
(f)前記被検体に関して試料を分析する工程、
を含む、試料中の被検体の量を決定する方法。
