用量・用法
本発明の女性ホルモン様活性増強剤は、経口投与によって投与される。本発明の女性ホルモン様活性増強剤の投与量については、女性ホルモンの減少の程度、年齢等に応じて適宜設定されるが、例えば、1日当たりのエクオール摂取量として、例えば0.1~100mg、好ましくは0.5~40mg、より好ましくは1.0~10mg、一層好ましくは1.5~5mg、特に好ましくは1.8~3mgが挙げられる。

試験例
エクオールとして、(S)-エクオール(和光純薬(株))を用意した。また、ニンジンエキスとして、高麗人参の細根を60v/v%エタノール水溶液で抽出し、乾燥することによって得たエキス末を用いた。エクオール及びニンジンエキスを表1に示す比率でジメチルスルホキシドに溶解し、培地を用いて希釈することで、被験試料液(1v/v%ジメチルスルホキシド)を調製した。なお、培地としては、ピルビン酸ナトリウム含有、L-グルタミン不含、フェノールレッド不含ダルベッコ改変イーグル培地(1.0g/Lグルコース)(液体)に、3v/v%のチャコール処理ウシ胎児血清、100U/mLペニシリン、100pg/mLストレプトマイシンを添加し、Lグルタミン濃度が0.5mMとなるように添加して調製したものを用いた。

比較例1に示すように、エクオール単独の女性ホルモン様作用は不十分であり、比較例2~4に示すように、ニンジンエキスについてはエクオールの5倍以上の量を用いることで単独で女性ホルモン様作用を示したが、その程度は不十分であった。一方で、実施例1及び2に示すようにエクオールとニンジンエキスとを所定比率で組み合わせることで、女性ホルモン様作用の相乗的な増強効果(相加効果を超える効果)が示された。中でも、実施例2に示す比率でエクオールとニンジンエキスと組み合わせることで、女性ホルモン様作用の相乗的な増強効果がより一層高められる(相加効果を超える程度がより一層高くなる)ことが認められた。一方で、比較例5に示すように、エクオールとニンジンエキスとが所定比率を超えて組み合わされると、女性ホルモン様作用の相加効果を下回る結果となった。つまり、実施例1及び2で認められる女性ホルモン様作用の増強効果は、エクオールとニンジンエキスとの所定比率での組み合わせに特有の効果であることが認められた。 As shown in Comparative Example 1, the female hormone-like effect of equol alone was insufficient, and as shown in Comparative Examples 2 to 4, carrot extract alone showed a female hormone-like effect when used in an amount more than five times that of equol, but the degree of the effect was insufficient. In other words, it was recognized that the enhancement effect of the female hormone-like effect observed in Examples 1 and 2 is an effect specific to the combination of equol and carrot extract at a specified ratio.
