Description
・実施例1
脂肪酸を含む原料として、純度80%のリノール酸(和光純薬工業株式会社製)2.8gを用い、これに炭酸カリウム(和光純薬工業株式会社製)7.0g、および、蒸留水300mLを加えて試験溶液を調製した。この時の試験溶液のpHは11であった。
収率(%)=(生成した13-oxoODAまたは9-oxoODAのwt%)/(使用した原料リノール酸の初期wt%)
試験溶液のリポキシゲナーゼとの反応時間を、36時間に替えて、24時間にしたこと以外は実施例1と同様に操作・分析を行った。
脂肪酸を含む原料として、純度80%のリノール酸(和光純薬工業株式会社製)2.8gを用い、これに炭酸カリウム(和光純薬工業株式会社製)4.0g、リン酸水素二カリウム(和光純薬工業株式会社製)23.3g、および、蒸留水300mLを加えて試験溶液を調製した。この時の試験溶液のpHは9.0であった。
脂肪酸を含む原料として、純度80%のリノール酸(和光純薬工業株式会社製)0.1gを用い、これに炭酸カリウム(和光純薬工業株式会社製)0.23g、および、蒸留水10mLを加えて試験溶液を調製した。この時の溶液のpHは11であった。
反応温度を、5℃に替えて、30℃にしたこと以外は比較例1と同様に操作・分析を行った。
脂肪酸を含む原料として、純度80%のリノール酸(和光純薬工業株式会社製)0.28gを用い、これに炭酸カリウム(和光純薬工業株式会社製)0.15g、炭酸水素ナトリウム(和光純薬工業株式会社製)0.05g、および、蒸留水10mLを加えて試験溶液を調製した。この時の溶液のpHは10.2であった。
反応温度を、5℃に替えて、30℃にしたこと以外は比較例5と同様に操作・分析を行った。
Claims (17)
- (a)リノール酸を含む原料に植物体由来のリポキシゲナーゼを含む酵素材料を作用させる工程、
(b)工程(a)で得られた反応混合物に含まれる前記リポキシゲナーゼの酵素反応を停止させる工程、および
(c)前記工程(b)を経て得られた生成物に脱水素酵素を作用させる工程
を含む、ケトオクタデカジエン酸の製造方法。 - 前記リノール酸を含む原料が、リノール酸を構成成分として含有する油脂を含む原料、リノール酸を含む原料、または、リノール酸である請求項1記載のケトオクタデカジエン酸の製造方法。
- 前記酵素材料を作用させる工程を、10℃以上、60℃以下で行う請求項1または2記載のケトオクタデカジエン酸の製造方法。
- 前記酵素材料を作用させる工程を、20℃以上、50℃以下で行う請求項1~3のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記酵素材料を作用させる工程において、前記酵素材料を0.5時間以上、72時間以下作用させる請求項1~4のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記酵素反応を停止させる工程を、前記リポキシゲナーゼの酵素活性を失活させることにより行う請求項1~5のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記失活が、加熱処理またはpH調整により行われる請求項6記載のケトオクタデカジエン酸の製造方法。
- 前記加熱処理を、前記反応混合物を70℃以上、140℃以下に加熱することにより行う請求項7記載のケトオクタデカジエン酸の製造方法。
- 前記pH調整を、前記反応混合物をpH6未満またはpH12超に調整することにより行う請求項7記載のケトオクタデカジエン酸の製造方法。
- 前記酵素反応を停止させる工程を、前記リポキシゲナーゼを前記反応混合物から除去することにより行う請求項1~5のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記酵素反応を停止させる工程を、前記反応混合物に前記リポキシゲナーゼの阻害剤を添加し、前記リポキシゲナーゼの酵素活性を阻害することにより行う請求項1~5のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記脱水素酵素が、アルコール脱水素酵素である請求項1~11のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記脱水素酵素を作用させる工程を、補因子の存在下で行う請求項1~12のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記補因子が、NAD+である請求項13記載のケトオクタデカジエン酸の製造方法。
- 前記脱水素酵素を作用させる工程を、0℃以上、50℃以下で行う請求項1~14のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記脱水素酵素を作用させる工程を、6以上、12以下のpHで行う請求項1~15のいずれか1項に記載のケトオクタデカジエン酸の製造方法。
- 前記リノール酸を含む原料に前記リポキシゲナーゼを含む酵素材料を作用させてのち0.5~24時間以内に前記加熱処理を行う請求項7記載のケトオクタデカジエン酸の製造方法。
