Description
〔発明の目的〕
本発明は抗菌剤の中間体として有用な新規3−
(キヌクリジニオ)メチル−3−セフエム誘導体
に関するものである。
従来、セフエム骨格の7位に置換チアジアゾリ
ルアセトアミド基または置換チアゾリルアセトア
ミド基を有する多くの化合物が知られている。例
えば、特開昭55−11600号、特開昭55−105689号、
特開昭57−24389号、特開昭57−81493号、特開昭
58−41887号、特開昭58−59992号、特開昭59−
219292号、特開昭51−149296号、特開昭52−
102293号、特開昭52−116492号、特開昭52−
125190号、特開昭54−154786号、特開昭57−
192394号等の公報記載の化合物があげられる。
しかし、そのセフエム骨格の3位に(4−カル
バモイル−1−キヌクリジニオ)メチル基を有す
る化合物は知られていない。本発明者等は、3位
に上記の基を有する化合物が優れた抗菌力を有す
ることを見い出した。本発明化合物はその新規な
合成中間体である。
したがつて、本発明の目的は、抗菌剤の中間体
として有用な新規化合物、その製造方法を提供す
ることにある。
〔発明の構成〕
本発明は、一般式
〔式中、nは0または1を示し、R1およびR2
が共に水素原子、R1がアミノ基の一価の保護基
でR2が水素原子、あるいはR1とR2でアミノ基の
二価の保護基を示し、Xは陰イオンを示し、R3
が−COO-のときはmは0、R3が−COOR4(R4は
カルボキシル基の保護基)のときはmは1を示
す〕で表わされる3−(キヌクリジニオ)メチル
−3−セフエム誘導体およびその塩である。
上記一般式()の化合物におけるR1のアミ
ノ基の一価の保護基としては、通常使用されるも
の、例えばホルミル、アセチル、クロロアセチ
ル、ジクロルアセチル、プロピオニル、フエニル
アセチル、2−チエニルアセチル、2−フリルア
セチル、フエノキシアセチルなどの置換基または
非置換の低級アルカノイル基;ベンジルオキシカ
ルボニル、t−ブトキシカルボニル、p−ニトロ
ベンジルオキシカルボニルなどの置換または非置
換の低級アルコキシカルボニル基;トリチル、p
−メトキシベンジル、ジフエニルメチルなどの置
換低級アルキル基;トリメチルシリル、t−ブチ
ルジメチルシリルなどの置換シリル基などがあげ
られる。また、R1とR2で形成するアミノ基の二
価の保護基としては、ベンジリデン、サリチリデ
ン、3,5−ジ(t−ブチル)−4−ハイドロキ
シベンジリデン、3,5−ジ(t−ブチル)ベン
ジリデンなどの置換または非置換のベンジリデン
基等があげられる。
R4のカルボキシル基の保護基としては、通常
使用されるもの、例えばメチル、エチル、プロピ
ル、t−ブチル、2,2,2−トリクロロエチ
ル、バレリルオキシメチル、ピバロイルオキシメ
チル、p−ニトロベンジル、p−メトキシベンジ
ル、ジフエニルメチルなどの置換または非置換の
低級アルキル基;トリメチルシリル、t−ブチル
ジメチルシリルなどの置換シリル基などがあげら
れる。
Xの陰イオンとしては、クロルイオン、ブロム
イオン、ヨードイオンなどのハロゲンイオン;硫
酸イオン、硝酸イオンなどの無機酸イオンなどが
あげられる。
一般式()の化合物の塩としては、塩酸塩、
臭化水素酸塩、沃化水素酸塩、硫酸塩、炭酸塩、
重炭酸塩などの無機酸塩;酢酸塩、マレイン酸
塩、乳酸塩、酒石酸塩、トリフルオロ酢酸塩など
の有機カルボン酸塩;メタンスルホン酸塩、ベン
ゼスルホン酸塩、トルエンスルホン酸塩などの有
機スルホン酸塩;アスパラギン酸塩、グルタミン
酸塩などのアミノ酸塩等があげられる。
本発明化合物は次に示す方法により製造するこ
とができる。
一般式
〔式中、n,R1,R2は前記の定義と同じ、R5
は水素原子またはカルボキシル基の保護基、Yは
ハロゲン原子または低級アルカノイルオキシ基を
示す〕で表わされる化合物またはその塩に式
で表わされる化合物またはその塩を反応させ、必
要により保護基を脱離およびまたはスルホキシド
を還元して前記一般式()の化合物またはその
塩を得る。
一般式()におけるYがハロゲン原子の場合
の上記反応は、例えばアセトン、テトラヒドロフ
ラン、N,N−ジメチルホルムアミド、塩化メチ
レン、クロロホルム、アセトニトリルなどの不活
性溶媒中、反応温度−10℃〜50℃で行なうことが
できる。
また、一般式()におけるYが低級アルカノ
イルオキシ基の場合の上記反応は、例えばクロロ
ホルム、塩化メチレン、テトラヒドロフラン、
N,N−ジメチルホルムアミド、ジオキサン、ア
セトンなどの不活性溶媒中、ヨードトリメチルシ
ランの存在下、反応温度−20℃〜60℃で行なうこ
とができる。
保護基の脱離は、使用した保護基の種類に応
じ、加水分解、還元あるいは酵素による加水分解
など常法により行なうことができる。また、スル
ホキシドの還元も、三塩化リンを反応させる等、
常法により行なうことができる。
一般式()の化合物のYのハロゲン原子とし
ては、塩素原子、臭素原子および沃素原子があげ
られる。また、Yの低級アルカノイルオキシ基と
してはアセチルオキシ、プロピオニルオキシなど
があげられる。
R3のカルボキシル基の保護基としては前記の
R4の説明でのべた基があげられる。また、一般
式()および一般式()の化合物の基として
は、上記反応を妨げないものであれば使用するこ
とができる。例えばナトリウム塩、カリウム塩な
どのアルカリ金属塩;カルシウム塩、マグネシウ
ム塩などのアルカリ土類金属塩;アンモニウム
塩;塩酸塩、硫酸塩、炭酸塩、重炭酸塩、臭化水
素酸塩、沃化水素酸塩などの無機酸塩;酢酸塩、
マレイン酸塩、乳酸塩、酒石酸塩、トリフルオロ
酢酸塩などの有機カルボン酸塩;メタンスルホン
酸塩、ベンゼンスルホン酸塩、トルエンスルホン
酸塩などの有機スルホン酸塩;トリメチルアミン
塩、トリエチルアミン塩、ピリジン塩、プロカイ
ン塩、ピコリン塩、ジシクロヘキシルアミン塩、
N,N′−ジベンジルエチレンジアミン塩、N−
メチルグルカミン塩、ジエタノールアミン塩、ト
リエタノールアミン塩、トリス(ヒドロキシメチ
ルアミノ)メタン塩などのアミン塩;アルギニン
塩、アスパラギン酸塩、リジン塩、グルタミン酸
塩、セリン塩などのアミノ酸塩等の塩の中より適
宜選択することができる。
本発明化合物を合成中間体として用い、その7
位のアミノ基へ公知方法で種々の基、例えば2−
(5−アミノ−1,2,4−チアジアゾール)−
(Z)−2−メトキシイミノアセチル基等を導入す
ることにより優れた抗菌活性を有する化合物を得
ることができる。
次に実施例を示し、本発明をさらに詳しく説明
する。
実施例 1
7β−ホルムアミド−3−(4−カルバモイル−1
−キヌクリジニオ)メチル−3−セフエム−4−
カルボキシレイト
7β−ホルムアミド−3−アセトキシメチル−
3−セフエム−4−カルボン酸1.2gを塩化メチ
レン12mlに懸濁し、N−メチル−N−(トリメチ
ルシリル)トリフルオロアセトアミド815μを
加えて30分間攪拌した。氷冷後、ヨードトリメチ
ルシラン1.25mlを加え、5分間攪拌後、室温にも
どし、さらに15分間攪拌した。溶媒を減圧留去
し、残渣をアセトニトリル12mlに溶解し、氷冷
下、4−カルバモイルキヌクリジン616mgを加え
1時間攪拌した。反応液にメタノール3mlさらに
ジエチルエーテル300ml加え生じた沈殿を取し
た。
この沈殿をシリカゲルのカラムクロマトグラフ
イー〔展開溶媒:アセトン−水(7:1)および
(5:1)〕で精製し、目的物140mgを得た。
実施例 2
7β−トリチルアミノ−3−(4−カルバモイル−
1−キヌクリジニオ)メチル−3−セフエム−4
−カルボキシレイト
7β−トリチルアミノ−3−アセトキシメチル
−3−セフエム−4−カルボン酸2.4gを塩化メ
チレン24mlに溶解し、N−メチル−N−(トリメ
チルシリル)トリフルオロアセトアミド960μ
を加えて30分間攪拌した。氷冷後、ヨードトリメ
チルシラン720μを加え、5分間攪拌後、室温
にもどし、さらに15分間攪拌した。溶媒を減圧留
去した。残渣をアセトニトリル24mlに溶解し、氷
冷下、4−カルバモイルキヌクリジン756mgを加
えて1時間攪拌した。反応液にメタノール3.2ml、
ついでジエチルエーテル240mlを加え、生じた沈
殿を取した。この沈殿をシリカゲルのカラムク
ロマトグラフイー〔展開溶媒:アセトン−水
(7:1)、(5:1)および(3:1)〕で精製し
て目的物207mgを得た。
実施例 3
7β−(2−チエニルアセトアミド)−3−(4−カ
ルバモイル−1−キヌクリジニオ)メチル−3−
セフエム−4−カルボキシレイト
7β−(2−チエニルアセトアミド)−3−アセ
トキシメチル−3−セフエム−4−カルボン酸
6.0gを塩化メチレン60mlに懸濁し、N−メチル
−N−(トリメチルシリル)トリフルオロアセト
アミド3.08mlを加え、30分間攪拌した。氷冷後、
ヨードトリメチルシラン4.73mlを加えて5分間攪
拌後、さらに室温で15分間攪拌した。溶媒を減圧
濃縮し、残渣をアセトニトリル60mlに溶解した。
氷冷後、4−カルバモイルキヌクリジン2.3gを
加え1時間攪拌した。反応液にメタノール6mlを
加え、次いでジエチルエーテル600mlを滴下した。
1時間攪拌した後、生じた沈殿を取しシリカゲ
ルのカラムクロマトグラフイー〔展開溶媒:アセ
トン−水(7:1)および(5:1)〕で精製し
て目的物700mgを得た。
実施例 4
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト 塩酸塩
実施例1で得られた化合物130mgをメタノール
5mlに懸濁し、室温で濃塩酸0.52ml加え、4時間
攪拌した。反応液を減圧濃縮し、ジエチルエーテ
ル−メタノールで結晶化し、目的物115mgを得た。
実施例 5
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト 塩酸塩
実施例2で得られた化合物100mgを50%ギ酸5
mlに懸濁し、室温で3時間30分攪拌した。これに
水20mlを加えて不溶物を去し、液を減圧濃縮
した。残渣を1N塩酸1mlに溶解し、これにイソ
プロパノール5mlおよびジエチルエーテル10mlを
加えた。析出した結晶を取し、n−ヘキサンで
洗浄後乾燥して目的物45mgを得た。
実施例 6
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト 塩酸塩
実施例3で得られた化合物600mgを塩化メチレ
ン30mlに懸濁し、N,N−ジメチルアニリン1.24
mlおよびクロロトリメチルシラン465μを加え、
30℃で3時間攪拌した。次いで反応液を−25℃に
冷却後、五塩化リン1.27gを加え1時間攪拌し
た。これに1,3−ブタンジオール1.3mlの塩化
メチレン25ml冷溶液を加え、同温度で10分間攪拌
した。反応液を0℃に加温してさらに40分間攪拌
後、生じた沈殿を取した。これをメタノール7
mlに溶解し、不溶物を去後、液に塩化メチレ
ン20mlおよびジエチルエーテル20mlを加えて析出
する結晶を取し、目的物30mgを得た。
実施例 7
t−ブチル 7β−アミノ−3−(4−カルバモイ
ル−1−キヌクリジニオ)メチル−3−セフエム
−4−カルボキシレイト 1−オキシド ブロマ
イド
t−ブチル 7β−アミノ−3−ブロムメチル
−3−セフエム−4−カルボキシレイト 1−オ
キシド 臭化水素酸塩600mgをN,N−ジメチル
ホルムアミド6mlに溶解し、4−カルバモイルキ
ヌクリジン456mgを加え、アルゴンガス気流中、
室温で14時間攪拌した。反応液にジエチルエーテ
ル120mlを加え、生じた沈殿を取し、これをn
−ヘキサンで洗浄して、目的物580mgを得た。
実施例 8
t−ブチル 7β−アミノ−3−(4−カルバモイ
ル−1−キヌクリジニオ)メチル−3−セフエム
−4−カルボキシレイト ブロマイド 塩酸塩
実施例7で得られた化合物570mgをN,N−ジ
メチルホルムアミド10mlに溶解し、−25℃で三塩
化リン500μを加え、30分間攪拌した。反応液
にジエチルエーテル50mlを加え、遊離した油を分
取し、ジエチルエーテル10mlで洗浄した後、減圧
下乾燥して目的物164mgを得た。
実施例 9
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト
実施例8で得られた化合物150mgに氷冷下、ギ
酸1.5mlおよび濃塩酸0.15mlを加え、4時間攪拌
した後、減圧下に濃縮した。残渣を氷水5mlに溶
解し、重炭酸ナトリウムで中和した。これを逆相
シリカゲルカラムクロマドグラフイー(展開溶
媒:水)で精製して目的物60mgを得た。
実施例 10
p−メトキシベンジル 7β−フエニルアセトア
ミド−3−(4−カルバモイル−1−キヌクリジ
ニオ)メチル−3−セフエム−4−カルボキシレ
イト ヨーダイド
p−メトキシベンジル 7β−フエニルアセト
アミド−3−クロロメチル−3−セフエム−4−
カルボキシレイト980mgをアセトン20mlに懸濁し、
ヨウ化ナトリウム362mgを加えて室温で1時間攪
拌した。これに、氷冷下、4−カルバモイルキヌ
クリジン313mgを加え、2時間攪拌した。反応液
を過し、液にジエチルエーテル70mlを加え
た。析出した結晶を取して目的物500mgを得た。
実施例 11
p−メトキシベンジル 7β−ホルムアミド−3
−(4−カルバモイル−1−キヌクリジニオ)メ
チル−3−セフエム−4−カルボキシレイト ヨ
ーダイド
p−メトキシベンジル 7β−ホルムアミド−
3−ヨードメチル−3−セフエム−4−カルボキ
シレイト9.3gの酢酸エチル溶液465mlに、氷冷攪
拌下、4−カルバモイルキヌクリジン2.94gのメ
タノール−酢酸エチル(1:4V/V)176ml溶液
を1時間かけて滴下した。30分間攪拌後、生じた
沈殿を取し、酢酸エチルついでジイソプロピル
エーテルで洗浄し、目的物12.0gを得た。
実施例 12
7β−ホルムアミド−3−(4−カルバモイル−1
−キヌクリジニオ)メチル−3−セフエム−4−
カルボキシレイト
実施例11で得られた化合物11.8gを氷冷した蟻
酸50mlに溶解した後、室温で10時間攪拌した。反
応液を過し、液をアセトン100ml中に滴下し
た。さらにジイソプロピルエーテル200mlを同溶
液に滴下し、生じた沈殿を取し、アセトンで洗
浄した。沈殿をジメチルホルムアミド30mlに溶解
し、溶液をアセトン150ml中に滴下した。生じた
沈殿を取し、アセトンおよびジイソプロピルエ
ーテルで洗浄後、減圧乾燥して目的物6.66gを得
た。
実施例 13
p−メトキシベンジル 7β−ベンジリデンアミ
ノ−3−(4−カルバモイル−1−キヌクリジニ
オ)メチル−3−セフエム−4−カルボキシレイ
ト ヨーダイド
p−メトキシベンジル 7β−ベンジリデンア
ミノ−3−ヨードメチル−3−セフエム−4−カ
ルボキシレイト5.5gを酢酸エチル330mlに溶解し
た。この溶液に4−カルバモイルキヌクリジン
1.39gのメタノール−酢酸エチル(17ml、68ml)
溶液を、氷冷下35分間かけて滴下した。10分間攪
拌後、生じた結晶を取し、酢酸エチルで洗浄、
乾燥して目的物5.67gを得た。
実施例 14
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト 塩酸塩
p−メトキシベンジル 7β−ベンジリデンア
ミノ−3−(4−カルバモイル−1−キヌクリジ
ニオ)メチル−3−セフエム−4−カルボキシレ
イト ヨーダイド500mgを蟻酸1mlに溶解後、濃
塩酸0.2mlを加え、室温で16時間攪拌した。反応
液にアセトン20mlを加え、析出した結晶で取し
た。これをアセトンで洗浄後、乾燥して目的物
330mgを得た。
実施例 15
7β−フエニルアセトアミド−3−(4−カルバモ
イル−1−キヌクリジニオ)メチル−3−セフエ
ム−4−カルボキシレイト
p−メトキシベンジル 7β−フエニルアセト
アミド−3−(4−カルバモイル−1−キヌクリ
ジニオ)メチル−3−セフエム−4−カルボキシ
レイト ヨーダイド13.46gを氷冷した蟻酸67ml
に溶解した後、室温で23時間、続いて30℃で1時
間攪拌した。反応液をアセトン2.5中に滴下し
た。これにシエチルエーテル500mlを滴下し、生
じた沈殿を取した。これをアセトン、ジエチル
エーテルで洗浄後、風乾して目的物6.09gを得
た。
実施例 16
7β−アミノ−3−(4−カルバモイル−1−キヌ
クリジニオ)メチル−3−セフエム−4−カルボ
キシレイト 塩酸塩
7β−フエニルアセトアミド−3−(4−カルバ
モイル−1−キヌクリジニオ)−3−セフエム−
4−カルボキシレイト1gを水19mlに懸濁し、リ
ン酸二カリウム水溶液で液のPHを8.0に調整して
基質液とした。キヤリアーフイクスドペニシリン
Gアミダーゼ〔商品名、大腸菌由来のペニシリン
Gアミダーゼ(キヤリア:ポリアクリルアミド)
ベーリンガーマンハイム山之内株式会社製〕1g
に水を加えて膨潤させた後、前記基質液を加え
た。反応温度を28℃、溶液のPHを8.0(リン酸二カ
リウム水溶液で調整)に保ちながら1時間攪拌し
た。これを過し、液を1N塩酸で酸性にした
後、減圧濃縮した。残渣をセパビーズSP207(商
品名、スチレン−ジビニルベンゼン共重合体系吸
着剤、三菱化成株式会社製)のカラムクロマトグ
ラフイー(展開溶媒:水、30%メタノール)にて
精製して目的物405mgを得た。

Examples include compounds described in publications such as No. 192394. However, no compound having a (4-carbamoyl-1-quinuclidinio)methyl group at the 3-position of its cefem skeleton is known. The present inventors have discovered that a compound having the above-mentioned group at the 3-position has excellent antibacterial activity. The compound of the present invention is a novel synthetic intermediate thereof. Therefore, an object of the present invention is to provide a novel compound useful as an intermediate for antibacterial agents and a method for producing the same.
