Description
【発明の詳細な説明】
〔発明の利用分野〕
本発明は、発酵槽の植菌装置に関するもので、
特にバイオハザード対策設備の発酵槽に適する植
菌装置に関するものである。
〔発明の背景〕
従来の発酵槽の植菌方法は、火炎植菌方法又は
注射植菌方法のいずれかであり、これらはいずれ
も植菌する場合、その構造上種菌中の微生物が周
囲の外気、あるいは植菌作業を行なう作業者を汚
染するという欠点があり、バイオハザード対策を
行なつた設備の中で使用することが難かしく、特
に微生物が周囲の生物系に対し危険な場合には、
従来方法の植菌装置は使用できなかつた。
従来の火炎植菌については、例えば「発酵工
業」木下祝郎著大日本図書株式会社昭和50年7月
1日発行、95頁において論じられている。
〔発明の目的〕
本発明の目的は、種菌を無菌的に、かつ、植菌
中の微生物が周囲を汚染するのを防止した発酵槽
の植菌装置を提供することにある。
〔発明の概要〕
本発明は、発酵槽の植菌口に植菌バルブを介し
て接続した植菌ラインにドレンラインを接続し、
前記植菌ラインと加熱殺菌用蒸気および冷却、圧
押し用無菌空気を供給する蒸気・無菌空気供給ラ
インとの間に、バイパスラインを備えた植菌ポツ
トユニツトを着脱可能に接続して、種菌を無菌的
に、かつ、植菌中の微生物が周囲の環境を汚染す
ることなく、発酵槽内に植菌するようにしたもの
である。
〔発明の実施例〕
以下、本発明の実施例を図面により説明する。
発酵槽1の上部の植菌口13には植菌バルブ12
を介して植菌ライン16が接続されており、植菌
ライン16にはドレンバルブ10を介してドレン
ライン11が接続されている。2は上蓋15をネ
ジ結合等により着脱可能に装着した植菌ポツト
2、上蓋15に圧押しバルブ3、T継手17を介
して取付けられたカツプラー9、植菌ポツト2の
下部にバルブ4を介して取付けられたカツプラー
8、バルブ4よりバイパスライン6を介してT継
手17に接続されたカツプラー7よりなる植菌ポ
ツトユニツト、14は加熱殺菌用蒸気および冷
却、圧押し用無菌空気を供給する蒸気・無菌空気
供給ラインであつて、植菌ライン16と蒸気・無
菌空気供給ライン14の間にカツプラー8,9を
介して植菌ポツトユニツト2が着脱可能に接続さ
れている。
上述の構成において、種菌の発酵槽1内への植
菌について説明する。まず、あらかじめオートク
レーブ(図示せず)で殺菌された植菌ポツト2、
上蓋15、圧押しバルブ3、T継手17、バルブ
4,5、バイパスライン6、カツプラー7,8,
9よりなる植菌ポツトユニツト2を安全キヤビネ
ツト(図示せず)の中に入れ、カツプラー7を離
脱して上蓋15を取外す。つぎに、圧押しバルブ
3およびバルブ4が閉じていることを確認した
後、植菌ポツト2内に種菌を入れ蓋15を取付
け、カツプラー7を結合して植菌ポツトユニツト
2を安全キヤビネツトより取出し、カツプラー8
を植菌ライン16に接続し、カツプラー9を蒸
気・無菌空気供給ライン14に接続する。しかし
て、バルブ5およびドレンバルブ10を開いて、
蒸気・無菌空気供給ライン14よりカツプラー
9,7、バイパスライン6、バルブ5を経て植菌
ライン16に蒸気を供給し、ドレンバルブ10を
介してドレンライン11より排出することによ
り、植菌ライン16を殺菌した後、無菌空気を供
給して植菌ライン16を冷却する。植菌ライン1
6の冷却が完了すると、バルブ5、ドレンバルブ
10を閉じ、圧押しバルブ3を開いて植菌ポツト
2内に無菌空気を供給して加圧し、バルブ4、植
菌バルブ12を開いて植菌ポツト2内の種菌を発
酵槽1内に投入植菌する。発酵槽1への植菌が終
了すると、上述と同様に植菌ライン16の殺菌を
行なつた後、カツプラー8,9を離脱させて植菌
ポツトユニツト2を発酵槽1より取外す。
〔発明の効果〕
本発明は以上述べたように、発酵槽の植菌口に
植菌バルブを介して植菌ラインを接続し、前記植
菌ラインにドレンバルブを介してドレンラインを
接続し、前記植菌ラインと蒸気・無菌無気供給ラ
インとの間にバイパスラインを備えた植菌ポツト
ユニツトを着脱可能に接続したものであるから、
植菌操作中種菌を完全に無菌的に保持するとがで
きると共に、植菌中の微生物が周囲の環境を汚染
するのを未然に防止することができ、バイオハザ
ードの心配が全くない発酵槽の植菌装置を得るこ
