試験例1.プロテアーゼ活性亢進表試験
ヒト真皮由来線維芽細胞NB1RGBを、0.5%NCS含有イーグル最少必須培地を入れた96穴マイクロプレートに1×104個/穴播種し、37℃,5.0%CO2の条件下に1日間プレ培養した後、製造例1~3の加水分解物溶液を2.0%の濃度(培地全量に対する溶液としての終濃度)となるように培地に添加し、同条件でさらに3日間培養した。その後、それぞれの試験区の細胞培養上清を除去し細胞溶解液(1%Tween 20を含むTris-HCLバッファー(pH 8.0):12mMトリスヒドロキシメチルアミノメタン、5mM EDTA、150mM NaCl、塩酸)100μLを用いて細胞を破砕したものを粗酵素液とした。粗酵素液の内50μLを別プレートに取分け、そこへ基質として65μM Suc-Leu-Leu-Val-Tyr-AMCを含むバッファー10μLを加え、37℃で1時間反応させた。反応終了後、蛍光プレートリーダ―(フルオロスキャン アセント、Thermo Labsystems社製)を用いて蛍光強度(励起波長:355nm、蛍光波長:460nm)を測定し、これをプロテアーゼとした。試料溶液に代えてPBS(-)を添加した試料無添加の場合(対照)についても上記と同様の操作を行い、ここに得られたプロテアーゼ活性に対する各試料添加時のプロテアーゼ活性の相対値を求め、プロテアーゼ活性率(%)とした。

After the reaction, the fluorescence intensity (excitation wavelength: 355nm, fluorescence wavelength: 460nm) was measured using a fluorescent plate reader (Fluoroscan Ascent, Thermo Labsystems) and this was taken as the protease. The same procedure as above was also carried out for the control case where PBS(-) was added instead of the sample solution, and the relative value of the protease activity when each sample was added to the protease activity obtained here was calculated, and this was taken as the protease activity rate (%).
