Description
本発明はデキストラナーゼを安定に配合した口
腔用組成物に関する。
従来、う蝕予防の有効成分として歯垢の形成を
防止するためデキストラナーゼを口腔用組成物に
配合することが知られている(特許第782154
号)。
しかるに、デキストラナーゼは口腔用組成物中
の水分、アニオン活性剤等の影響で失活し易い問
題があり、このため従来よりデキストラナーゼを
口腔用組成物中に安定に配合するための方法が
種々提案されている(特許第802787号、同924229
号、特開昭53−26334号など)が、まだその安定
化は特に高温下において保存された場合十分では
なく、従つてデキストラナーゼを口腔用組成物中
においてより安定化させ得る方法が望まれてい
る。
本発明者らはデキストラナーゼの口腔用組成物
への安定化配合につき種々検討を行なつた結果、
デキストラナーゼと共に下記式(1)
NH2−CH2−R−COOH ……(1)
(但し、Rはシクロヘキサン環もしくはn−C4H8
基を示す)
で示されるオメガアミノ酸又はそのアルキルエス
テルもしくはアリールエステル誘導体、特にトラ
ネキサム酸、イプシロンアミノカプロン酸を配合
すると、デキストラナーゼが口腔用組成物中にお
いて安定化し、40℃の高温下に口腔用組成物を長
期間保存した後でもデキストラナーゼの残存率が
高く、デキストラナーゼの効果が有効に発揮され
ると共に、デキストラナーゼと歯槽膿漏予防効果
をもつトラネキサム酸、イプシロンアミノカプロ
ン酸等のオメガアミノ酸、その誘導体との併用に
より口腔用組成物の有効性が高まり、う蝕予防効
果並びに歯槽膿漏予防効果が相乗的に高くなるこ
とを知見し、本発明をなすに至つたものである。
以下、本発明につき詳しく説明する。
本発明に係る口腔用組成物は、練歯磨、潤製歯
磨、液状歯磨などの歯磨類、マウスウツシユ、ト
ローチ、チユーインガム、歯肉マツサージクリー
ム等として用いられるもので、デキストラナーゼ
と共に前記(1)式で示されるオメガアミノ酸又はそ
のアルキルエステルもしくはアリールエステル誘
導体の1種又は2種以上を配合するものである。
この場合、デキストラナーゼの配合量は、通常
歯磨1g当り100〜100000単位(u/g歯磨)で
あり、特に好ましくは1000〜50000u/g歯磨配
合される。なお、ここで1単位とは、デキストラ
ンを基質として反応を行なつた場合、1分間当り
グルコース1μgに相当する遊離還元糖を生ずる
酵素量をいう。
また、オメガアミノ酸としては、前記(1)式にお
いてRがシクロヘキサン環であるトラネキサム
酸、n−C4H8基であるイプシロンアミノカプロ
ン酸が使用でき、またこれらのアルキルエステル
もしくはアリールエステル誘導体としてはイプシ
ロンアミノカプロン酸ヘキシルエステル、イプシ
ロンアミノカプロン酸ヘプチルエステル、トラネ
キサム酸ヘキシルエステル、トラネキサム酸フエ
ニルエステル等が使用できるが、特に歯槽膿漏予
防の面よりトラネキサム酸、イプシロンアミノカ
プロン酸が好適に用いられる。
上記オメガアミノ酸、その誘導体の配合量は、
デキストラナーゼの安定化能の面から0.01%(重
量%、以下同じ)以上、また口腔用組成物の使用
感、安定性の面から3%以下とすることが好まし
く、この範囲においてデキストラナーゼの安定化
効果が発揮されると共に、う蝕予防、歯槽膿漏予
防効果も良好に発揮され、また使用感も良好なも
のである。なお、オメガアミノ酸、その誘導体の
より好ましい配合量は0.03〜1%である。
本発明においては、口腔用組成物の種類等に応
じて通常使用される適宜な成分が配合され得る。
例えば、歯磨類の場合には、第2リン酸カルシ
ウム・2水和物及び無水物、炭酸カルシウム、ピ
ロリン酸カルシウム、酸化アルミニウム化合物、
シリカ、ケイ酸アルミニウム、不溶性メタリン酸
ナトリウム、第3リン酸マグネシウム、炭酸マグ
ネシウム、硫酸カルシウム、合成樹脂等の1種又
は2種以上を配合し得る(配合量通常20%〜90
%、練歯磨の場合には20%〜60%)。特に、これ
らのうちでは下記一般式(2)
Al2O3・nH2O ……(2)
(但し、0≦n、好ましくは0≦n≦3)
で示される酸化アルミニウム化合物(アルミナ、
アルミナ水和物)が好適に用いられ、この酸化ア
ルミニウム化合物を主研磨剤として使用すること
により、デキストラナーゼの安定化をより確実に
達成し得る。この場合、酸化アルミニウム化合物
としてはα−アルミナ、α−アルミナ水和物が好
ましい。アルミナ水和物はジプサイト、バイヤラ
イト、ベーマイト、ダイアスポア等が好適に使用
でき、特に3水和物がマイルドな研磨剤として優
れているが、これらのうちではジプサイトが供給
性などの点で好ましく用いられる。
また、練歯磨の場合には粘結剤としてカラゲナ
ン、カルボキシメチルセルロースナトリウムなど
のセルロース誘導体、アルギン酸ナトリウムなど
のアルカリ金属アルギネート、ビーガム、キサン
タンガムなどのガム類、ポリビニルアルコールな
どの合成粘結剤、シリカゲル、アルミニウムシリ
カゲルなどの無機粘結剤等の1種又は2種以上が
配合され得る(配合量通常0.3〜5%)。特に、有
効成分としてアルカリ金属モノフルオロホスフエ
ートを配合するときは、歯磨組成物の安定性、使
用感の面からカラゲナンとアルギン酸アルカリ金
属塩とを併用することが好ましい。この場合通常
のカラゲナン(カツパ−/イオタ−混合系)に比
べてカツパ−カラゲナンは歯磨組成物の肌あれ防
止の点で効果的であるから、歯磨組成物の安定性
の点でカラゲナンとしてカツパ−カラゲナンを使
用することが好ましい。
更に、歯磨類の製造において、粘稠剤としてソ
ルビツト、グリセリン、エチレングリコール、プ
ロピレングリコール、1・3−ブチレングリコー
ル、ポリエチレングリコール、キシリツト、マル
チツト、ラクチツク等の1種又は2種以上を配合
し得る(配合量通常10〜70%)。この場合粘結剤
分散液としてプロピレングリコールを1〜5%用
いることが望ましいが、多量に配合すると保存に
よるデキストラナーゼ残存率が低下するため、主
粘稠剤としてソルビツトを配合し、プロピレング
リコール1重量部とソルビツト6〜60重量部とを
併用することが望ましい。
更にまた、歯磨類を製造するに際し、ラウリル
硫酸ナトリウム、ミリスチル硫酸ナトリウム等の
アルキル基の炭素数が8〜18である高級アルキル
硫酸の水溶性塩、ソジウムラウリルモノグリセラ
イドスルホネート、ソジウムココナツツモノグリ
セライドスルホネート等の脂肪酸基の炭素数が10
〜18である高級脂肪酸モノグリセライドスルホネ
ートの水溶性塩、高級脂肪酸ソジウムモノグリセ
ライドモノサルフエート、ソジウム−N−メチル
−N−パルミトイルタウライド、ソジウム−N−
ラウロイルザルコシネート、ソジウム−N−ラウ
ロイル−β−アラニン等の脂肪酸基の炭素数が12
〜18である高級脂肪酸と低級脂肪族アミノ酸との
アマイドの塩といつたアニオン活性剤、ラウリン
酸ジエタノールアミド等の脂肪酸ジエタノールア
ミド、ステアリルモノグリセライド、シヨ糖モノ
及びジラウレート等の脂肪酸基の炭素数が12〜18
であるシヨ糖脂肪酸エステル、ラクトース脂肪酸
エステル、ラクチトール脂肪酸エステル、マルチ
トール脂肪酸エステル、エチレングリコール約60
モルが付加したソルビタンモノステアレート縮合
物、エチレンオキサイドとプロピレンオキサイド
の重合物及びポリオキシエチレンポリオキシプロ
ピレンモノラウリルエステル等の誘導体といつた
ノニオン活性剤、ベタイン型、アミノ酸型等の両
性活性剤などの1種または2種以上の界面活性剤
(配合量通常0.5〜7%)を配合し得る。香料とし
てはペパーミント、スペアミント等の精油、l−
メントール、カルボン、オイゲノール、アネトー
ル等の香料素材が単独で又はこれらを組合せて全
体の0.1〜5%程度配合し得る。サツカリンナト
リウム、ステビオサイド、ネオヘスペリジルジヒ
ドロカルコン、グリチルリチン、ペリラルチン、
p−メトキシシンナミツクアルデヒドなどの甘味
剤、p−ヒドロキシメチルベンゾイツクアシド、
p−ヒドロキシブチルベンゾイツクアシドなどの
防腐剤、その他の成分を配合し得、例えば練歯磨
の場合には上記した所望の成分を適量の水と練合
することにより製造し得る。
また、マウスウツシユ、トローチ、チユーイン
ガム、歯肉マツサージクリーム等を製造する場合

キストラナーゼ、オメガアミノ酸類に加えて更に
アミラーゼ、プロテアーゼ、ムタナーゼ、リゾチ
ーム、リテツクエンザイム等の酵素、ソジウムモ
ノフルオロホスフエート、ポタジウムモノフルオ
ロホスフエートなどのアルカリ金属モノフルオロ
ホスフエート、フツ化ナトリウム、フツ化第1錫
等のフツ化物、クロルヘキシジン塩類、アルミニ
ウムクロルヒドロキシルアラントイン、ジヒドロ
コレステロール、グリチルリチン塩類、グリセロ
ホスフエート、塩化ナトリウム、水溶性無機リン
酸化合物等の有効成分を1種又は2種以上配合し
得る。特に、ソジウムモノフルオロホスフエート
などのアルカリ金属モノフルオロホスフエートを
デキストラナーゼと併用すると、デキストラナー
ゼを安定化し、長期保存後におけるデキストラナ
ーゼの歯磨組成物中での残存率を高く維持するこ
とができるので望ましい。この場合、アルカリ金
属モノフルオロホスフエートの配合量は0.1〜1
重量%とすることが好ましい。また、デキストラ
ナーゼとムタナーゼとを組合せる場合、歯垢の溶
解と再付着防止の相乗作用が生じ、デキストラナ
ーゼとリテツクエンザイムとの組合せはリテツク
エンザイムの効力を強くする等のメリツトがあ
る。なお、水溶性無機リン酸化合物としては正リ
ン酸、ピロリン酸、ポリリン酸のカリウム塩又は
ナトリウム塩があるが、カリウム塩が好ましい。
本発明に係る口腔用組成物は、デキストラナー
ゼにオメガアミノ酸又はそのアルキルエステルも
しくはアリールエステル誘導体の1種又は2種以
上を併用したことにより、デキストラナーゼが安
定化し、デキストラナーゼの歯垢除去効果が長期
間に亘り高く保持されると共に、このように歯垢
除去効果が高まることによつてう蝕予防のみなら
ず歯槽膿漏予防効果も高まり、デキストラナーゼ
とトラネキサム酸、イプシロンアミノカプロン酸
等のオメガアミノ酸、その誘導体との併用でより
高いう蝕予防効果並びに歯槽膿漏予防効果を発揮
するものである。
次に実験例を示し、本発明の効果を具体的に説
明する。
