Description
(1)有効成分、数平均分子量3,800未満の両親媒性ブロック共重合体、及びC1~C5アルコールを混合する工程、(2)得られた工程(1)の混合物からC1~C5アルコールを除去して、混合物中のC1~C5アルコール含量を2.5w/v%未満に下げる工程、及び(3)得られた工程(2)の混合物に水性媒質を加えて、ミセルを形成させる工程を含むナノ粒子の製造方法を提供する。
本発明において、用語「ナノ粒子」は、ミセル又はミセル組成物と同じ意味で使用することができる。一実施形態では、「ナノ粒子」は、サブミクロンサイズ(例えば、粒径)、すなわち、1マイクロメートル未満(例えば、10~900nm、又は10~500nm)を有することができる。
モノメトキシポリエチレングリコール-(ポリ-D,L-乳酸)共重合体(mPEG-PDLLA)19gとドセタキセル1gを秤量し、エタノール(EtOH)4mLを加え、完全に溶解して透明な溶液になるまで60℃で撹拌した。次に、丸底フラスコ付きの回転式減圧蒸留器を利用して、下記表1に示す温度及び時間条件に従って、減圧蒸留によりエタノールを除去した。その後、温度を28℃に下げ、室温の蒸留水230mLを加え、溶液が青色で透明になるまで反応を行い、3時間以内に高分子ミセルを形成させた。
下記表2に示す条件に従って減圧蒸留によりエタノールを除去したことを除いて、前記実施例1~3及び比較例1と同様の手順を行った。エタノール除去後、残留エタノール含量は約0.50w/v%であった。その後、下記表2に示すミセル化温度まで温度を下げ、室温の蒸留水230mLを加え、所定時間反応させて、高分子ミセルを形成した。
ろ過したミセルを下記表3に示す条件下で保管したことを除いて、前記実施例1と同様の手順を行い、凍結乾燥剤としてのマンニトール5gを加えて完全に溶解し、空隙サイズ200nmのフィルターでろ過した後、凍結乾燥して、ドセタキセルを含有する高分子ミセル組成物を粉末で製造した。
実施例1~3及び5、並びに比較例1、3、4のそれぞれで製造されたドセタキセルを含む高分子ミセル2gに生理食塩水38mLを加え、IKA VIBRAX撹拌機を使用して溶解した後、溶液を静置して、30分ごとにドセタキセルの析出を観察した。その結果を下記表4に示す。
試験溶液は、実施例1~4及び比較例2のそれぞれで製造されたドセタキセルが含まれた高分子ミセルの調製液を45μmのろ紙でろ過し、ろ過した溶液1mLをHPLC移動相溶液4mLと混合した。調製された試験液を以下の条件でHPLC分析し、類縁物質含量を比較した。類縁物質の含量(%)を下記表5に示す。
カラム:粒径5μm、孔径100Åのシリカゲル粒子を有し、C18をコートした長さ250mm、内径4.6mmのステンレスカラム又はそれに類似したもの
移動相:アセトニトリル/メタノール/水混合液(26/32/42、v/v/v)
流速:1.5mL/分
検出器:紫外吸光光度計(測定波長232nm)
実施例1~3及び比較例1、高分子の分子量が異なる比較例5のそれぞれで調製したドセタキセルが含まれた高分子ミセル2gに生理食塩水38mLを加え、IKA VIBRAX撹拌機を使用して500rpmの撹拌速度で凍結乾燥固形物が完全に溶解するのにかかる時間(再溶解時間)を測定した。その結果を下記表6に示す。
Claims (4)
- (1)有効成分、数平均分子量3000~3700の両親媒性ブロック共重合体及びエタノールを混合する工程、
(2)得られた工程(1)の混合物からエタノールを除去して、混合物中のエタノール含量を0.8w/v%以下に下げる工程、
(3)得られた工程(2)の混合物に水性媒質を加えて、ミセルを形成させる工程及び
(4)工程(3)で形成されたミセルを凍結乾燥する工程
を含み、
両親媒性ブロック共重合体が、ポリエチレングリコール、モノメトキシポリエチレングリコール及びそれらの組み合わせよりなる群から選ばれる親水性ブロック(A)、並びに
ポリラクチド、ポリグリコリド、ポリ(乳酸-グリコリド)及びそれらの組み合わせよりなる群から選ばれる疎水性ブロック(B)を含み、
エタノールの除去が30℃超45℃以下の温度で行われ、
ミセルの形成が35℃未満の温度で行われ、
工程(3)で形成されるミセルが3時間以内に凍結乾燥されることを特徴とするナノ粒子の製造方法。 - ミセルを形成させる工程が、形成されたミセルをろ過する過程を含む請求項1に記載のナノ粒子の製造方法。
- 形成されたミセルが、凍結乾燥が行われる前まで0℃~15℃に維持される請求項1に記載のナノ粒子の製造方法。
- 有効成分が、難水溶性薬物である請求項1~3のいずれか1項に記載のナノ粒子の製造方法。
