Description
(産業上の利用分野)
この発明は、クロストリジウム属に属する新規
な細菌に関するものである。この細菌は、二酸化
炭素と水素とから酢酸を製造する方法に用いるこ
とができる。
(従来技術)
常温の条件下で二酸化炭素と水素とを資化し
て、生育培地中に酢酸を蓄積する微生物として、
クロストリジウム・アセチカム、アセトバクテリ
ウム・ウツデイ、ユーバクテリウム・リモサム、
ブチリバクテリウム・メチロトロフイカム、クロ
ストリジウム・ストレインCV−AA1そして我々
が創製したクロストリジウム・エスピーNo.68−2
微工研菌寄第7367号(FERM−PNo.7367)、クロ
ストリジウム・エスピーNo.307微工研菌寄第7487
号(FERM−PNo.7487)、クロストリジウム・エ
スピーNo.484微工研菌寄第7488号(FERM−PNo.
7488)、アセトバクテリウム・エスピーNo.446微工
研菌寄第7563号(FERM−PNo.7563)などが知
られていた。また高温の条件下では、アセトゲニ
ウム・キヴイ、クロストリジウム・サーモオート
トロフイカム、がある。
(発明の目的)
上記のような菌が知られているものの、二酸化
炭素と水素とからの酢酸の製造を工業的に実施す
るために、解決すべき課題はまだ多く、中でも酢
酸の蓄積濃度および生産速度の高い菌を得ること
は重要である。そのためには、公知の菌種にもと
づいた育種改良とならんで、二酸化炭素と水素と
から酢酸を製造しうる新菌種の創製がきわめて重
要な手段となる。
そして、PH6以下に最適生育領域を持ち、二酸
化炭素と水素から酢酸を製造する能力のある菌は
まだ報告されていない。
本発明はこのような目的のもとに、二酸化炭素
と水素を資化して、培地中に酢酸を蓄積する新規
微生物を得ることを目的とする。
(発明の構成)
本発明は幅1.5μm、長さ6.0〜8.0μmの嫌気性菌
で単独もしくは2〜3連の直桿菌であり胞子を作
る点で、クロストリジウム属に属する菌であると
考えられるが、二酸化炭素と水素で生育し、グラ
ム染色陰性でありかつPH4.0〜7.0の範囲で生育し
鞭毛の無い直桿菌で以下に示す性質のため公知の
菌と相違しており、新菌種であると考えられる。
正式の種名はまだ付されていないので、本発明で
はクロストリジウム・エスピーNo.672と表示する。
次にクロストリジウム・エスピーNo.672創製法
および菌学的性質を示す。
(創製法)
本菌は熊本県の阿蘇の泥より下記の方法により
分離した。すなわち第1表に示す液体培地5mlを
試験管へ分注し減菌後、嫌気グロープボツクス中
で約0.3gの土壌を添加し、ブチルゴム栓で密栓
後、気相を水素(67%)と二酸化炭素(33%)を
含む除菌ガスに置換し、30℃で静置培養し、約3
週間毎に植え継ぎを行つた。2回液体培地で植え
継いだのち、第1表の培地に寒天3%を加えた寒
天培地を用いてロールチユーブ法(メソツズ・イ
ン・マイクロバイオロジー、3巻B、117頁
(1969)アカデミツク・プレス)により単菌分離
し本菌を得た。
(菌学的性質)
本発明の菌株の菌学的性質を示す。この菌学的
性質の検討には、「アンアエロブ・ラボラトリ
ー・マニユアル第4版」(Anaerobe Laboratory
Manual、、The V.I.P.Anaerobe Laboratory

University、Blackburg(1972))およびバージエ
ーズ・マニユアル・オブ・デターミネイテイブ・

第1表の基本培地に下記炭素源(特に記載が無
いときは1%)を含む液体培地5mlを直径18mmの
試験管に加え、無菌培地を作成し本菌を植菌し気
相を窒素(67%)と二酸化炭素(33%)を含む除
菌ガスに置換し、30℃で14日間静置培養した。生
育は600nmの濁度を分光計(スペトロニツク20、
島津製作所製)で測定した。600nmの濁度が炭
素源を含まないコントロールとの差が0.1未満の
ものを「資化しない」、0.1以上0.2未満のものを
「わずかに資化する」0.2以上のものを「資化す
る」とした。
資化するもの:D−グルコース、D−フラクトー
ス、キシロース、D−リボース、ラムノース、

第1表の基本培地に上記の試験で資化すること
の知られた炭素源を1%もしくは0.5%添加し、
気相を窒素(67%)と二酸化炭素(33%)を含む
除菌ガスに置換し、本菌を植菌、30℃で静置培養
した。すべての炭素源において培地中には有機酸
としての酢酸と酪酸が生産された。
(在来の類似種との比較など)
上記の菌学的性質から、No.672は、偏嫌気性の
グラム陰性有胞子桿菌で、その主要醗酵酸代謝産
物が二酸化炭素と水素からは酢酸、その他の資化
する炭素源から酪酸であることを特徴とする菌株
である。この性状からバージーズ・マニユアル・
オブ・デターミネイテイブ・バクテリオロジー第
8版、バージーズ・マニユアル・オブ・システマ
テイツク・バクテリオロジー(ボリユーム1)及
びアンアエロブ・ラボラトリー・マニユアル第4
版にもとずき検索するとクロストリジウム
(Clostridium)に属する菌株であると考えられ
る。そこでアンアエロブ・ラボラトリー・マニユ
アル第4版に従つて同定していくとクロストリジ
ウム・フアラツクス(C.fallax)に行きあたる。
またバージーズ・マニユアル・オブ・デターミネ
テイブ・バクテリオロジー第8版及び、バージー
ズ・マニユアル・オブ・システマテイツク・バク
テリオロジー(ボリユーム1)には諸性状がNo.
672と一致する菌種の記載はなかつた。No.672とク
ロストリジウム・フアラツクスの性状を比較した
ところ共に偏性嫌気性の有胞子桿菌である点で一
致したが、第2表に示す点で両菌の性状は違つて
いた。

However, it grows on carbon dioxide and hydrogen, is negative for Gram staining, grows in the pH range of 4.0 to 7.0, and is a straight bacillus without flagella.It is different from known bacteria due to the following properties, and is a new bacterial species. Since the official species name has not yet been assigned, it is designated as Clostridium sp. No. 672 in the present invention. Next, we will show the method for creating Clostridium sp. No. 672 and its mycological properties. (Creation method) This bacterium was isolated from Aso mud in Kumamoto Prefecture by the following method. That is, 5 ml of the liquid medium shown in Table 1 was dispensed into test tubes, sterilized, approximately 0.3 g of soil was added in an anaerobic globe box, and after sealing with a butyl rubber stopper, the gas phase was mixed with hydrogen (67%) and carbon dioxide. (培養方法)
培養方法は原則的には、一般の微生物の場合と
同様であるが、酸素の混入を防ぐことが必要であ
り、実験室的には、ゴム栓等で密栓した培養器中
で、静置あるいは振盪する方法が用いられる。や
や大きい規模では、通常用いられる醗酵槽がその
まま利用でき、装置内の酸素は、窒素などの不活
性気体あるいは原料気体などで置換することによ
り嫌気的な雰囲気をつくることが可能である。醗
酵槽の形式は特に問わないが、普通に使用される
攪拌混合槽のほか、一段あるいは多段の気泡塔
型、ドラフトチユーブ型の醗酵槽も利用できる。
培養に用いる炭素源は、通常、二酸化炭素ガス
として供給するが、培地中に溶解二酸化炭素のあ
るいは炭酸塩、炭素水素塩として加えることもで
きる。窒素源は塩化アンモニウムのごときアンモ
ニウム塩や硝酸ソーダのような硝酸塩のごとく、
通常の醗酵に用いうる各種の窒素化合物を用いる
ことができる。
その他必要に応じ、リン酸二水素カリ、硫酸マ
グネシウム、硫酸マンガン、塩化ナトリウム、硫
酸鉄、塩化コバルト、塩化カルシウム、硫酸亜
鉛、硫酸銅、明ばん、モリブデン酸ソーダ、硼酸
などの無機化合物、あるいは酵母エキスなどのビ
タミン源を添加することは、通常行なわれる通り
である。
以下具体例により本発明を説明する。
実施例 1
クロストリジウム・エスピーNo.672株を以下の
ように培養した。第1表に示す培地を試験管へ5
ml分注減菌後、同培地で培養を行つた培養液
100μを嫌気グローブボツクス(フアーマ社、
アナエロボツクス)中で添加し、ブチルゴム栓で
密栓したのち気相を水素(67%)と二酸化炭素
(33%)を含む除菌ガスに置換し、30℃で静置培
養した。
培養液の一部を遠心分離機により菌体を分離
し、この上清をリン酸で酸性にして、ガスクロマ
トグラフイーにより生成物の定量を行なつた。
その結果、静置培養10日間で0.82g/の酢酸
を生成していた。
実施例 2
L字型試験管を用い、実施例1と同様に準備し
てクロストリジウム・エスピーNo.672株の振盪培
養を行なつた。測定方法も実施例1と同様に行な
い生成物を分析した結果10日間で1.15g/の酢
